P34 is an antimicrobial peptide produced by a Bacillus sp. strain isolated from the intestinal contents of a fish in the Brazilian Amazon basin with reported antibacterial activity. The aim of this work was to evaluate the peptide P34 for its in vitro antiviral properties against canine adenovirus type 2 (CAV-2), canine coronavirus (CCoV), canine distemper virus (CDV), canine parvovirus type 2 (CPV-2), equine arteritis virus (EAV), equine influenza virus (EIV), feline calicivirus (FCV) and feline herpesvirus type 1 (FHV-1). The results showed that the peptide P34 exhibited antiviral activity against EAV and FHV-1. The peptide P34 inhibited the replication of EAV by 99.9% and FHV-1 by 94.4%. Virucidal activity was detected only against EAV. When P34 and EAV were incubated for 6 h at 37 °C the viral titer reduced from 104.5 TCID50 to 102.75 TCID50, showing a percent of inhibition of 98.6%. In conclusion, our results demonstrated that P34 inhibited EAV and FHV-1 replication in infected cell cultures and it showed virucidal activity against EAV. Since there is documented resistance to the current drugs used against herpesviruses and there is no treatment for equine viral arteritis, it is advisable to search for new antiviral compounds to overcome these infections.
O vírus da imunodeficiência felina (FIV) pertence à família Retroviridae e apresenta estrutura molecular e patogenia similar ao vírus da imunodeficiência humana (HIV). A infecção pelo FIV pode determinar síndrome de imunodeficiência adquirida em gatos (AIDS felina). A eficácia do tratamento em humanos com HIV é obtida com uma combinação de drogas com diferentes mecanismos de ação que promovem a inibição de fases críticas do ciclo de replicação dos retrovírus, sendo tal esquema terapêutico denominado pela sigla HAART (Highly Active Antiretroviral Therapy). Antirretrovirais como zidovudina, lamivudina, fozivudina, estampidina, TL3 e plerixafor foram testados in vivo frente ao FIV no regime de monoterapia e induziram supressão de carga viral. Seguindo a tendência de tratamento anti-HIV, futuros estudos devem testar fármacos antirretrovirais em regime de associação, com o propósito de desenvolver um protocolo HAART em medicina felina. A presente revisão tem o objetivo de discutir e propor condutas na terapia antirretroviral (TARV) em felinos, além de apresentar os principais fármacos antirretrovirais com eficácia comprovada frente ao FIV.
O peptídeo antimicrobiano identificado como P34 foi obtido a partir de uma nova espécie de Bacillus isolada do conteúdo intestinal do peixe Piau-com-pinta (Leporinus sp.), presente na região da Bacia Amazônica. A atividade do P34 já foi demonstrada in vitro contra Listeria monocytogenes e herpesvírus bovino. Entretanto, para o desenvolvimento de novos agentes terapêuticos é essencial, inicialmente, uma avaliação dos seus níveis de toxicidade. O presente trabalho relata a avaliação da citotoxicidade do peptídeo P34 in vitro, usando o ensaio de vermelho neutro como método de determinação da viabilidade celular. Células CRFK (Crandell rees feline kidney), RK13 (Rabbit kidney), VERO (African green monkey kidney), MDCK (Madin-Darby canine kidney) e MDBK (Madin-Darby bovine kidney) foram cultivadas durante 24 horas e posteriormente expostas ao antimicrobiano P34, em diferentes concentrações, por 48 horas. A análise pelo ensaio de vermelho neutro demonstrou diferentes níveis de toxicidade, variando de acordo com a linhagem celular avaliada. As linhagens que permitiram uma concentração mais elevada do peptídeo P34 foram a RK13, MDBK e MDCK, com concentrações de 1,15 µg.ml-1, 1,07 µg.ml-1 e 1,0 µg.ml-1. Já as linhagens VERO e CRFK toleraram uma menor concentração do peptídeo para não apresentarem efeito citotóxico, sendo as concentrações de 0,75 µg.ml-1 e 0,5 µg.ml-1, respectivamente.
Previous studies have demonstrated the antimicrobial activity of the peptide P34. In this study, the antiviral potential of P34 and the in vitro mechanism of action were investigated against bovine alphaherpesvirus type 1 (BoHV1). P34 exhibited low toxicity, a high selectivity index (22.9) and a percentage of inhibition of up to 100% in MDBK cells. Results from antiviral assays indicated that P34 did not interact with cell receptors, but it was able to inhibit the viral penetration immediately after pre-adsorption. In addition, BoHV1 growth curve in MDBK cells in the presence of P34 revealed a significant reduction in virus titer only 8h post-infection, also suggesting an important role at late stages of the replicative cycle. Virucidal effect was observed only in cytotoxic concentrations of the peptide. These findings showed that the antimicrobial peptide P34 may be considered as a potential novel inhibitor of in vitro herpesviruses and must encourage further investigation of its antiherpetic activity in animal models as well as against a wide spectrum of viruses.
Dentre as propriedades biológicas das moléculas derivadas da classe da 4-tiazolidinona, a atividade antiviral tem atraído a atenção dos pesquisadores. Neste estudo, foi determinada a citotoxicidade e a atividade antiviral de cinco compostos derivados da 2-picolilamina da classe da 4-tiazolidinona (V19, V20, V23, V28 e V29) frente ao vírus da diarreia viral bovina (BVDV) em células MDBK (Madin-Darby Bovine Kidney). A atividade antiviral foi determinada pela comparação entre os títulos obtidos pelo método de diluição limitante na presença ou ausência de cada molécula. As moléculas V20 e V28 na maior concentração não citotóxica foram capazes de inibir a produção das partículas virais, reduzindo os títulos virais quando comparadas com o controle, e resultaram em um percentual de inibição de 78% (p<0,01) contra o BVDV. As moléculas V20 e V28 foram consideradas não tóxicas nas concentrações 0,063 mg/mL e 0,07 mg/mL, respectivamente, quando avaliadas através do ensaio de vermelho neutro como método de determinação da viabilidade celular. As outras tiazolidinonas avaliadas (V19, V23 e V29) não demonstraram atividade contra o BVDV e foram tóxicas acima de 0,078 mg/mL. Em conclusão, os compostos V20 e V28 derivados da 2-picolilamina da classe da 4-tiazolidinona apresentaram atividade antiviral frente ao BVDV, o que os tornam alvos para mais estudos visando elucidar seus mecanismos de ação antiviral.
Felid alphaherpesvirus type 1 (FHV-1) is an important cause of respiratory and ocular diseases in cats worldwide. Mice have been widely used to study the pathogenesis of several human and animal viruses, especially herpesviruses. This study aimed to verify whether BALB/c mice are susceptible to FHV-1 infection. The animals were intranasally inoculated with FHV-1 and their clinical signs were observed from 3 days post-infection (dpi). At 10 dpi, the animals were euthanized and the lungs, liver, spleen, and kidneys were collected for histopathological examination and quantitative polymerase chain reaction. The results showed that mice were infected with FHV-1 and reproduced several features of the disease observed in its natural host. Histological lesions and viral DNA were found in all sampled tissues, with a higher frequency of FHV-1 DNA copies detected in the lungs. All mice were seroconverted to FHV-1 at 7 dpi. To our knowledge, this is the first report of experimental infection of BALB/c mice with FHV-1. Our findings demonstrate that this murine model can contribute to understanding of FHV-1 pathogenesis and may be useful for trials against this virus.
