Membrane-enriched homogenates of fresh and cultured (48 h) porcine lymphocytes were assayed for the presence of specific LHRH receptors by saturation and displacement analysis using [D-Ser-(TBU)6, des-Gly-NH2(10)] LHRH-EA as the labeled and unlabeled ligand. Membrane-enriched homogenates of porcine pituitaries served as positive controls while porcine granulosa cell membranes and crude liver homogenates served as negative controls. Specific high-affinity LHRH receptors were found in porcine pituitaries (Kd = 0.3 nM) and cultured lymphocytes (Kd = 13 nM) but not in fresh lymphocytes. No specific binding was observed in negative control tissues. Porcine lymphocytes have measurable high-affinity LHRH receptors after 48 h of culture.
Resumo -O objetivo deste trabalho foi avaliar a ação imunoesterilizadora de uma vacina anti-hormônio liberador de hormônio luteinizante (LHRH), composta por ovalbumina-LHRH-7 e tiorredoxina-LHRH-7, em touros mestiços Nelore. Vinte e seis touros, com dois anos de idade, foram distribuídos aleatoriamente em dois grupos de 13 animais. No grupo I, os animais receberam uma dose e dois reforços da vacina nos dias 0, 141, e 287 do experimento. No grupo II, os animais não receberam nenhum tratamento (controle). Para avaliar o efeito da vacina nos touros, foi realizada a mensuração da circunferência escrotal no início do experimento e no dia do abate, 741 dias depois. Por ocasião do abate, também foi coletada uma amostra dos testículos para avaliação histológica. O grupo imunizado apresentou circunferência escrotal ao abate de 22±5,98 cm, menor do que a do grupo controle que foi de 35,6±2,4 cm. Na análise histológica dos animais do grupo imunizado, foi observada degeneração testicular com ausência de espermatozoides em 85% dos animais avaliados, os outros 15% apresentaram redução no número de espermatozoides, em comparação aos animais do grupo controle. A vacina anti-LHRH, com fusão de proteínas, é efetiva na castração imunológica de touros e deve ser considerada como alternativa para utilização na produção bovina extensiva no Brasil.Termos para indexação: Bos taurus indicus, imunoesterilização, LHRH, touros Nelore. Testicular characteristics of bulls immunosterilized with anti-luteinizing hormone-releasing hormone vaccineAbstract -The objective of this study was to evaluate the immunosterilization action of the anti-luteinizing hormone-releasing hormone (LHRH) vaccine, composed with ovalbumin-LHRH-7 and thioredoxin-LHRH-7, in Nelore-cross bulls. Twenty-six 2-year old bulls were randomly assigned in two groups of 13 animals each. The animals of group I received a primary and two booster injections of the vaccine on days 0, 141, and 287 of the experiment. In group II, the control group, the bulls did not receive any type of treatment. Scrotal circumference was measured in the beginning of the experiment and at slaughter, after 741 days. A testicular sample was collected at slaughter and processed for histological evaluation. The scrotal circumference in the immunized group was smaller, 22±5.98 cm, compared to the one of the control group (35.6±2.4 cm), at slaughter. Histological analyses of the testis showed testicular degeneration and absence of sperms in 85% of the animals in the treated group, and a reduced number of sperms in the other 15%, in comparison to the control group. The vaccine anti-LHRH with protein fusion is effective in immune castration of bulls and should be considered as an alternative tool for extensive livestock production in Brazil.
This study was conducted to determine the effects of testosterone-propionate exposure during fetal development on sexual differentiation and growth rates in heifers. Ten pregnant cows were given subcutaneous injections of testosterone-propionate (250 mg/injection) every other day during d 40 to 60 of gestation. Four cows aborted after the end of testosterone treatment, while four heifers (androgenized females) and two bulls (androgenized males) were produced from the six remaining pregnant, testosterone-propionate treated cows. Calves from cows that did not receive exogenous hormone treatment were used as controls. At 8 mo of age, the androgenized heifers and control heifers and control steers were challenged with 1 mg estradiol-17 beta to induce a preovulatory luteinizing hormone (LH) surge. Two weeks later, pituitary responsiveness to exogenous luteinizing hormone releasing hormone (LHRH; 75 micrograms) was evaluated in androgenized heifers and in control heifers and control steers. To monitor growth rates, all animals were weighed at 28-d intervals from birth to 380 d of age. Androgenized females exhibited a partially masculinized phenotype as well as internal male reproductive structures. Treatment with estradiol-17 beta first depressed (P less than .05) serum LH concentrations in all animals, then induced (P less than .05) a preovulatory-like LH surge in control and androgenized females. Control steers did not (P greater than .05) exhibit a preovulatory-like LH surge following administration of estradiol-17 beta. Exogenous LHRH treatment stimulated peak LH concentrations (P less than .05) to a greater extent in control and androgenized females than in control steers.(ABSTRACT TRUNCATED AT 250 WORDS)
Nipple fruit (Solanum mammosum) has been considered to have great pharmaceutical potential because of its high amounts of solamargine and solasonine. This study aimed to examine the effect of nipple fruit at different concentrations (0.5%, 1%, and 2% (w/v)) on the viability, acid, bile, lysozyme, and gastric juice tolerance, and protease activity of Lactobacillus acidophilus LA K. The viability was studied in MRS broth. Acid tolerance was determined by adjusting the pH to 2, whereas bile tolerance was examined with oxgall 0.3% (w/v) in MRS broth. Lysozyme resistance was investigated in an electrolyte solution with lysozyme (100 mg/L), while gastric juice tolerance was analyzed with pepsin and NaCl. Protease activity was determined spectrophotometrically at 340 nm in skim milk with o-phthaldialdehyde reagent. L. acidophilus LA K was incubated anaerobically (37 °C). Microbial growth was determined every 2 h for 10 h of incubation. Acid tolerance was determined at 0, 5, and 15 min, whereas bile tolerance was analyzed at 0, 4, and 8 h of incubation. Lysozyme tolerance was determined at 0, 1, and 2 h of incubation, while gastric juice tolerance was determined at pH 2, 3, 4, 5, and 7. Protease activity was evaluated at 0, 12, and 24 h incubation. Nipple fruit’s chemical and bioactive compounds were also examined to discuss their impact on the survival of L. acidophilus LA K. Nipple fruit did not affect microbial growth, bile, and acid tolerance. Nipple fruit at 2% had higher survivability on the simulated gastric juice and lysozyme resistance and increased protease activity.
