A cultura da batata é de expressiva relevância econômica e social, com grande importância na dieta alimentar de muitos países devido às suas características nutricionais. No entanto, muitos fatores influenciam a qualidade dos tubérculos, entre eles pragas e doenças, como a sarna da batata causada por bactérias do gênero Streptomyces e fitonematoides dos gêneros Pratylenchus e Meloidogyne. Os sintomas causados por esses patógenos podem ser muito semelhantes, podendo resultar em um diagnóstico equivocado do agente causal e, consequentemente, medidas incorretas de manejo. A partir dessa informação, objetivou-se avaliar a ação isolada ou conjunta de S. scabiei, P. brachyurus e M. incognita, verificando se a ocorrência desses patógenos influencia a severidade dos sintomas, degradando tubérculos de batata. Foram realizados três experimentos em casa de vegetação, inoculando-se os fitopatógenos no início do ciclo da cultura, a qual foi conduzida até seu desenvolvimento completo para as avaliações dos tubérculos. A partir dos resultados, foi possível observar a influência do clima no desenvolvimento da batata e consequentemente na interação entre a cultura e os fitopatógenos. Constatou-se semelhança nos danos causados por esses fitopatógenos, o que pode levar a identificação e manejo equivocado destes agentes. Os resultados mostraram que a incidência de lesões causadas por S. scabiei foi em média superior a 12%, enquanto os sintomas causados por P. brachyurus e M. incognita variaram de 3 a 6%, indicando maior patogenicidade da bactéria, uma vez que essa atua desde a formação dos primeiros tubérculos e os nematoides precisam de maior período para penetração e multiplicação para causar danos mais acentuados.
Two new actinobacteria, designated strains IBSBF 2807T and IBSBF 2953T, isolated from scab lesions on potato tubers grown in the southern Brazilian states of Rio Grande do Sul and Santa Catarina, respectively, were characterized and identified through a polyphasic approach. Phylogenetic analyses of 16S rRNA sequences revealed that these two strains belong to the genus Streptomyces . Multilocus sequence analysis using five concatenated genes, atpD, gyrB, recA, rpoB and trpB, allocated strains IBSBF 2807T and IBSBF 2953T in distinct branches of Streptomyces phytopathogenic strains. PCR-RFLP analysis of the atpD gene also confirmed that these strains differ from the type strains of Streptomyces associated with potato scab. The morphological, physiological and biochemical characterization, along with the overall genome-related index properties, indicated that these two strains could be distinguished from their closest phylogenetic relatives and each other. According to the data, IBSBF 2807T and IBSBF 2953T represent two new Streptomyces species related to potato scab. The proposed names for these strains are Streptomyces hilarionis sp. nov. (IBSBF 2807T=CBMAI 2674T=ICMP 24297T=MUM 22.66T) and Streptomyces hayashii sp. nov (IBSBF 2953T=CBMAI 2675T=ICMP 24301T=MUM 22.68T).
