Was evaluated the pattern of growth among females and males of tambaqui by Gompertz nonlinear regression model. Five traits of economic importance were measured on 145 animals during the three years, totaling 981 morphometric data analyzed. Different curves were adjusted between males and females for body weight, height and head length and only one curve was adjusted to the width and body length. The asymptotic weight (a) and relative growth rate to maturity (k) were different between sexes in animals with ± 5 kg; slaughter weight practiced by a specific niche market, very profitable. However, there was no difference between males and females up to ± 2 kg; slaughter weight established to supply the bigger consumer market. Females showed weight greater than males (± 280 g), which are more suitable for fish farming purposes defined for the niche market to larger animals. In general, males had lower maximum growth rate (8.66 g / day) than females (9.34 g / day), however, reached faster than females, 476 and 486 days growth rate, respectively. The height and length body are the traits that contributed most to the weight at 516 days (P <0.001).
The objective of this study was to evaluate the genetic diversity and the parental contribution of Piaractus mesopotamicus in the production of offspring in the semi-natural system of reproduction. Twenty parental fishes (eleven males and nine females) and the total of 100 larvae were evaluated by microsatellite marker. The parents and offspring had thirty-one alleles and heterozygosity of 0.550 and 0.563, respectively. The females were fertilised by two up to six males while the males fertilised three up to five females. The contribution of the females and males to the offspring were 66.6 and 58%, respectively. Such results indicated no loss in the genetic variability in the offspring, and the parents had multiple paternity and reasonable contribution to the offspring production.
Qualitative parameters of piapara semen (Leporinus elongatus) were evaluated before and after hormonal induction with carp pituitary extract at 2.5 mg.kg -1 of live weight. The progressive motility, the spermatic vigor and the lifetime of the spermatozoa were higher before the hormonal induction (P > 0.05). The percentage of normal spermatozoa and spermatozoa with secondary pathologies did not differ (P > 0.05) between treatments: before induction (44.0 and 44.4%, respectively) and after-induction (44.3 and 46.7%, respectively). However, the percentage of primary pathologies was higher (P < 0.05) for the semen collected before induction than for the semen collected after induction; the estimates were 12.2 and 8.0%, respectively. The most frequent pathologies were the taillessness with the frequencies of 27.4 and 36.3% followed by the headlessness for which the estimates were 10.1 and 3.9%, before and after induction respectively. The semen collected before the hormonal induction presented better qualitative parameters.Keywords: spermatic cells, spermatic morphology, fish. Parâmetros qualitativos do sêmen de piapara (Leporinus elongatus Valenciennes, 1850) ResumoOs parâmetros qualitativos do sêmen de piaparas (Leporinus elongatus) foram avaliados antes e após a indução hormonal com o extrato de hipófise de carpa na dosagem 2,5 mg.kg -1 de peso vivo. A motilidade progressiva, o vigor espermático e o tempo de vida dos espermatozóides apresentaram valores superiores (P < 0,05) no sêmen coletado antes da indução hormonal. Já a estimativa da porcentagem de espermatozóides normais ou com patologias leves, na pré-indução (44,0 e 44,4%, respectivamente) e na pós-indução hormonal (44,3 e 46,7%, respectivamente) não diferiram (P > 0,05) entre os tratamentos. Porém a estimativa de patologias graves foi maior (P < 0,05) no sêmen de pré-indução do que no pós-induzido com 12,2 e 8%, respectivamente. A patologia mais freqüente foi cabeça solta com 27,4 e 36,3% seguida por cauda solta com 10,1 e 3,9%, antes e após a indução hormonal respectivamente. De acordo com os resultados, o sêmen de piapara coletado na pré-indução hormonal apresentou os melhores parâmetros qualitativos.Palavras-chave: células espermáticas, morfologia espermática, peixes.
RESUMO O objetivo deste trabalho foi avaliar o efeito do diluente com dimetilsulfóxido (DMSO) sobre a estrutura e funcionalidade do espermatozóide de tambaqui Colossoma macropomum criopreservado. O sêmen foi diluído 1/9 (v/v) em Beltsville Thawing Solution acrescido de 5, 10, 15 e 20% de DMSO. Após dois minutos foi congelado em botijão dry shipper (-76°C/12h), sendo armazenado em botijão de NL (-196°C). Após 15 dias o sêmen criopreservado foi avaliado quanto às taxas de motilidade, latência, fertilização, eclosão, integridade de membrana e de DNA, funcionalidade mitocondrial e viabilidade. As taxas de fertilização e eclosão com 10% DMSO não diferiram (P > 0,05) do sêmen fresco. Os diluentes com 5 e 10% de DMSO foram superiores (P < 0,05), nas avaliações de motilidade, latência e integridade de DNA do espermatozóide, às concentrações 15 e 20%. Entretanto, a viabilidade celular nas concentrações 5 e 10% de DMSO foi inferior (P < 0,05) à obtida com 20% de DMSO, enquanto com 15% de DMSO não diferiu (P > 0,05) das demais concentrações. Considerando os resultados in vivo e in vitro, nas condições experimentais relatadas, o DMSO na concentração de 10% foi associado à melhor manutenção da qualidade de sêmen criopreservado de Tambaqui.
Cryoprotectant solutions are used to protect the sperm from alterations caused by the low temperature in the cryopreservation process. We evaluated the quality of Colossoma macropomum semen after freezing, using dimethyl sulfoxide (DMSO) as a cryoprotectant, combined with two extender solutions (T1 -Solution 1: Glucose 90.0 g/L, Sodium Citrate 6.0 g/L, EDTA 1.5 g/L, Sodium Bicarbonate 1.5 g/L, Potassium Chloride 0.8 g/L, Gentamycin Sulphate 0.2 g/L, and T2 -Solution 2: Glucose 90.0 g/L, ACP ® -104 10.0 g/L). Motility rate and motility time did not differ between T1 and T2 and were lower than fresh semen. The number of normal sperm was significantly different in treatments T1 (15.1%) and T2 (21.9%), and both showed a reduction in the percentage of normal sperm compared to fresh semen (57.4%). The values found for the rates of fertilization and hatching, mitochondrial functionality and sperm DNA, did not differ between the treatments (T1 and T2). Regarding membrane integrity, there was a higher percentage of spermatozoa with intact membranes in T1 (53.4%) than T2 (43.7%). The extender solutions, combined with 10% DMSO, maintained the sperm DNA intact in almost all the C. macropomum sperm cells, however there was a loss in their functionality.As soluções crioprotetoras são utilizadas para proteger os espermatozoides das alterações causadas por baixas temperaturas durante o processo de criopreservação. Avaliamos a qualidade do sêmen de Colossoma macropomum após o congelamento, utilizando dimetilsulfóxido (DMSO) como crioprotetor, combinado com duas soluções diluidoras (T1 -Solução 1: Glicose 90,0 g/L, Citrato de Sódio 6,0 g/L, EDTA 1,5 g/L, Bicarbonato de Sódio 1,5 g/L, Cloreto de Potássio 0,8 g/L, Sulfato de Gentamicina 0,2 g/L, e T2 -Solução 2: Glicose 90,0 g/L, ACP®-104 10,0 g/L). A taxa de motilidade (%) e o tempo de motilidade (s) não diferiram entre T1 e T2, porém foram mais baixos do que no sêmen fresco. O número de espermatozoides normais foi significativamente diferente nos tratamentos T1 (15,1%) e T2 (21,9%), e ambos mostraram uma redução na porcentagem de espermatozoides normais, comparado ao sêmen fresco (57,4%). Os valores encontrados para as taxas de fertilização e eclosão, funcionalidade mitocondrial e DNA do esperma, não diferiram entre os tratamentos (T1 e T2). Para a integridade da membrana, houve uma porcentagem mais elevada de espermatozóides com a membrana intacta em T1 (53,4%) do que T2 (43,7%). As soluções diluentes combinadas com DMSO a 10% preservaram o DNA espermático intacto em quase todas as células do sêmen de C. macropomum, mas houve perda na funcionalidade dos mesmos.
This study aimed to evaluate muscle organization in tambaqui in order to describe the muscle growth process. We analyzed the morphometric pattern of fibers from white muscle of young-adults (300 days) by smaller diameter. The organization of white muscle exhibited a typical morphological pattern found in other fish species. Heavier animals showed higher frequency of larger diameter fibers (>50 μm) and smaller animals had higher frequency of smaller diameter fibers (<20 μm) (P=0.005). However, both animals showed the same frequency of intermediate diameter fibers (20-50 μm). Body weight showed a positive correlation with muscle diameter fiber (r=0.45), being 20-50 μm the diameters that contributed the most to animal weight (P <0.0001). A weak correlation between fiber diameter and animal sex was observed (r=0.2). Females showed higher frequency of large fiber diameters (>50 μm) than males. However, there was no difference between body weight and sex (P=0.8). Our results suggest that muscle growth is by hypertrophy and hyperplasia due to a mosaic appearance from different diameters fibers, which is characteristic of large size fish species.O objetivo deste trabalho foi avaliar a organização muscular em tambaqui, a fim de descrever o processo de crescimento muscular. Foi analisado o padrão morfométrico das fibras do músculo branco de animais com 300 dias de idade usando o método de diâmetro menor. O músculo branco apresentou uma organização morfológica padrão encontrado em peixes. Animais de maior peso apresentaram maior frequência de fibras de maior diâmetro (> 50 μm) e os animais de menor peso apresentaram maior frequência de fibras de menor diâmetro (<20 μm) (P = 0,005). Entretanto, ambos os animais, com maior e menor peso, apresentaram frequências semelhantes de fibras de diâmetro intermediário (20-50 μm). O parâmetro peso corporal mostrou correlação positiva com o diâmetro da fibra muscular (r = 0,45), sendo as fibras de diâmetro intermediários (20-50 μm) que mais contribuíram para o peso do animal (P <0,0001). Fêmeas apresentaram maior frequência de fibras de maior diâmetro (>50 μm) que machos. Observou-se uma fraca correlação entre o diâmetro da fibra e o sexo dos animais (r = 0,2). Apesar de fraca, a correlação estimada é corroborada pela fibras de grandes diâmetros (> 50 μm) serem mais frequente nas fêmeas que nos machos. No entanto, não houve diferença entre o peso corporal dos animais aos 300 dias de idade e sexo (P = 0,8). Os resultados encontrados sugerem que o crescimento muscular ocorre por hipertrofia e hiperplasia, caracterizado pela aparência em mosaico de fibras de diferentes diâmetros, característico de peixes de grande tamanho.
The aim of this study was to evaluate the association between proteins in the seminal plasma of tambaqui Colossoma macropomum (Cuvier, 1818) with seminal quality indicators after thawing. The semen was cryopreserved with a dilution based on BTS with 8% DMSO. A 200 µL sample of semen from each animal was diluted in 800 µL BTS, centrifuged at 800 rpm, and the supernatant was cryopreserved to further analyze of the protein profile of seminal plasma through one-dimensional electrophoresis (SDS-PAGE). After 15 days of cryopreservation, a cryopreserved semen straw was thawed to analyze both qualitative and quantitative parameters. When considering all collections, the SDS-PAGE identified 15 protein bands in the seminal plasma of tambaqui. When the interaction (presence or absence) between proteins observed in the seminal plasma and the post thawed spermatic parameters was evaluated, we observed a great influence of the presence of proteins on spermatic quality. A greater (P<0.05) fertilization rate was observed with the presence of proteins 12, 34, 44, 85, and 90 kDa. Proteins in seminal plasma of tambaqui influenced the spermatic quality after thawing, and thus, they can be utilized as an indicator of sperm quality, especially the proteins with a molecular weight ≤ 50 kDa.
