Adding nanofillers to PLA/PCL blends to change their surface and interface properties can improve their phase morphology. Here the selective localization of CNTs and organoclays as the third component in the blend is studied. It is found that clay is selectively localized in the PLA phase and at the phase interface whereas CNTs are mainly found in the PCL phase and at the phase interface. With a reduced viscosity ratio of the blend matrices, the CNTs change their preferred localization from PCL to PLA. The effects of the different selective localization of clay and CNTs on the morphologies are studied. In addition, the crystallization behavior of ternary systems also shows a strong dependence on the selective localization of nanofillers. magnified image
COVID-19 remains a serious emerging global health problem, and little is known about the role of oropharynx commensal microbes in infection susceptibility and severity. Here, we present the oropharyngeal microbiota characteristics identified by shotgun metagenomic sequencing analyses of oropharynx swab specimens from 31 COVID-19 patients, 29 influenza B patients, and 28 healthy controls. Our results revealed a distinct oropharyngeal microbiota composition in the COVID-19 patients, characterized by enrichment of opportunistic pathogens such as Veillonella and Megasphaera and depletion of Pseudopropionibacterium, Rothia, and Streptococcus. Based on the relative abundance of the oropharyngeal microbiome, we built a microbial classifier to distinguish COVID-19 patients from flu patients and healthy controls with an AUC of 0.889, in which Veillonella was identified as the most prominent biomarker for COVID-19 group. Several members of the genus Veillonella, especially Veillonella parvula which was highly enriched in the oropharynx of our COVID-19 patients, were also overrepresented in the BALF of COVID-19 patients, indicating that the oral cavity acts as a natural reservoir for pathogens to induce co-infections in the lungs of COVID-19 patients. We also found the increased ratios of Klebsiella sp., Acinetobacter sp., and Serratia sp. were correlated with both disease severity and elevated systemic inflammation markers (neutrophil–lymphocyte ratio, NLR), suggesting that these oropharynx microbiota alterations may impact COVID-19 severity by influencing the inflammatory response. Moreover, the oropharyngeal microbiome of COVID-19 patients exhibited a significant enrichment in amino acid metabolism and xenobiotic biodegradation and metabolism. In addition, all 26 drug classes of antimicrobial resistance genes were detected in the COVID-19 group, and were significantly enriched in critical cases. In conclusion, we found that oropharyngeal microbiota alterations and functional differences were associated with COVID-19 severity.
Summary This paper describes successful practices applied during polymer flooding at Daqing that will be of considerable value to future chemical floods, both in China and elsewhere. On the basis of laboratory findings, new concepts have been developed that expand conventional ideas concerning favorable conditions for mobility improvement by polymer flooding. Particular advances integrate reservoir-engineering approaches and technology that is basic for successful application of polymer flooding. These include the following:Proper consideration must be given to the permeability contrast among the oil zones and to interwell continuity, involving the optimum combination of oil strata during flooding and well-pattern design, respectively;Higher polymer molecular weights, a broader range of polymer molecular weights, and higher polymer concentrations are desirable in the injected slugs;The entire polymer-flooding process should be characterized in five stages--with its dynamic behavior distinguished by water-cut changes; -Additional techniques should be considered, such as dynamic monitoring using well logging, well testing, and tracers; effective techniques are also needed for surface mixing, injection facilities, oil production, and produced-water treatment; andContinuous innovation must be a priority during polymer flooding. Introduction China's Daqing oil field entered its ultrahigh-water-cut period after 30 years of exploitation. Just before large-scale polymer-flooding application, the average water-cut was more than 90%. The Daqing oil-field is a large river-delta/lacustrine facies, multilayered with complex geologic conditions and heterogeneous sandstone in an inland basin. After 30 years of waterflooding, many channels and high-permeability streaks were identified in this oil field (Wang and Qian 2002). Laboratory research began in the 1960s, investigating the potential of enhanced-oil-recovery (EOR) processes in the Daqing oil field. After a single-injector polymer flood with a small well spacing of 75 m in 1972, polymer flooding was set on pilot test. During the late 1980s, a pilot project in central Daqing was expanded to a multiwell pattern with larger well spacing. Favorable results from these tests--along with extensive research and engineering from the mid-1980s through the 1990s--confirmed that polymer flooding was the preferred method to improve areal- and vertical-sweep efficiency at Daqing and to provide mobility control (Wang et al. 2002, Wang and Liu 2004). Consequently, the world's largest polymer flood was implemented at Daqing, beginning in 1996. By 2007, 22.3% of total production from the Daqing oil field was attributed to polymer flooding. Polymer flooding boosted the ultimate recovery for the field to more than 50% of original oil in place (OOIP)--10 to 12% OOIP more than from waterflooding. At the end of 2007, oil production from polymer flooding at the Daqing oil field was more than 10 million tons (73 million bbl) per year (sustained for 6 years). The focus of this paper is on polymer flooding, in which sweep efficiency is improved by reducing the water/oil mobility ratio in the reservoir. This paper is not concerned with the use of chemical gel treatments, which attempt to block water flow through fractures and high-permeability strata. Applications of chemical gel treatments in China have been covered elsewhere (Liu et al. 2006).
Glycosides are known to be precursors of the alcoholic aroma compounds of black tea. They are hydrolyzed by endogenous glycosidases during the manufacturing process. Changes in the amounts of these glycosides during the manufacturing process were investigated by using a capillary gas chromatographic--mass spectrometric analysis after trifluoroacetyl derivatization of the tea glycosidic fractions. Primeverosides were 3-fold more abundant than glucosides in fresh leaves, but they decreased greatly during the manufacturing process, especially during the stage of rolling. After the final stage of fermentation, primeverosides had almost disappeared, whereas glucosides were substantially unchanged. These results show that hydrolysis of the glycosides mainly occurred during the stage of rolling and confirm that primeverosides are the main black tea aroma precursors. This was also supported by the changes in the glycosidase activities in tea leaves. The glycosidase activities remained at a high level during withering but decreased drastically after rolling.
The ezrin-radixin-moesin proteins provide a regulated linkage between membrane proteins and the cortical cytoskeleton and also participate in signal transduction pathways. Ezrin is localized to the apical membrane of parietal cells and couples the protein kinase A activation cascade to the regulated HCl secretion. Our recent proteomic study revealed a protein complex of ezrin-ACAP4-ARF6 essential for volatile membrane remodeling (Fang, Z., Miao, Y., Ding, X., Deng, H., Liu, S., Wang, F., Zhou, R., Watson, C., Fu, C., Hu, Q., Lillard, J. W., Jr., Powell, M., Chen, Y., Forte, J. G., and Yao, X. (2006) Mol. Cell Proteomics 5, 1437-1449). However, knowledge of whether ACAP4 physically interacts with ezrin and how their interaction is integrated into membrane-cytoskeletal remodeling has remained elusive. Here we provide the first evidence that ezrin interacts with ACAP4 in a protein kinase A-mediated phosphorylation-dependent manner through the N-terminal 400 amino acids of ACAP4. ACAP4 locates in the cytoplasmic membrane in resting parietal cells but translocates to the apical plasma membrane upon histamine stimulation. ACAP4 was precipitated with ezrin from secreting but not resting parietal cell lysates, suggesting a phospho-regulated interaction. Indeed, this interaction is abolished by phosphatase treatment and validated by an in vitro reconstitution assay using phospho-mimicking ezrin S66D . Importantly, ezrin specifies the apical distribution of ACAP4 in secreting parietal cells because either suppression of ezrin or overexpression of non-phosphorylatable ezrin prevents the apical localization of ACAP4. In addition, overexpressing GTPase-activating protein-deficient ACAP4 results in an inhibition of apical membrane-cytoskeletal remodeling and gastric acid secretion. Taken together, these results define a novel molecular mechanism linking ACAP4-ezrin interaction to polarized epithelial secretion.
Summary This paper investigates the potential of various approaches for improving sweep in parts of the Daqing oil field that have been enhanced-oil-recovery (EOR) targets. Our studies indicated that the polymer flood should have provided excellent sweep throughout the vast majority of the patterns under consideration. However, because alkaline/surfactant/polymer (ASP) flooding is being considered to increase recovery efficiency from the Daqing oil field, mobility control and sweep improvement will be especially important and challenging during implementation of any future ASP process. Fractures were present in a number of Daqing wells (both injectors and producers). Because the fractures were narrow and far from the wellbore, severe channeling did not occur. On the contrary, fractures near the wellbore aided reservoir sweep. Nearwellbore fractures increased the injectivity index substantially during the injection of polymer solutions and increased oil-productivity index in the production wells. These observations may be valuable during implementation of future floods where very-low-mobility chemical (i.e., ASP) banks must be injected to maintain mobility control. Several modes of polymer degradation were considered, with mechanical (shear) degradation being of greatest concern. Appropriate use of near-wellbore fractures may mitigate mechanical degradation effectively, as well as improving injectivity. Several new polymers show potential for cost-effective improvements at Daqing. Increased polymer concentration was also considered. A number of other approaches are (or have been) under investigation, including colloidal dispersion gels, foams, ASP foams, steam, microbes, and polymer solutions prepared with reduced salinity.
