We studied the importance of proper model assumption in the context of Bayesian phylogenetics by examining >5,000 Bayesian analyses and six nested models of nucleotide substitution. Model misspecification can strongly bias bipartition posterior probability estimates. These biases were most pronounced when rate heterogeneity was ignored. The type of bias seen at a particular bipartition appeared to be strongly influenced by the lengths of the branches surrounding that bipartition. In the Felsenstein zone, posterior probability estimates of bipartitions were biased when the assumed model was underparameterized but were unbiased when the assumed model was overparameterized. For the inverse Felsenstein zone, however, both underparameterization and overparameterization led to biased bipartition posterior probabilities, although the bias caused by overparameterization was less pronounced and disappeared with increased sequence length. Model parameter estimates were also affected by model misspecification. Underparameterization caused a bias in some parameter estimates, such as branch lengths and the gamma shape parameter, whereas overparameterization caused a decrease in the precision of some parameter estimates. We caution researchers to assure that the most appropriate model is assumed by employing both a priori model choice methods and a posteriori model adequacy tests.
This study provides the first descriptions of sperm storage at the tissue and cellular levels in a female frog or toad. Oviducal anatomy was studied by light and electron microscopy in Ascaphus truei from north coastal California. Ascaphus truei is one of the few species of anurans in which fertilization is internal. Unlike other anurans with internal fertilization, however, mating in A. truei consists of a unique combination of amplectic and copulatory mechanisms that we term "copulexus." Posterior to a short, aglandular infundibular region, the oviduct possesses: 1) a proximal, convoluted ampullary region where intrinsic tubular glands secrete gelatinous envelopes around eggs; 2) a middle ovisac region where fertilization occurs; and 3) a distal oviducal sinus formed by medial junction of the ovisacs. Sperm storage tubules (SSTs) occur in the anterior portions of the ovisacs and consist of simple tubular glands. SSTs and the rest of the oviducal lining stain positively with the periodic acid-Schiff's procedure for neutral carbohydrates and this reaction is especially intense in reproductively active females. Sperm were found in the SSTs of gravid females as well as some nonvitellogenic females. The sperm are in orderly bundles in the SSTs, and although occasionally sperm nuclei were embedded in the epithelium, no evidence for spermiophagy was found. Oviducal sperm storage in A. truei is homoplastic, with closest structural similarities to squamate reptiles. Oviduct/sperm design constraints appear to limit the options for expression of features associated with oviducal sperm storage.

Ultrastructure of the spermathecae after the breeding season. -Acta Zoologica (Stockholm) 82 : 49-56.Sperm storage in cloacal spermathecae was studied in females of Triturus v. vulgaris from southern England killed at the end of the breeding season in June. This species mates and oviposits eggs in ponds from March to June. Included in the sample were 12 unmated females collected in terrestrial situations in March and mated in the laboratory. Some of these females oviposited viable eggs in the laboratory whereas others did not oviposit after mating. In addition, we examined five females with unknown mating histories that were collected from a breeding pond in June. We found that all of the specimens contained some stored sperm and were similar in spermathecal ultrastructure. The spermathecae exhibited characteristics of secretory epithelium at the end of a cycle, including irregular heterochromatic nuclei surrounded by scant cytoplasm, absence of organelles involved in synthetic activities, few secretory vacuoles, and wide intercellular canaliculi. Spermiophagy by the spermathecal epithelium was extensive. In contrast, spermathecae from females at the beginning of the breeding season as reported in our previous study were actively producing a PAS+ secretion and did not exhibit spermiophagy. Spermiophagy is a means of eliminating sperm prior to the next breeding season.
