Des expériences de microponctions tubulaires réalisées chez le Rat dans différents états (animaux non diurétiques, en diurèse saline, en diurèse par le mannitol et en diurèse par I’urée) sont rapportées dans ce travail. Pour chaque microponction, ont été déterminés: le débit tubulaire, les concentrations de I’inuline marquée, du sodium et du potassium ainsi que la pression osmotique. Les résultats fournis par 236 échantillons de fluide tubulaire proximal ou distal apparaissent, dans le cas du sodium et de I’inuline, en accord avec ceux précédemment obtenus dans ces types de conditions expérimentales. Pour le potassium, sa concentration reste voisine de celle du plasma le long du tubule contourné proximal dans tous les états étudiés, malgré une réabsorption fractionnaire de I’eau très variable d’un état à I’autre; de plus, il existe une indication de sécrétion tubulaire de potassium dans la portion terminale des néphrons en diurèse par l’urée et par le mannitol. En ce qui concerne la pression osmotique, une hypoosmolarité franche a été trouvée dans tous les échantillons distaux ponctionnés à un débit ne dépassant pas le débit tubulaire; le rapport (F/P)osm moyen le plus bas a été note dans le groupe des Rats non diurétiques. Enfin, le taux de filtration glomérulaire des néphrons ponctionnés a été trouvé sensiblement égal dans tous les groupes (moyennes de 30 à 32 nl/min/glomérule) malgré une clearance rénale de I’inuline variable d’un état à I’autre (et notamment d’un facteur 2 entre le groupe des Rats en diurèse à I’urée et le groupe des Rats en diurèse saline).
Les concentrations des ions sodium, potassium, chlore (dans quelques cas), de I’inuline 3H méthoxy et de I’urée 14C, ainsi que la pression osmotique ont été mesurées dans des échantillons de microponctions proximales et distales, dans le plasma et dans I’urine urétérale de Psammomys obesus non diurétiques. Les résultats indiquent chez cette espèce désertique et par rapport au Rat: 1. une filtration glomérulaire par néphron faible (6 nl/min/glomérule); 2. une réabsorption fractionnaire de I’eau à la fin du tubule contourné proximal basse (F/P) in de I’ordre de 2, en rapport avec la faible longueur de ce segment; 3. un comportement du sodium, du potassium et de la pression osmotique le long du néphron comparable à celui observé chez le Rat; 4. un rapport moyen (F/P) in de 3,3 à I’entrée du tubule distal indiquant que la réabsorption nette d’eau dans l’anse de Henle est limitée; 5. un débit d’urée à I’entrée du tubule distal en moyenne 2,3 fois supérieur à son debit de filtration indiquant une importante addition nette d’urée dans I’anse de Henle. Ces deux derniers résultats sont discutés en relation avec le mécanisme médullaire de concentration de I’urine, puisque chez le Psammomys les néphrons accessibles à la surface du cortex possèdent tous des anses longues.
Le comportement de l’Inuline-3H, de l’Urée-14C, du Sodium, du Potassium et de la pression osmotique le long des tubules corticaux accessibles a été étudié par microponction chez trois lignées de souris (RAP, CBA, Pérou) en diurèse saline. Dans l’ensemble, les résultats obtenus chez cette espèce ne sont pas qualitativement différents de ceux rapportés antérieurement pour d’autres espèces de rongeurs places dans des conditions expérimentales voisines. Les differences les plus notables observées résultent de la petite taille des reins et de la réduction de la longueur des néphrons; ainsi, les tubules proximaux superficiels (pars recta comprise) sont courts (Pérou: 3,8 mm, CBA: 6,2 mm, RAP: 6,5 mm); corrélativement, les taux de filtration mesurés par néphron sont bas (respectivement 5,8, 8,5 et 12,4 nl/min) et la reabsorption fractionnaire de l’eau est limitée le long du tubule proximal: (F/PIn) = 2 à la fin de la portion accessible. En outre, dans les conditions de ces expériences, on observe chez les 3 lignées de souris une excrétion tubulaire de potassium dans la portion terminale des néphrons ainsi qu’un «recyclage» médullaire d’urée entre canaux collecteurs et anses de Henle attesté par une addition nette d’urée entre le tubule proximal et le tubule distal (F/P)urée/inuiine = 0,7 environ à la fin du proximal et 1,9 (RAP), 0,9 (CBA) et 1,34 (Pérou) dans le tubule distal.
