Melanoma en agresif kanser türlerindendir ve tedavilerin yetersiz olmasından kaynaklı ileri aşamlarada kötü prognoz ile karakterizedir. Kurkuminin kolon, pankreas, prostat, karaciğer ve multiple miyelom dahil olmak üzere çeşitli kanser türlerinde lezyonlara karşı etkili olduğu kanıtlanmıştır. Biz de çalışmamızda kurkuminin malign melanoma hücrelerine etkisini araştırmayı amaçladık. Çalışmamızda SK-MEL – 30 insan melanoma hücre hattı kullanıldı. SK-MEL – 30 melanoma hücreleri kültüre edildikten sonra 2.5 µg, 6.75 µg, 12.5 µg, 15 µg ve 25 µg kurkumin ile 24 ve 48 saat süre ile inkübe edildi. Annexin V/PI ve Kaspaz 3/7 analizleri ile hücre canlılıkları ve apoptoz açısından değerlendirildi. Annexin V/PI analizi sonucunda 25 µg kurkuminin 48 saat süre sonunda malin melanoma hücrelerinde canlılığı yaklaşık %50 oranında azalttığı tespit edildi. Kaspaze 3/7 analizi sonucunda kurkuminin doz oranı arttıkça hücre ölümünün gerçekleştiği ve 25 µg kurkumin uygulamasının melanoma hücrelerinde apoptozu indükleyerek hücre ölümüne neden olduğu gösterildi. Sonuç olarak bu çalışma ile kurkuminin malin melanoma hücrelerine karşı antiproliferatif ve apoptozu indükleyici etkisi olduğu belirlendi. Kurkuminin klinik etkilerinin tespit edilebilmesi için daha kapsamlı çalışmaların yapılması gerektiğini önermekteyiz.
Numerous cellular processes are controlled by the ubiquitin‐proteasome‐mediated degradation pathway, involve the 97‐kDa valosin‐containing protein (p97/VCP). Small p97/VCP‐interacting protein (SVIP) was first discovered as one of the novel androgen‐responsive genes as well as one of the many cofactors controlling p97/VCP. The aim of the study was to investigate localization and immunoexpression of p97/VCP and SVIP in rat ovarian tissue. The histomorphological examination of rat ovarian tissue was performed by using haematoxylin–eosin (HE) staining. Using the immunohistochemical technique, cellular location and expression of p97/VCP and SVIP in rat ovarian tissue were examined. The nuclear and cytoplasmic immunoexpression of p97/VCP and SVIP was observed in the different stages of ovarian follicles and corpus luteum in the rat ovaries. The immunolocalization of SVIP and VCP in the rat ovaries suggest that they may be involved in the oogenesis. Further studies should be performed about the function of the VCP and SVIP in the female reproductive tract.
