The results of experiments in dogs with and without two glucose loads and in the isolated pancreas to compare the pharmacodynamics of two low-dosage arylsulphonylureas, glipizide and glibenclamide, are described and discussed.-The results agree with those obtained by other authors confirming that glibenclamide shows delayed but prolonged activity on both plasma insulin and glucose levels. Moreover glibenclamide counteracts the hyperglycaemia induced by the second glucose load less effieently. Glipizide acts faster on insulin and glucose levels, which return quickly to normal. When a second glucose load was given it was still more active in reducing plasma glucose levels. The dynamics of insulin secretion following glipizide more closely resembles tolbutamide than glibenelamide.

C 49.58 H 4.20 C1 29.30 2.6-Bis-lbis-(~-chlor-afhyl)-aminomefhylj-benzo[l.2.4.5jbisimidazol-dihydrochlorid (I) : 3 g trockenes ZV (freie Base) wurden mit 5 ccm Chloroform und I5 ccm Thionylchlorid 1 Stde. a d dem Wasserbad unter RiickfluD erhitzt. Nach Abtreiben des Thionylchlorids i. Vak. wurde der Riickstand in wenig heiBem Wasser gelBst, mit Kohle behandelt und nach Abfiltrieren derselben in Eis gestellt. Die Substanz schied sich in schwach gelblichen Kristallen aus (0.5 9). Aus der i. Vak. eingeengten Mutterlauge konnte noch einmal die gleiche Menge erhalten werden. Ausb. 1 g (24 % d. Th.). (Eingegangen am 25. Juni 1958)p-Toluolsulfonyl-isocyanat reagiert mit N-monosubstituierten Acetamiden in ahnlicher Weise wie rnit Formamiden. Sind die Amide aliphatisch substituiert, so erfolgt die Amidin-Bildung um so leichter, je verzweigter die Kette ist. Bei der Substitution durch die tert.Butylgruppe bildet sich auch bei tiefer Temperatur sofort das entsprechende Amidin. Die Darstellung ringartig substituierter Amidine wird beschrieben. Die Konstitution und die IR-Spektren der Amidine werden diskutiert.In den vorhergehenden Mitteilungenl) wurde gezeigt, daB sich aus p-Toluolsulfonyl-isocyanat und monosubstituierten Saureamiden teilweise schon bei Zimmertemperatur, im allgemeinen aber im SchmelzfluB oder beim Erhitzen in Liisungsmitteln, substituierte Amidine bilden. So entstehen mit N-Phenyl-formamid und mit N-Cyclohexyl-formamid N-p-Toluolsulfonyl-N'-phenyl-formamidin bzw. N-p-Toluolsulfonyl-N'-cyclohexyl-formamidip ; nur mit N-n-Butyl-formamid haben wir in der Schmelze kein Amidin isolieren konnen. Diese Ausnahme erklaren wir damit, daB ahnlich wie bei den von R. HUISGEN und H. RHIMLINGER~) studierten Nitroso-acyl-1) I.
Die Konstitution der bei der Reaktion von p‐Toluolsulfonyl‐isocyanat mit N‐Cyclohexyl‐formamid isolierten Verbindung vom Schmp. 124–126°, die bei der Verseifung N‐p‐Toluolsulfonyl‐N′‐cyclohexyl‐harnstoff und in der Hitze N‐p‐Toluolsulfonyl‐N′‐cyclohexyl‐formamidin liefert, wird diskutiert. Weiterhin werden einige charakteristische Eigenschaften der IR‐Spektren dieser und ähnlicher Verbindungen mitgeteilt.
N-p-Toluolsulfonyl-N.N'-dimethyl-acetamidin (I) : Der Suspension von 2.4 g N.N'-Dimethylacetamidin-hydrochlorid in 20 a m Aceton setzte man 5 ccm 50-proz. Natronlauge, dann unter Ktihlung 5.6 g ~Toluolsulfonyl-chlorid, gelbst in 30 a m Aceton, zu. Nach 2 Stdn. wurde in Eiswasser gegossen und der Niederschlag aus Toluol umkristallisiert. Ausb. 82 % d. Th., Schmp. 71 -72". CllH16N20zS (240.3) Ber. C 55.34 H 6.67 S 13.34 Gef. C 55.0 H 6.54 S 13.11 N. N'-Bis-p-toluolsulfonyl-acetamidin ( X I I I ) : Der Suspension von 4.5 g Acetamidin-hydrochlorid in 50 ccm Aceton frigte man 30 ccm 50-proz. Natronlauge und anschlieBend 19 g p-Toluolsulfonyl-chlorid, gel6st in 80 ccm Aceton, zu. Nach 2 Stdn. wurde die klare Lbsung in Eiswasser gegossen, angesauert und der ausgefallene Niederschlag aus Benzol kristallisiert. Ausb. 85 % d. Th.; Schmp. 130-131".
