Bu çalışmada Elazığ iline bağlı Merkez ve Maden ilçelerindeki domates ve biber alanlarında fitoplazma hastalık etmenlerinin durumları belirlenmiştir. Güdümlü örnekleme yöntemine göre yapılan bu sörveyler sonucu 27 bitki örneği toplanmıştır. Moleküler analizler Phytoplasma F, Phytoplasma R ve Phytoplasma Probe üniversal primerleri kullanılarak qPCR ve R16mF2/R16mR1 ile R162Fn/R16R2 üniversal primerleri kullanılarak Nested qPCR ile gerçekleştirilmiştir. Moleküler analizler sonucunda 4 örneğin fitoplazma ile enfekteli olduğu belirlenmiştir. Fitoplazma örneklerinin 16S rDNA bölgesinden elde edilen 1.25 kb’lik amplifikasyon ürünlerine BLAST ve RFLP analizleri uygulanmıştır. Bu analizler sonucunda domates alanlarında tespit edilen 2 örneğin Candidatus phytoplasma solani (ELZ 5, Acc. No:ON171767; ELZ 17, Acc. No:ON171769) ve 1 örneğin Candidatus phytoplasma trifolii (ELZ 25, Acc. No:ON171770) olduğu, biber üretim alanlarında tespit edilen 1 örneğin ise Candidatus phytoplasma trifolii (ELZ 14, Acc. No:ON171768) olduğu belirlenmiştir. Fitoplazma izolatlarına ait 16S rDNA dizilerinin BLAST karşılaştırmasında dünyadaki diğer fitoplazma izolatları ile %99-100 arasında benzerlik gösterdiği, yapılan filogenetik analizler sonucunda ise elde edilen izolatların referans izolatları ile aynı grupta yer aldığı tespit edilmiştir.
