Background Many agronomic traits have been bred into modern wheat varieties, but wheat ( Triticum aestivum L.) continues to be vulnerable to heat stress, with high night-time temperature (HNT) stress shown to have large negative impact on yield and quality. Global mean temperature during the day is consistently warming with the minimum night temperature increasing at a much quicker pace. Currently, there is no system or method that allows crop scientists to impose HNT stress at key developmental stages on wheat or crops in general under field conditions, involving diverse genotypes and maintaining a dynamic temperature differential within the tents compared to the outside. Results Through implementation of a side roll up and a top ventilation system, heaters, and a custom cyber-physical system using a Raspberry Pi, the heat tents were able to consistently maintain an elevated temperature through the night to differentiate heat stress impact on different genotypes. When the tents were placed in their day-time setting they were able to maintain ambient day-time temperature without having to be removed and replaced on the plots. Data averaged from multiple sensors over three consecutive weeks resulted in a consistent but small temperature difference of 0.25 °C within the tents, indicating even distribution of heat. While targeting a temperature differential of 4 °C, the tents were able to maintain an average differential of 3.2 °C consistently throughout the night-time heat stress period, compared to the outside ambient conditions. The impact of HNT stress was confirmed through a statistically significant yield reduction in eleven of the twelve genotypes tested. The average yield under HNT stress was reduced by 20.3% compared to the controls, with the highest reduction being 41.4% and a lowest reduction of 6.9%. Recommendations for fine-tuning the system are provided. Conclusion This methodology is easily accessible and can be widely utilized due to its flexibility and ease of construction. This system can be modified and improved based on some of the recommendations and has the potential to be used across other crops or plants as it is not reliant on access to any hardwired utilities. The method tested will help the crop community to quantify the impact of HNT stress, identify novel donors that induce tolerance to HNT and help the breeders develop crop varieties that are resilient to changing climate. Sorghum is often exposed to suboptimal low temperature stress under field conditions, particularly at the seedling establishment stage. Enhancing chilling tolerance will facilitate earlier planting and so minimize the negative impacts of other stresses experienced at later growth stages. Genome-wide association mapping was performed on a sorghum association panel grown under control (30/20 °C; day/night) and chilling (20/10 °C) conditions. Genomic regions on chromosome 7, controlling the emergence index and seedling (root and shoot) vigor, were associated with increased chilling tolerance but they did not co-localize with undesirable tannin content quantitative trait loci (QTLs). Shoot and root samples from highly contrasting haplotype pairs expressing differential responses to chilling stress were used to identify candidate genes. Three candidate genes (an alpha/beta hydrolase domain protein, a DnaJ/Hsp40 motif-containing protein, and a YTH domain-containing RNA-binding protein) were expressed at significantly higher levels under chilling stress in the tolerant haplotype compared with the sensitive haplotype and BTx623. Moreover, two CBF/DREB1A transcription factors on chromosome 2 showed a divergent response to chilling in the contrasting haplotypes. These studies identify haplotype differences on chromosome 7 that modulate chilling tolerance by either regulating CBF or feeding back into this signaling pathway. We have identified new candidate genes that will be useful markers in ongoing efforts to develop tannin-free chilling-tolerant sorghum hybrids.

Plant leaves grow and change their orientation as well their emission of chlorophyll fluorescence in time. All these dynamic plant properties can be semi-automatically monitored by a 3D imaging system that generates plant models by the method of coded light illumination, fluorescence imaging and computer 3D reconstruction. Here, we describe the essentials of the method, as well as the system hardware. We show that the technique can reconstruct, with a high fidelity, the leaf size, the leaf angle and the plant height. The method fails with wilted plants when leaves overlap obscuring their true area. This effect, naturally, also interferes when the method is applied to measure plant growth under water stress. The method is, however, very potent in capturing the plant dynamics under mild stress and without stress. The 3D reconstruction is also highly effective in correcting geometrical factors that distort measurements of chlorophyll fluorescence emission of naturally positioned plant leaves.

Abstract The rate of light-induced inactivation of photosystem II (PSII) at 22 and 4 °C was measured from natural accessions of Arabidopsis thaliana (Rschew, Tenela, Columbia-0, Coimbra) grown under optimal conditions (21 °C), and at 4 °C from plants shifted to 4 °C for 2 weeks. Measurements were done in the absence and presence of lincomycin (to block repair). PSII activity was assayed with the chlorophyll a fluorescence parameter F v / F m and with light-saturated rate of oxygen evolution using a quinone acceptor. When grown at 21 °C, Rschew was the most tolerant to photoinhibition and Coimbra the least. Damage to PSII, judged from fitting the decrease in oxygen evolution or F v / F m to a first-order equation, proceeded more slowly or equally at 4 than at 22 °C. The 2-week cold-treatment decreased photoinhibition at 4 °C consistently in Columbia-0 and Coimbra, whereas in Rschew and Tenela the results depended on the method used to assay photoinhibition. The rate of singlet oxygen production by isolated thylakoid membranes, measured with histidine, stayed the same or slightly decreased with decreasing temperature. On the other hand, measurements of singlet oxygen from leaves with Singlet Oxygen Sensor Green suggest that in vivo more singlet oxygen is produced at 4 °C. Under high light, the PSII electron acceptor Q A was more reduced at 4 than at 22 °C. Singlet oxygen production, in vitro or in vivo, did not decrease due to the cold-treatment. Epidermal flavonols increased during the cold-treatment and, in Columbia-0 and Coimbra, the amount correlated with photoinhibition tolerance. Increasing temperatures can severely affect wheat (Triticum aestivum L.) production, particularly when it coincides with the grain-filling period. Heat stress induces rapid senescence resulting in early maturity and shortened grain-filling period. In this study, the applicability of in vivo chlorophyll fluorescence (Chl-F) and chlorophyll index to track rate of senescence in flag leaves and spikes exposed to heat stress were investigated. Seven winter wheat varieties were exposed to post-flowering heat stress using growth chambers [35/15 • C (heat stress) and 25/15 • C (control) day/night] and unique field-based heat tents (imposed +6 • C higher than ambient). Effective quantum yield of photosystem II (PSII) (QY) was recorded temporally in flag leaves and spikes, and compared with in vitro chlorophyll-a (Chl-a) concentration and non-invasive estimation of chlorophyll and flavonoids index. Time point indicating the start of senescence (changepoint, CP) for QY was advanced by 0-8 and 0-6 d in flag leaves and spikes, respectively, under heat stress. In the chamber experiment, sustained heat stress induced accelerated decline of QY, particularly in wheat cultivars Larry and WB4458. Stronger positive relationship between days to senescence in spikes and thousand kernel weight indicated an extended period of assimilate supply from sink compared to the source tissue, during grain filling. Capturing heat stressinduced changes in photosynthetic pigments and QY at high temporal frequency presents an effective phenotyping approach for testing genetic diversity in largescale field experiments involving different crops.

Response of rice (Oryza sativa) exposed to both biotic and abiotic stresses can be quantified by employing fast and accurate optical methods. In this study, the overall stress responses of (i) 12 near-isogenic lines (NILs) in the genetic background of the rice blast-susceptible cultivar Lijiangxintuanheigu (LTH) and (ii) four NILs in the genetic background of the bacterial blight-susceptible cultivar IR24, were inspected by means of Chl fluorescence (Chl-F) imaging. The distribution of the maximum and effective quantum yield of PSII (Fv/FM and QY) and steady-state Chl-F (Ft) were found to be effective in differentiating symptomatic leaf tissue for both rice blast and bacterial blight, which correlated well with 30 cycles of rice blast and six cycles of bacterial blight previously screened using classical (manual) approaches. Subsequently, identified Chl-F parameters allowing detection under ambient light (QY and Ft) were tested across both biotic and abiotic (drought) stress experiments, for rice cultivars contrasting for drought stress response (N22, IR64 and NSIC Rc 222). Their applicability has been proven for both rice blast and bacterial blight; however, QY failed to detect the effect of drought. In addition to Chl-F, the usefulness of 11 selected vegetation indices (Vis) was tested on these three cultivars exposed to particular stresses: (i) rice blast was detectable by Vis calculated from the visible spectrum; (ii) bacterial blight by near-infrared-related Vis; and (iii) drought by Vis calculated from the visible spectrum. The key Chl-F parameters and/or Vis have been summarized and discussed.
