Penelitian ini bertujuan mengetahui keragaman fenotipe serta genotipe dan morfometrik antar ayamKampung, ayam Bangkok dan ayam Katai, ayam Birma, Bagon dan Magon. Penelitian ini dilakukan di Sembilanlokasi berbeda di enam Kecamatan Kota Medan menggunakan 368 ekor ayam. Pengambilan data dilakukan dengancara pengamatan pada sifat kualitatif meliputi warna bulu, pola bulu, corak bulu, kerlip bulu, warna shank danbentuk jengger dan morfometrik tubuh. Data yang diperoleh diolah dengan analisis diskriminan sederhana denganmenggunakan program SAS (Statistical Analysis System) dan Dendogram menggunakan program MEGA(Molekuler Evolusioner Genetic Analysis). Hasil penelitian uji kebebasan menunjukkan bahwa adanyaketerhubungan yang sangat nyata (X2> X20,05) antara sifat pola bulu, corak bulu, kerlip bulu, warna shank danbentuk jengger, tetapi tidak adanya keterhubungan (X2< X20,05) antara warna bulu dan jenis ayam yang diamati.Kemurnian fenotifik masing-masing jenis ayam ialah jenis Katai 100%, Kampung 98,46%, Bangkok 84,13%, Bagon75,00%, Magon 65,00% dan Birma 40,00%. Faktor peubah pembeda morfologi tubuh dari keenam jenis ayam ialahpanjang femur, panjang tibia, panjang shank, lingkar shank, panjang sayap, panjang maxilla dan panjang jari ketiga.Jarak genetik ayam Bagon dengan ayam Magon memiliki jarak genetik yang dekat, sebaliknya ayam Kampung danayam Katai memiliki jarak genetik yang jauh.
