Oxidative stress is the result of an imbalance between oxidant and antioxidant species, with predominance of oxidative species with harmful action of reactive oxygen species (ROS) and reactive nitrogen species (RNS) on cells. Changes in the levels of nitric oxide (NO • ) can be the cause and/ or a result of various pathophysiological processes. The main objective of this review is to address the relationship between oxidative stress and atherosclerosis in order to better understand the main features of this disease.Keywords: oxidative stress; nitric oxide; atherosclerosis. INTRODUÇÃOA arteriosclerose é uma doença inflamatória que se destaca como principal causa de morbi-mortalidade no mundo. No Brasil, ocorrem aproximadamente 300 mil óbitos/ano, causados por doenças cardiovasculares associadas à arteriosclerose e dados ainda mais assustadores são encontrados nos Estados Unidos, onde as doenças cardiovasculares são responsáveis por cerca de 80% das mortes de pessoas com mais de 65 anos.1-3 Atualmente, as doenças inflamatórias têm sido relacionadas a um desbalanço redox e efeitos do excesso de espécies oxidantes nos organismos. Por isso, tem-se observado um enorme interesse no estudo de antioxidantes devido, principalmente, às descobertas sobre o efeito dos radicais livres no organismo.4 A oxidação é uma parte fundamental da via aeróbica e do metabolismo e, assim, os radicais livres são produzidos naturalmente ou por alguma disfunção biológica. Esses radicais livres, cujo elétron desemparelhado se encontra centrado nos átomos de oxigênio ou nitrogênio, são denominados espécies reativas de oxigênio (ROS), ou espécies reativas de nitrogênio (RNS) ambas radicalares (com um ou mais elétrons desemparelhados) ou não. 5,6 No organismo, os radicais livres encontram-se envolvidos na fagocitose, regulação do crescimento celular, sinalização intercelular e síntese de substâncias biológicas importantes. No entanto, seu excesso acarreta efeitos prejudiciais, tais como a lipoperoxidação de membranas e oxidação de proteínas.4,7 ROS e RNS incluem os radicais livres, como o ânion superóxido, radicais hidroxila e peroxila, bem como espécies não radicalares, como peróxido de hidrogênio, peroxinitrito e oxigênio singlete. As ROS são produzidas continuamente na cadeia respiratória das mitocôndrias com a redução de um elétron do oxigênio e molecular. NAD(P)H oxidase, xantina oxidase, mieloperoxidase, cicloxigenase e lipoxigenase são grandes fontes enzimáticas de ROS em células de mamíferos, enquanto a radiação UV representa um exemplo de um fator ambiental de geração de ROS. 8 Em altas concentrações, ROS e RNS podem danificar macromoléculas celulares, incluindo DNA, proteínas e lipídios, podendo levar à morte celular subsequente. Neonothopanus gardneri (N. gardneri) is a species of bioluminescent fungus belonging to the order Agaricales (Marasmiaceae) found in South America. Its existence was first reported in 1840 by George Gardner in his travels to Brazil, where it is popularly called "coco flower". Found mainly in decaying leaves and in the trunk of dwarf palm trees called "pindoba" (Attalea oleifera) or babaçu (Orbignya phalerata), recently N. gardneri had some of its bioactives isolated and their respective structures elucidated. Thus, this paper aims to present and discuss the findings of the works produced involving this theme. Thus, for the development of this literature review, books and scientific articles were searched in the following databases: Scopus, PubMed, Science Direct, web of science, Royal Society of Chemistry (RSC) Publishing and Google Scholar (1990-2021). The following keywords were used to filter the productions: "Neonothopanus", "Neonothopanus gardneri", "Bioactivities", "Bioprospecting", "Secondary metabolite", "Endophytic" and "bioluminescence". Finally, it is possible to observe that studies involving this species of bioluminescent fungus have focused on explaining the mechanism of light production and its potential biological activities, among them, antitumor, antioxidant, antimicrobial and antileishmanial effects.
AT1 receptor is an interesting biological target involved in several important diseases, such as blood hypertension and cardiovascular pathologies. In this study we investigated the main electrostatic and steric features of a series of AT1 antagonists related to hypertensive activity using structure and ligand-based strategies (docking and CoMFA). The generated 3D model had good internal and external consistency and was used to predict the potency of an external test set. The predicted values of pIC50 are in good agreement with the experimental results of biological activity, indicating that the 3D model can be used to predict the biological property of untested compounds. The electrostatic and steric CoMFA maps showed molecular recognition patterns, which were analyzed with structure-based molecular modeling studies (docking). The most and the least potent compounds docked into the AT1 binding site were subjected to molecular dynamics simulations with the aim to verify the stability and the flexibility of the ligand-receptor interactions. These results provided valuable insights on the electronic/structural requirements to design novel AT1 antagonists.
O Sarcoma 180 (S180) é um tipo de câncer de mama, que está associado à uma alta morbimortalidade e, por isso, a elucidação dos mecanismos moleculares é importante. O avanço da Química Medicinal no planejamento in silico de fármacos e vem despertando o interesse de cientistas e indústrias farmacêuticas. Assim, este trabalho tem como principal objetivo avaliar parâmetros farmacocinéticos (ADME) in silico, por meio do software PreADMET, para ligantes derivados de pirrole de anel A oito, substituídos de duocarmicina B2, com avaliação antitumoral em S180. Os resultados inéditos obtidos corroboram com informações moleculares que são essenciais para prever a biodisponibilidade oral dos potentes candidatos à fármacos.
Antagonists of AT1 receptor are interesting substances to develop drugs that can be used for the treatment of hypertension and other diseases that affect the cardiovascular system. This study investigates the main interactions between various AT1 antagonists and the biological target by applying fragment-based drug design (Hologram QSAR - HQSAR). The proposed HQSAR model yielded significant correlation coefficients (q(2) = 0.764 and r(2) = 0.914), indicating that the method is rigorous and reliable. All models were externally validated using a test set and the results showed good agreement between the experimental and predicted data (r(2) test = 0.740). Therefore, our model can positively contribute to understand the structural features involved in the main interactions between the AT1 antagonists and the residues of the binding site.
