The mechanisms underlying the transport of leptin into the brain are still largely unclear. While the leptin receptor has been implicated in the transport process, recent evidence has suggested an additional role of LRP2 (megalin). To evaluate the function of LRP2 for leptin transport across the blood-brain barrier (BBB), we developed a novel leptin-luciferase fusion protein (pLG), which stimulated leptin signaling and was transported in an in vitro BBB model based on porcine endothelial cells. The LRP inhibitor RAP did not affect leptin transport, arguing against a role of LRP2. In line with this, the selective deletion of LRP2 in brain endothelial cells and epithelial cells of the choroid plexus did not influence bodyweight, body composition, food intake, or energy expenditure of mice. These findings suggest that LRP2 at the BBB is not involved in the transport of leptin into the brain, nor in the development of obesity as has previously been described.

Geburtstage in alter Freundschaft gewidmetIn den Jahren 1952-1956 1 führten wir mit der einzelligen Grünalge Scenedesmus obliquus, gezüchtet an der Kohlenstoffbiologischen Forschungsstation in Essen, umfangreiche Fütterungsversuche an wachsenden Albinoratten durch. Unsere Versuche erstreckten sich auf mehrere Chargen dieses Algenmaterials, das aus mehr als 1000 Einzelernten stammte. Die Scenedesmus-Algen waren in Essen nach dem Abzentrifugieren unter Infrarot-Strahlern bei höchstens 60-70° getrocknet und vermählen worden. In den Versuchen zeigte sich, daß das getrocknete Algenmaterial bzw. sein zu 60-70% verdauliches Protein im allgemeinen höchste biologische Wertigkeit und ausgesprochene Lebernekrose verhütende Eigenschaften besitzen kann.Zwölf verschiedene und zu verschiedenen Zeiten geerntete Chargen von jeweils mehreren Küogramm Trockenalgen zeigten eine erstaunliche Gleichmäßig-keit in der Höhe der diätetischen Eigenschaften. Drei weitere Chargen (13,14, 15) waren weniger wertvoll. Dabei handelt es sich aber um in Abwasser gezüchtete Algen 2 . Die zuletzt untersuchten Proben Nr. 22-26 waren wieder ebenso gut wie die Proben l -12. In der Kohlenstoff biologischen Forschungsstation war inzwischen ein neuer Stamm von Scenedesmus obliquus eingesetzt worden. (Die Proben 17-21 hatten nämlich die Qualität der Proben 1 -12 nicht erreicht.)Zusammen mit dem Ergebnis unserer Vitamin E-Bestimmungen 3 und den von H. Lundin 4 , Stockholm, und Mitarbeitern an unseren verfaßt von K. Lang, ist der Hinweis verabsäumt worden, daß es sich bei den in diätetischer Hinsicht schlechter befundenen Algen in unserer kurzen Originalmitteilung, Naturwissenschaften 43, 498 [1956], um Abwasser-Algen handelte. 8 Gemeinsam mit E.
