A perícia criminal é uma ciência que está no centro da resolução de todos os tipos de crimes, desde aqueles que envolvem vítimas fatais até aqueles relacionados ao meio ambiente. O objetivo geral do artigo foi descrever as principais técnicas moleculares padronizadas para identificação de agentes biológicos em violência sexual. Foi realizada uma revisão sistemática. Os artigos empregados vão desde o caráter forense a fundamentação teórica acerca de definições científicas sobre temas que englobam a área da pesquisa e para isso foram utilizados critérios de inclusão e exclusão segundo a metodologia PRISMA. Foram selecionados 10 artigos para compor a revisão. Identificou-se após apurar vários métodos da biologia molecular o reconhecimento das principais técnicas utilizadas, a PCR e a eletroforese, como sua complementar. Mesmo com pouca quantidade de material genético encontrado em uma cena do crime, como DNA presente em um cigarro ou um copo utilizado pelo suspeito na hora do crime, a técnica de PCR e suas variações possibilita amplificar, multiplicando o DNA até a amostra ser suficiente para análise, fazendo a tipagem do material. Conclui-se a partir dessa revisão que as principais técnicas utilizadas são Técnicas de PCR (cadeia de polimerase), Técnica de eletroforese e de extração de DNA e técnica de sequenciamento de DNA. Verificou-se que análise forense é importante para buscar maior exatidão e precisão quantitativa e qualitativa para resultados que comprovem a origem da identificação da amostra analisada.
