Study of mutant phenotypes is a fundamental method for understanding gene function. The construction of a near-complete collection of yeast knockouts (YKO) and the unique molecular barcodes (or TAGs) that identify each strain has enabled quantitative functional profiling of Saccharomyces cerevisiae. By using these TAGs and the SGA reporter, MFA1pr-HIS3, which facilitates conversion of heterozygous diploid YKO strains into haploid mutants, we have developed a set of highly efficient microarray-based techniques, collectively referred as dSLAM (diploid-based synthetic lethality analysis on microarrays), to probe genome-wide gene-chemical and gene-gene interactions. Direct comparison revealed that these techniques are more robust than existing methods in functional profiling of the yeast genome. Widespread application of these tools will elucidate a comprehensive yeast genetic network.

Reactions of the unsymmetrical phenol ligand 2-(bis(2-pyridylmethyl)aminomethyl)-6-((2-pyridylmethyl)(benzyl)aminomethyl)-4-methylphenol with Mn(OAc)(2).4H(2)O or Mn(H(2)O)(6)(ClO(4))(2) in the presence of NaOBz affords the dimanganese(II) complexes 1(CH(3)OH), [Mn(2)(L)(OAc)(2)(CH(3)OH)](ClO(4)), and 2(H(2)O), [Mn(2)(L)(OBz)(2)(H(2)O)](ClO(4)), respectively. On the other hand, reaction of the ligand with hydrated manganese(III) acetate furnishes the mixed-valent derivative 3(H(2)O), [Mn(2)(L)(OAc)(2)(H(2)O)](ClO(4))( 2). The three complexes have been characterized by X-ray crystallography. 1(CH(3)OH) crystallizes in the monoclinic system, space group P2(1)/c, with a = 10.9215(6) A, b = 20.2318(12) A, c = 19.1354(12) A, alpha = 90 degrees, beta = 97.5310(10) degrees, gamma = 90 degrees, V = 4191.7 A(3), and Z = 4. 2(H(2)O) crystallizes in the monoclinic system, space group P2(1)/n, with a = 10.9215(6) A, b = 20.2318(12) A, c = 19.1354(12) A, alpha = 90 degrees, beta = 97.5310(10) degrees, gamma = 90 degrees, V = 4191.7 A(3), and Z = 4. 3(H(2)O) crystallizes in the monoclinic system, space group P2(1)/c, with a = 11.144(6) A, b = 18.737(10) A, c = 23.949(13) A, alpha = 90 degrees, beta = 95.910(10) degrees, gamma = 90 degrees, V = 4974(5) A(3), and Z = 4. Magnetic measurements revealed that the three compounds exhibit very similar magnetic exchange interactions -J = 4.3(3) cm(-)(1). They were used to establish tentative magneto-structural correlations which show that for the dimanganese(II) complexes -J decreases when the Mn-O(phenoxo) distance increases as expected from orbital overlap considerations. For the dimanganese(II,III) complexes, crystallographic results show that the Mn(II)-O(phenoxo) and Mn(III)-O(phenoxo) bond lengths are inversely correlated. An interesting magneto-structural correlation is found between -J and the difference between these bond lengths, delta(Mn)(-)(O) = d(Mn)()II(-)(O) - d(Mn)()III(-)(O): the smaller this difference, the larger -J. Electrochemical studies show that the mixed-valence state is favored in 1-3 by ca. 100 mV with respect to analogous complexes of symmetrical ligands, owing to the asymmetry of the electron density as found in the analogous diiron complexes.

In recent years, metal–organic frameworks (MOFs) have been extensively investigated for diverse heterogeneous catalysis due to their diversity of structures and outstanding physical and chemical properties. Currently, most related work focuses on employing MOFs as porous substrate materials to fabricate confined nanoparticle or heteroatom-doped electrocatalysts which have to be annealed at high temperature before application. However, the annealing process would destroy the structure completely and lose the intrinsic active sites in MOFs framework. Herein, a simple solvothermal process is used to synthesize a series of Fe/Ni bimetallic MOFs. The as-prepared MOFs are applied directly as highly efficient oxygen evolution reaction (OER) electrocatalysts with no post-annealing treatment. The bimetallic FeNi-MOFs show higher OER activity than single metal MOFs and commercial precious RuO2 catalysts. With the optimized FeNi-MOF as the catalyst, the OER current densities of 50 and 100 mA/cm2 can be achieved at the overpotentials of only 270 and 287 mV, respectively. Meanwhile, a small Tafel slope of 49 mV/dec was obtained. Moreover, this catalyst shows high electrochemical stability in strong basic solution. This work demonstrates that through structural optimization, bimetallic and multimetallic MOFs may have promising potentials as advanced catalysts for electrochemical energy conversion.

Energy-efficient, low-cost, and highly durable catalysts for the electrochemical hydrogen evolution reaction (HER) and urea oxidation reaction (UOR) are extremely important for related sustainable energy systems. In the present work, hierarchical coassembled cobalt molybdenum sulfide nanosheets deposited on carbon cloth (CC) were synthesized as catalysts for hydrogen evolution and urea oxidation. By adjusting the doping amount of Mo, 2D nanosheets with different morphologies and compositions (Co x Mo y S-CC) can be obtained. The as-prepared nanosheet materials with abundant active sites exhibit superior properties on the electrochemical HER and UOR in alkaline medium. Significantly, the Mo-doping concentration and composition of the formed nanosheets have large effects on the electrocatalytic activity. The fabricated nanosheets with optimal Mo doping (Co3Mo1S-CC) illustrate the best catalytic properties for the HER in N2-saturated 1.0 M KOH. A small overpotential (85 mV) is needed to meet the current density of 10 mA/cm2. This study indicates that the doping of an appropriate amount of molybdenum into CoS2 nanosheets can efficiently improve the catalytic performance. Also, the nanosheet catalyst exhibits an extremely high electrocatalytic activity for the UOR, and the electrochemical results indicate that a relatively low cell voltage of 1.50 V is needed to obtain the current density of 10 mA/cm2. The present work demonstrates the potential application of CoMoS nanosheets in the energy electrocatalysis area and the insights into performance-boosting through heteroatom doping and optimization of the composition and structure.

Background: Macrophage polarization plays a pivotal role in the process of inflammation which is common in chronic kidney disease (CKD). Macrophages polarization under the condition of CKD remains poorly understood. Here we tested the hypothesis that CKD promotes macrophage M1 polarization. Methods: A rat model of CKD was established by reduced renal mass (RRM). Polarization of macrophages was induced in ex vivo macrophages from RRM rats and cultured ones under the condition of uremic serum. The markers were evaluated by RT-PCR, western blot, and flow cytometer. Results: Our data showed that macrophages from RRM rats displayed enhanced M1 and impaired M2 polarization as revealed by increased M1 markers (tumor necrosis factor α, IL-6, IL-12p40, nitric oxide) and decreased M2 markers (IL-10, CD206, arginase activity) in response to LPS and IL-4 induction, respectively. Treatment with uremic sera in peritoneal and bone marrow derived macrophages from normal rats led to similar results. Moreover, macrophages from RRM rats and cultured under the condition of uremic sera had reduced mitochondrial biogenesis. The disturbed macrophage polarization and mitochondrial biogenesis were accompanied by reduced activity of adenosine monophosphate-activated protein (AMP)-activated kinase (AMPK). Enhancing activation of AMPK restored mitochondrial biogenesis and M2 macrophage polarization. Conclusion: These observations suggest that CKD disturbs macrophage polarization and mitochondrial biogenesis through inhibition of AMPK. This might provide a novel therapeutic strategy for intervention of chronic inflammation in CKD.
