Sea turtles have been proposed as health indicators of marine habitats and carriers of antibiotic-resistant bacterial strains, for their longevity and migratory lifestyle. Up to now, a few studies evaluated the antibacterial resistant flora of Mediterranean loggerhead sea turtles (Caretta caretta) and most of them were carried out on stranded or recovered animals. In this study, the isolation and the antibiotic resistance profile of 90 Gram negative bacteria from cloacal swabs of 33 Mediterranean wild captured loggerhead sea turtles are described. Among sea turtles found in their foraging sites, 23 were in good health and 10 needed recovery for different health problems (hereafter named weak). Isolated cloacal bacteria belonged mainly to Enterobacteriaceae (59%), Shewanellaceae (31%) and Vibrionaceae families (5%). Although slight differences in the bacterial composition, healthy and weak sea turtles shared antibiotic-resistant strains. In total, 74 strains were endowed with one or multi resistance (up to five different drugs) phenotypes, mainly towards ampicillin (~70%) or sulfamethoxazole/trimethoprim (more than 30%). Hence, our results confirmed the presence of antibiotic-resistant strains also in healthy marine animals and the role of the loggerhead sea turtles in spreading antibiotic-resistant bacteria.

A protocol that utilises data (trueness/recovery, precision and robustness) from validation tests to calculate measurement uncertainty was described and applied to a sector field inductively coupled plasma mass spectrometry (SF-ICP-MS)-based method for the determination of Be, Cd, Hg, Ir, Pb, Pd, Pt, Rh, Sb, U, Tl and W in human serum. The method was validated according to criteria issued by international bodies such as AOAC, Eurachem and ISO and the uncertainty in the analytical measurements was estimated following the Eurachem/Citac guide. The methodology was based on dilution of human serum with water and analysis by serum-matched standard calibration. The method quantification limits ranged 0.02 microg/L (Tl, Ir) to 0.26 microg/L (Hg). The coefficients of regression were greater than 0.9991 over a range of two orders of magnitude of concentration. The mean trueness was 101% and the mean recovery on three levels of fortification (1-, 1.5-, and 2-times the baseline serum level) ranged between 93.3% and 106%. The maximum relative standard deviation values for repeatability and within-laboratory reproducibility were 12.8% and 13.5%. The method was robust to slight variations of some critical factors relevant to the sample preparation and SF-ICP-MS instrumentation. The relative expanded uncertainty over three levels of concentration ranged from 11.6% (Hg) to 27.6% (Pt), and the uncertainty on the within-laboratory reproducibility, which included factors such as time, analyst and calibration, represented the main contribution to the overall uncertainty.

1. Annual and seasonal encounter rates, life stages and the main threats to loggerhead sea turtles (Caretta caretta) in the Aeolian Archipelago (southern Italy) were studied. 2. Dedicated boat surveys resulted in 258 surface observations and 138 captures of healthy (n = 309), ailing (n = 66) and dead (n = 21) turtles from 2009 to 2013.3. Loggerheads were encountered at the sea water surface while resting (87%) or feeding on pelagic prey (13%).4. The loggerhead encounter rate (observations per km surveyed) was higher during the autumn, suggesting the presence of potential foraging/overwintering habitats in the area.5. The mean (± SD) curved carapace length (CCL) was 48.8 ± 10.7 cm, with 65% of the individuals ranging from 40-70 cm in size. Smaller turtles were encountered more frequently during the spring months, probably as a result of the abundant pelagic prey within the coastal area. During the colder season, larger turtles were more common in the afternoon than in the morning, suggesting that the time needed for rewarming might increase with turtle size.6. Ingestion of anthropogenic debris was reported in 48.5% of the rescued turtles. Individual mortality was mainly related to longline fishing (70.6%), with debris entanglements/ingestion frequently associated with these records. Longline bycatch and boat collisions were higher in summer, whereas debris ingestion was highest in spring. Different threats might affect particular life stages because the longline bycatch was more frequent for larger turtles, whereas boat collisions were more frequent with smaller individuals. 7. Migratory patterns, habitat characteristics and seasonal changes in sea temperature and currents might influence the seasonal occurrence of loggerhead turtles in this area.8. These results increase the current ecological knowledge of the factors driving loggerhead turtle life and are important for implementing management plans for its conservation in the Mediterranean Sea.
