RESUMOO objetivo deste trabalho foi avaliar a influência de diferentes condições de preparo de amostra para a determinação de nitrogênio total (NT) em tecido foliar. Durante o estudo três procedimentos foram avaliados: No primeiro foi utilizada uma mistura catalítica a base de selênio/sulfato de potássio em meio ácido. No segundo e terceiro foram utilizadas uma mistura composta por sulfato de cobre/sulfato de potássio em meio ácido sendo que no método três menores quantidades de reagentes foram utilizadas. De acordo com os resultados os valores de NT obtidos pelos três métodos apresentam coeficiente de variação (CV) menor que 15% e desvio padrão menor que 0,7. Ainda de acordo com resultados do teste t não existe diferença estatística entre a média das variáveis. Desta forma, o terceiro método empregando quantidades reduzidas de reagentes e mistura catalítica a base de sulfato de cobre/sulfato de potássio é o mais indicado de acordo com o estudo. Devido a menor quantidade de reagentes utilizados, redução do custo das analises, menor quantidade de resíduos gerados e a substituição do selênio na mistura catalítica que é uma ameaça ao meio ambiente e potencial carcinogênico.Palavras-chave: Análise Química; Química Verde; Tecido Vegetal. The purpose of this work was to evaluate the influence of different conditions of the sample preparation to determination of total nitrogen (TN) in foliar tissue.
