O objetivo desse trabalho foi apresentar uma revisão bibliográfica sobre as tecnologias utilizadas na Genética Forense, enfatizando o uso de marcadores moleculares para a identificação humana. Apresento aqui alguns exemplos do potencial da Biologia Molecular para auxiliar na investigação criminal, bem como na definição de parentesco (maternidade e paternidade). A utilização desses marcadores é atualmente a peça fundamental para os testes de DNA forense. Estes sistemas são, na sua maioria, baseados na análise de painéis de sequências microssatélites especÃficas (STRs). Foi possÃvel discorrer sobre o uso forense do DNA, sobre a presença de regiões hipervariáveis no material genético, o papel de marcadores moleculares, bem como abordar técnicas de análise de DNA e suas aplicações. A busca por novas metodologias se faz importante para reduzir os custos e impulsionar uma nova cultura genética na Ciência Forense, as quais terão impacto no futuro do DNA forense com a expansão da Biologia Molecular.
Terrestrial molluscs can become agricultural pests and transmit parasites, playing an important role in different biological communities. In the present study, we evaluated the diversity and abundance of this group in two horticultural areas in Rio de Janeiro (Manguinhos and Jacarepaguá neighborhoods), as well the presence of parasitic nematodes associated with these molluscs. We collected specimens in the austral spring and summer, with four sites being sampled within each study area, including malabar spinach, sweet potato, chicory greens, and cassava plantations, and one site in an adjacent, non-cultivated area. We collected a total of 522 live mollusc specimens and identified 16 species from 10 different families. The greatest abundance of molluscs was recorded in summer (363) and at Jacarepaguá (309). Overall, 174 (57%) of the 303 specimens analyzed parasitologically were positive for nematodes. Larvae of superfamily Metastrongyloidea, which includes nematodes that are a concern for public health and veterinary medicine, were found parasitizing the slug Sarasinula linguaeformis, in Manguinhos. Our results contribute to understanding of the diversity of terrestrial molluscs occurring in kitchen gardens in the city of Rio de Janeiro, and provide important insights for generating subsidies for health education actions and control of parasitic diseases transmitted by them.
ResumoO objetivo deste trabalho foi realizar uma análise comparativa de laudos oriundos do Instituto de Análise Laboratoriais Forense -IALF, pertencente à Coordenadoria Geral de Perícia do Mato Grosso do Sul em Campo Grande, a fim de observar a presença de vestígio de esperma encontrado nas vestimentas e nas coletas realizadas por meio de suabes de vítimas de estupro. Foram anali sados 895 laudos do período de 2012 a 2015. Os testes estatísticos foram realizados por meio do programa Graphpad Prism 6.0 e os resultados expressos pela diferença entre as médias. A percentagem de amostras positivas oriundas dos exames realizados apenas nas vestes foi estatisticamente significante em relação aos exames realizados nos suabes (p = 0,0063), bem como os outros locais de coleta (p = 0,0031). As coletas realizadas por suabes foram as que apresentaram menor positividade para a presença de esperma, demonstrando que muito embora seja o método de coleta mais utilizado, ele não seria o mais indicado para a recuperação do material biológico. Todavia, embora tenha sido possível avaliar um número maior de amostras positivas quando havia laudo conjunto de vestes e suabes, estes não apresentaram diferença estatisticamente significante quando comparado às amostras de suabes sozinhas (p = 0,0528). Diante das análises, podemos concluir que o suabe sozinho não apresenta uma boa perspectiva de recuperação dos fluidos seminais. Desta forma, recomenda-se que nos casos de crimes sexuais, as vítimas, sempre que possível, devem se submeter ao exame de corpo de delito, apresentando as vestes que utilizava no momento do fato.Palavras-Chave: PSA; Teste imunocromatográfico; Material biológico. AbstractThe aim of this research was to make a comparative analysis of reports from the Institute of Laboratory Forensic Analysis -IALF, belonging to the General Coordination of Expertise of Mato Grosso do Sul in Campo Grande city, in order to observe the presence of seminal fluid found in clothing and in collections performed by swabs in rape victims. We analyzed 895 reports from the period from 2012 to 2015. The statistical tests were performed using the Graphpad Prism 6.0 program and the results expressed by the difference between the means. The percentage of positive samples obtained from the examinations carried out only on the garments was statistically significant in relation to the tests performed by swabs (p = 0.0063), as well as the other collection sites (p = 0.0031). The collections made by swabs were the ones that showed the least positivity for the presence of sperm, demonstrating that althou gh it is the most used collection method, it would not be the most indicated for the recovery of sperm material. However, although it was possible to evaluate a greater number of positive samples when there was a set of clothes and swabs, they did not present a statistically significant difference when compared to only swab samples (p = 0.0528).
