The prevalence of Toxoplasma gondii in free-ranging chickens (Gallus domesticus) is a good indicator of the prevalence of the parasite's oocysts in soil because chicken feed from the ground. The prevalence of T. gondii in free-range chickens from Ghana, Indonesia, Italy, Poland, and Vietnam was determined using the modified agglutination test (MAT). Antibodies to T. gondii were found in 41 (64%) of 64 chickens from Ghana, 24 (24.4%) of 98 chickens from Indonesia, 10 (12.5%) of 80 chickens from Italy, 6 (30%) of 20 chickens from Poland, and 81 (24.2%) of 330 chickens from Vietnam. Hearts and brains of chickens were bioassayed for T. gondii. Viable T. gondii was isolated from 2 chickens from Ghana, 1 chicken from Indonesia, 3 chickens from Italy, 2 chickens from Poland, and 1 chicken from Vietnam. Toxoplasma gondii isolates from 9 chickens were genotyped using 10 PCR-RFLP markers including SAG1, SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, and Apico. A total of 7 genotypes was identified; the 3 isolates from chickens from Italy were clonal type II, and the others were nonclonal. This is the first report of genetic characterization of T. gondii isolates from animals from these countries.
Background and Aim: Toxoplasma gondii is a unicellular coccidian parasite distributed globally and is an important zoonotic pathogen. Approximately 30% of the human population worldwide is chronically infected with T. gondii. The pathogenicity of this species depends on the type originating from the clonal population. Techniques for more accurately determining the type of T. gondii have recently been developed using genetic markers. Specifically, T. gondii has been typed using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). This study aimed to identify sets of PCR-RFLP markers that have high power to discriminate genotyping of T. gondii and are easy to use and are easy to use. The objective of this study was to characterize virulent strain isolates of T. gondii by PCR-RFLP using 10 markers with DdeI. Materials and Methods: T. gondii tachyzoites (RH virulent strain) were derived from culture cells at the Indonesian Research Center for Veterinary Sciences. Genotyping was performed on T. gondii DNA extracted from cell cultured tachyzoites using 10 genetic markers of PCR-RFLP, namely, B1#1, B1#2, B1#3, SAG1#1, SAG1#2, P30, BAG1, ROP1, GRA1, and GRA7, with digestion using the restriction enzyme DdeI. Results: The 10 genes were amplified by PCR. Among them, three genetic markers, B1#3, ROP1, and GRA1, were genotyped by the PCR-RFLP using restriction enzyme DdeI. Overall, the findings showed that the specific RFLP profile of digestion of gene regions by DdeI could be used as a specific marker for the virulent biotype causative of toxoplasmosis. In addition, virulent strains of T. gondii can be easily detected by these markers. Conclusion: Three pairs of primers (B1#3, ROP1, and GRA1) with DdeI have proven useful for the diagnosis of acute toxoplasmosis (virulent strain biotype I). This proposed method is relatively simple, rapid, cheap, and can be performed in most laboratories, providing a practical approach for the routine analysis of T. gondii strains.
Bovine reproductive diseases are endemic in Indonesia, but comprehensive information about their infectious causes is not available. Therefore, our aim for this study was to detect several infectious agents that cause reproductive diseases in Indonesian beef and dairy cows. A total of 152 cow serum samples collected by Faculty of Veterinary Medicine of Brawijaya University and Veterinary Disease Investigation Centre as a part of the mandatory and regularly surveillance system from three provinces during 2019–2020 were used. The samples were then sent to Indonesian Research Centre for Veterinary Science (IRCVS) for further detection of seven reproductive diseases by enzyme-linked immunosorbent assay (ELISA). Seven reproductive diseases to be tested in parallel are neosporosis, chlamydiosis, brucellosis, Q fever, bovine viral diarrhea (BVD), infectious bovine rhinotracheitis (IBR), and BHV-4 infection. The dominant reproductive diseases in Indonesian cows were BVD (45.69%), chlamydiosis (31.58%), IBR (20.53%), neosporosis (11.84%), and BHV-4 infection (10.53%). The seroprevalence of IBR, BHV-4 infection, neosporosis, and brucellosis varied significantly P < 0.05 between dairy and beef cattle. The most dominant reproductive diseases in aborted cows were chlamydiosis (45%), BVD (41%), and neosporosis (10%). The conclusion drawn from this study is that the dominant reproductive diseases in Indonesian cows are BVD, chlamydiosis, IBR, neosporosis, and BHV-4 infection. Chlamydiosis, BVD, and neosporosis are common among aborted cow. Chlamydiosis, neosporosis, and BHV-4 infection should be included in the national priority list in Indonesia. Control and preventive measures should be focused on high-risk areas and animals like stray cat and dog.
Surra merupakan salah satu penyakit menular pada berbagai jenis hewan yang disebabkan oleh protozoa darah. Surra disebabkan oleh Trypanosoma evansi (T. evansi) dan seringkali berakibat fatal berupa kematian yang tinggi terutama pada kuda, kerbau dan unta. Salah satu pengendalian penyakit Surra dapat dilakukan dengan pengobatan menggunakan trypanosidal yang efektif. Efektifitas trypanosidal untuk Surra harus didasarkan pada hasil uji sensitivitas. Oleh sebab itu perlu diteliti efektivitas berbagai trypanosidal terhadap beberapa isolat T. evansi yang berasal dari beberapa daerah kasus maupun wabah Surra di Indonesia agar diketahui kesesuaian dan efikasinya. Metode pengujian dilakukan dengan desain pre test-post test. Mencit DDY diinfeksi dengan beberapa isolat T. evansi dari beberapa daerah kasus dan diamati parasitemianya. Setelah parasitemia mencapai puncaknya, maka mencit diobati dengan masing-masing trypanosidal dengan dosis bertingkat. Selanjutnya mencit diamati perubahan parasitemianya selama kurang lebih satu bulan. Hasil penelitian menunjukkan bahwa setiap isolat memiliki kepekaan yang berbeda terhadap trypanosidal sehingga penggunaan trypanosidal cenderung spesifik lokasi. Secara umum, suramin dan melarsomine dihydrochloride merupakan dua trypanosidal yang paling efektif untuk beberapa isolat di Indonesia. Adapun isometamidium chloride tidak direkomendasikan digunakan untuk pengobatan kasus surra di Indonesia.
