The toxin 6-hydroxydopamine (6-OHDA) is a highly oxidizable dopamine (DA) analog that is widely used for reproducing several cell processes identified in Parkinson’s disease (PD). Due to the close similarity of its neurotoxic mechanism to those of DA, it is suitable as a model for testing the effects of potentially neuroprotective drugs. This study aimed to evaluate the effect of alpha-lipoic acid (LA) on brain oxidative stress (OS) in unilateral intrastriatal (6-OHDA) injected rats. Forty male Wistar rats, four months old (220–260 g), were evaluated. Half of them received LA (35 mg/kg i.p.) from the start to the end of the experiment. On day 2 of the trial, ten LA-supplemented rats and ten non-LA-supplemented rats were subjected to the apomorphine test. Brain homogenates were evaluated for thiobarbituric acid-reactive substances (TBARS) and glutathione peroxidase (GPx) activity. The same evaluation procedures were repeated on day 14 with the remaining animals. An increased TBARS level and decreased GPx activity, suggestive for OS, were recorded in homogenates on day 14 vs. day 2 of the experiment in the 6-OHDA treated rats. The simultaneous application of LA mitigated these changes. Our study demonstrates that the low dose of LA could be of value for decreasing the OS of the neurotoxic 6-OHDA, supporting the need for further studies of the benefit of LA treatment in PD.
Rationale Narcissus cv. Hawera has been found to biosynthesize some Sceletium‐type alkaloids with antidepressant and anxiolytic activities. This ornamental plant has been poorly studied as a source of bioactive alkaloids including some contraversive reports on in vitro and intact plants. In this study, a detailed GC‐MS characterization of its alkaloid fractions is presented. Methods GC‐MS was used for the identification of compounds in the alkaloid fractions. Both underivatized and silylated samples were analyzed simultaneously. Elevated plus maze and tail suspension tests were used to assay the anxiolytic and antidepressant activities. Ellman’s and MTT‐dye reduction assays were used to evaluate the acetylcholinesterase (AChE) inhibitory and cytotoxicity activities, respectively. Results Of the 29 alkaloids, 13 of Sceletium‐type were detected. Two new alkaloids were identified as 2‐oxo‐mesembrine and 2‐oxo‐epi‐mesembrenol. Lycorine was found as a major compound (43.5%) in the crude silylated methanol extract. After the elimination of lycorine by pre‐crystallization, the major alkaloids were 40.8% 6‐epi‐mesembranol, 16.2% 6‐epi‐mesembrenol, and 13.8% sanguinine. This fraction showed anxiolytic and antidepressant‐like activities as well as potent AChE inhibitory and antineoplastic activities. Conclusions Silylation of the alkaloid fractions from Narcissus cv. Hawera provides better separation, structural information, and improved sensitivity for compounds with two and more hydroxyl groups. The lycorine‐free alkaloid fraction shows a great potential for further pharmacological studies.
Cold atmospheric plasma (CAP) applications in various fields, such as biology, medicine and agriculture, have significantly grown during recent years. Many new types of plasma sources operating at atmospheric pressure in open air were developed. In order to use such plasmas for the treatment of biological systems, plasma properties should fulfil strong requirements. One of the most important is the prevention from heating damage. That is why in many cases, the post-discharge region is used for treatment, but the short living particles in the active discharge zone and reactions with them are missed in that case. We use the active region of surface-wave-sustained argon plasma for biological systems treatment. The previous investigations showed good bactericidal, virucidal, seeds germination and decontamination effects at a short treatment time, but the discharge conditions for bio-medical applications need specific adjustment. A detailed theoretical and experimental investigation of the plasma characteristics and their possible optimization in order to meet the requirements for bio-medical applications are presented in this paper. The length of the plasma torch, the temperature at the treatment sample position and the microwave radiation there are estimated and optimized by the appropriate choice of discharge tube size, argon flow rate and microwave power.
The rising burden of depression, which will soon be the second most common cause of disability in the world, is requesting new ways to treat and prevent it. Due to high number of significant adverse drug reactions of the conventional treatment, the modern pharmaceutical industry is more often turning their focus to novel plant-based solutions. We performed literature research based on standard literature search engines – PubMed, Google Scholar, Science Direct. A standard set of keywords related to our topic e.g. “Depression”, “Mesembrine type alkaloids”, “Narcissus” was used. The review describes the classical monoamine theory of depression and connects it with the newly found biochemical, genetic and morphological alterations associated with the major depressive disorder. The purpose of this review is to highlight the most important aspects of the pathophysiology of depression and to explore the possibilities to use mesembrine-like alkaloids isolated from Narcissus cv. Hawera in its treatment. We describe their effect on brain biochemistry and possible future investigations.
The objective of this study was to measure bone mineral density (BMD) in middle-aged men with and without the metabolic syndrome according to the International diabetes federation (IDF) definition from 2005. We studied 80 men (mean age: 51.9 +/- 9.0 y, mean body mass index (BMI): 32.0 +/- 1.7 kg/m2) with and 92 men without the metabolic syndrome (mean age: 52.6 +/- 15.1 y, mean BMI: 24.9 +/- 2.8 kg/m2). Height (cm), weight (kg), waist circumference (cm) and blood pressure were measured. Fasting plasma glucose (FPG) and blood lipids were determined. BMD at the lumbar spine and total hip was measured by dual X-ray absorptiometry on a Hologic QDR 4500 bone densitometer. In men around 59.3% had a waist circumference > 94 cm (abdominal obesity). Among them 58.7% showed abnormal BP values. Around 30.7% had FPG > or = 5.6 mmol/L and 22.7% had low high density lipoprotein (HDL)-cholesterol and 36.6% had hypertriglyceridemia. In men with the metabolic syndrome, mean lumbar spine BMD was 0.986 +/- 0.210 g/cm2 and total hip BMD - 1.012 +/- 0.209 g/cm2. The corresponding values in men without this syndrome were 0.934 +/- 0.179 g/cm2 and 0.894 +/- 0.189 g/cm2, respectively. The inter-group BMD difference reached statistical significance only at the hip (p = 0.039). Respectively, the prevalence of osteoporosis at the central sites was significantly higher in men without the metabolic syndrome (MS) (13.2 versus 20.8%, p = 0.03). Our data confirmed the trend for higher BMD in the studied men with the metabolic syndrome.
