La maladie de Gaucher, l'une des sphingolipidoses les plus courantes, a été la première maladie lysosomale à être traitée par une enzymothérapie substitutive. Ce traitement instauré précocement peut prévenir les complications parfois irréversibles de cette pathologie. Bien que les symptômes de la maladie apparaissent pendant l'enfance chez la majorité des patients, le diagnostic est souvent différé pendant de nombreuses années, même jusqu’à l’âge adulte. Le diagnostic est difficile en raison d'une grande variabilité des manifestations cliniques et de la sévérité des symptômes. En outre, les dosages enzymatiques pour confirmer le diagnostic ne se font que dans des laboratoires spécialisés. Une meilleure connaissance des manifestations cliniques et biologiques de cette maladie permettra au praticien de savoir l’évoquer devant un enfant symptomatique.
