To be used in regenerative medicine, cells should be checked for various conditions, including cell aging. This study aimed to learn senescent profile and its relation to cell viability, proliferation and cell size in various passages, which were done in α-MEM-10% PRP medium, until senescenceassociated β-galactosidase (SA-β-Gal) positive cells were found. Stem cells were isolated from umbilical cord tissue by multiple harvest explant method, cultured in α-MEM-10% PRP until P-17 and stained using SA-β-Gal staining. Viability, Population Doubling Time (PDT), percentage SA-β-Gal (+) and cell size at 30% confluent and at senescent staining were analyzed. Passages with SA-β-Gal (+) and (-) were compared in term of viability, PDT and cell size at 30% confluent. Further, cell size at senescent staining between SA-β-Gal (+) and (-) groups were compared. Viability and PDT showed no significant difference between SA-β-Gal (+) and (-) groups, while cell size at 30% confluence showed significant increase in SA-β-Gal (+) compared to (-) groups. Further, cell size in senescent staining showed significantly smaller cell size in SA-β-Gal (-) compared to SA-β-Gal (+) cells. Moreover, this study showed that even in SA-β-Gal (+) group, viability was greater than 91%, PDT was less than 2.1 days and cell size was less than 2602 µm 2 . In conclusion, umbilical cord derived MSCs that were cultured in α-MEM-10% PRP began to undergo aging at P-10. Morphological criteria of UC-MSC aging were cell size greater than 2602 µm 2 with a change in morphology toward a rounded shape.
In most cryopreservation medium, Fetal Bovine Serum (FBS) is used as supplement, while it is well known that FBS contains xenoproteins that can be incorporated into the cells and may be harmfull, as they can elicit immune response. Therefore, finding other xenofree materials as FBS alternative in cryopreservation medium is very important. Platelet Rich Plasma (PRP) is albumin rich and is a candidate for FBS alternative as cryopreservation supplement. Albumin is a natural extracellular cryoprotective agent that stabilizes impaired cell membrane during cryopreservation. This was an in vitro analytical study to to compare the effect of PRP and FBS as supplement in cryopreservation medium on human umbilical cord stem cells. In this study the stem cells were isolated from an umbilical cord tissue by explant method and propagated untill we got enough cells for cryopreservation. Cryopreservations were done using eight types of protocol, which differed in type and concentration of supplement and cell concentration. The effect of the eight protocols were compared in terms of post cryopreservation cell viability, morphology, cell size and proliferation. There were no difference between FBS and PRP supplemented cryopreservation media in term of cell viability and morphology. PRP supplemented medium showed better post cryopreservation performance in cell size and proliferation. PRP can be used as an alternative to FBS in cryopreservation medium for human umbilical cord tissue derived stem cells.
LATAR BELAKANG Hipertensi didefinisikan tekanan darah sistolik ≥ 140 mmHg atau tekanan darah diastolik ≥ 90 mmHg. RISKESDAS 2018 menunjukkan prevalensi hipertensi di Indonesia adalah 34,11%. Hipertensi merupakan risiko kesehatan umum pada pekerja yang terpapar kebisingan keras (≥ 85 dB(A)). Frekuensi, intensitas, durasi paparan, tipe bising dapat mengganggu aktivitas tubuh: sistem pendengaran dan non-pendengaran. Hipertensi merupakan penyakit multi-faktor yang dapat dipicu dari berbagai sumber seperti paparan bising. Sehingga penelitian ini dilakukan dengan tujuan untuk mengetahui hubungan paparan bising dengan hipertensi. METODE Penelitian ini dilakukan dengan menggunakan metode analitik observasional dengan pendekatan potong lintang yang mengikutsertakan 78 responden laki-laki dari 22-53 tahun di PT. X pabrik kabel yang beroperasi di Jawa Barat. Alat pengumpul data berupa kuesioner sosiodemografi, spyghmomanometer, microtoise, timbangan berat badan. Variabel yang diteliti adalah hipertensi, intensitas bising, tipe bising, masa kerja, usia, jumlah konsumsi rokok. Analisis data menggunakan uji chi-square dan Kolmogorov-Smirnov dengan SPSS (p<0,05). HASIL Didapatkan tekanan darah normal 23,1%, pre-hipertensi 39,7%, hipertensi 1 25,6%, hipertensi 2 11,5%.Intensitas bising <85 dB(A) 32,1%, ≥85 dB(A) 67,9%. Tipe bising kontinu 12,8%, intermitten 80,8%, impulsif 6,4%. Masa kerja <10 tahun 23,1%, ≥10 tahun 76,9%. Pekerja tidak merokok 35,9%, merokok 1-10 batang 24,4%, >10 batang 39,7%. KESIMPULAN Terdapat hubungan bermakna antara intensitas bising dan usia dengan hipertensi pada karyawan pabrik industri kabel (p=0,007; p=0,019). Tidak terdapat hubungan bermakna antara tipe bising, masa kerja, dan jumlah konsumsi rokok dengan hipertensi (p=0,281; p=0,139; p=0,257). : Background: Multiple harvest explant method (MHEM) derived umbilical cord mesenchymal stem cells (UC-MSCs) was shown to undergo senescent beginning at passage-10 (P-10). However, for the same cells, there are no senescent data after cryopreservation and passage. Therefore, this study aimed to analyze the senescent profile of cryopreserved MHEM derived UC-MSCs after serial passages. Methods: MHEM derived UC-MSCs were isolated and cultured in platelet lysate (PL) containing medium as described previously. The cells were cryopreserved at passage-1 (P-1) in 10% dimethyl sulfoxide (DMSO) and 10% PL containing alpha minimal essential medium (αMEM). Cell density at cryopreservation was 500 000 cells/mL. After one month, the cells were thawed and recultured until P-8 in six 12-well plate. At 80-90% confluent, two wells were harvested and the cells were recultured into six wells, and the four remaining wells were subjected to senescent (β-galactosidase) staining, which was done for all passages. Random photographs were taken from all stained wells, and senescent percentage was recorded. Results: No senescent cells were observed at P-2. Senescent cells began to appear at P-3. Percentage (mean ± SD) of senescent cells from P-3 through P-8 were 0.04 ± 0.02, 1.18 ± 1.82, 0.14 ± 0.18, 0.49 ± 0.00, 0.58 ±0.91, and 0.07 ± 0.07, respectively. Results: No senescent cells were observed at P-2. Senescent cells began to appear at P-3. Percentage (mean ± SD) of senescent cells from P-3 through P-8 were 0.04 ± 0.02, 1.18 ± 1.82, 0.14 ± 0.18, 0.49 ± 0.00, 0.58 ±0.91, and 0.07 ± 0.07, respectively. Conclusion: No senescent cells were observed at P-2 (cumulative population doubling [CPD] > 9.38). Senescent cells began to appear at P-3 (CPD > 14.06), but in all passages until P-8 (CPD >34.34) the senescent percentage was below 5%.
Aortic enlargement is not an uncommon incidental finding with potential omission of clinical implications. During routine dissection in the middle of the thorax, there was an incidental finding of a disproportionately enlarged aorta in comparison to normal-sized heart at gross observation despite normal anatomy in local and adjacent structures. Incidental finding of aortic enlargement should not be disregarded as it might contribute to pathological processes. The use of indices could give a more comprehensive appraisal of cardiovascular system dimensions.
