Paracoccidioidomycosis (PCM) is an endemic disease of humans from Latin America that is caused by Paracoccidioides brasiliensis and P. lutzii, with most cases of PCM in domestic animals being associated with P. brasiliensis. This study presents the clinical, cytological, mycological, serological, and molecular findings associated with P. brasiliensis in a dog from Southern Brazil. Fine needle biopsies were collected from the skin and several lymph nodes of a 5-year-old female Labrador dog that had enlargement of most superficial lymph nodes. Cytology of the skin and lymph nodes revealed pyogranulomatous dermatitis and lymphadenitis associated with fine-necked, budding fungal structures consistent with the Paracoccidioides genus of organisms; mycological culture derived from the lymph node aspirate demonstrated similar budding structures. Serological assays using exoantigens obtained from the fungal culture demonstrated that the fungal organisms derived from the lymph node were antigenically similar to P. brasiliensis by immunodiffusion and Western blot. A PCR assay, using the fungal culture as input, amplified a partial segment of the internal transcribed spacer 1 and 2 regions of P. brasiliensis; direct sequencing and phylogenetic analyses confirmed the PCR product as P. brasiliensis. The combined cytological, mycological, serological, and molecular findings confirmed a diagnosis of fungal dermatitis and lymphadenitis due to P. brasiliensis in this dog. This case represents the third description of clinical PCM in dogs and the first confirmation of mycotic dermatitis associated with P. brasiliensis in this species. The participation of dogs in the possible dissemination of PCM is reviewed, and it is proposed that dogs are probable accidental hosts in the epidemiological cycle associated with P. brasiliensis.
Renicolids are parasites that inhabit the renal tubules and ureters of molluscivorous and piscivorous birds. Puffinus puffinus is a migratory seabird that was identified as the definitive host of Renicola spp. Studies focusing on the renicolid species and the resulting renal lesions are valuable for their association with causes of stranding in seabirds. The aim of this study was to identify the renicolid trematodes and evaluate the histological findings in two P. puffinus stranded on the coast of Paraná state, Brazil. The parasites were evaluated by histologic, ultrastructural and molecular assays, while tissue changes were analyzed by histologic methods. The morphological and morphometrical characteristics of the parasites, along with polymerase chain reaction and sequencing assays (ribosomal and mitochondrial regions), identified the species as Renicola sloanei. The results also suggest that this helminth can be the adult form of Cercaria pythionike. The dilation of collecting ducts was the main histological finding in the kidneys. In conclusion, R. sloanei was identified, and for the first time, P. puffinus was described as a host of this digenean inducing mild renal changes.
Background: Renal cystic diseases (RCD) are characterized by cystic structures on renal parenchyma associated with obstructive lesions, membranous disruptions, and/or growth disturbances. The polycystic kidney disease (PKD) shows specific pathological characteristics, related to mutations on PKD1 and/or PKD2 chromosome locus on humans. In Persian cats and bull terriers the condition is like the human “adult-onset” PKD, while in Perendale sheep the “childhood”-like is described. In cetaceans, RCD are reported, however the characterization of PKD is scarcely described. This report aims to describe two cases of PKD and one of RCD in stranded Steno bredanensis, and to discuss the disease associated factors.Cases: Four rough-toothed-dolphins were found stranded in the Paraná coast, southern Brazil between 2016 to 2018, through the Santos Basin Beach Monitoring Project (PMP-BS), one of the systematic monitoring programs required by Brazilian Institute of Environment and Renewable Natural Resources (IBAMA) for the environmental licensing process of oil production and transport by Petrobras in the presalt province. In three animals histological sampling was performed. One of them (animal 3) was founded alive, presenting altered buoyancy with lateralization to the right and signs of pneumonia. The blood analysis showed anemia and leukocytosis. The animal showed poor clinical prognosis, and even with supportive treatment, come to death four days after the rescue. Routine autopsies were performed on all animals. Animals 1 and 2 presented macroscopically enlarged kidneys containing disseminated cystic structures in the parenchyma. On microscopic examination, the cortical region showed diffuse cystic structures delimited by variable thickness of fibrous tissue, usually compressing adjacent glomeruli, without concomitant inflammatory process. In these cases, the death was associated with the end stage renal disease. Animal 3 showed grossly few cystic structures, well delimited and replacing some reniculi. Tracheitis, granulomatous pneumonia, esophagitis, gastritis, enteritis and papilloma on penis and palate were observed. Microscopically, the cysts were lined by a single layer of columnar to cuboidal epithelial cells surrounded by extensive fibrotic tissue. Multifocal tubular necrosis was also noticed. Multifocal moderate nonsuppurative encephalitis with parasitic eggs and bacterial granulomatous hemorrhagic pneumonia were observed. In this case, the death was associated with the lesions in the nervous system. Discussion: Data concerning polycystic kidney disease on cetaceans and wild animals is limited, and no primary genetic pathway was associated. In the present study, the gross and histological aspects observed on two animals (1 and 2) are similar to the characteristics found in the human adult form of PKD, while the characteristics observed on animal 3 are consistent with usual cystic disease. In addition, the animals are aged like humans where the end stage renal disease occurs in patients around 70 years old. The kidney histological aspects observed in all animals are similar, however, animal 3 showed no renomegaly, a characteristic of PKD. Considering the genetic pathway involved in humans and some animal’s breeds, investigation on gene mutations in S. bredanensis could help to define if this is also a genetic disorder and increase the knowledge about PKD. Mycobacteriosis is a well-known disease that has been reported in a variety of reptiles species. However, the occurrence of mycobacteriosis in sea turtles has been rarely documented, and most reports are from captive-reared animals. A juvenile green turtle (Chelonia mydas) was found stranded in March 2020, in Guanabara Bay, one of the most urbanized and polluted coastal areas in Rio de Janeiro, Southeastern Brazil. The turtle was in fair body condition and exhibited multiple cutaneous fibropapillomas, covering almost 60% of its body. Despite continued medical therapy, the animal died 60 days after initial supportive care. Necropsy revealed granulomatous pneumonia, hydropericardium, small granulomas in the myocardium, liver, spleen, thyroid and brain and caseous exudate in the meninges of the brain. The histopathological analysis showed disseminated granulomatous inflammation in various organs examined during necropsy. Intrahistiocytic acid-fast bacteria were observed in the liver, heart, lungs, kidneys, spleen, brain and adrenals. The blood culture collected shortly after the animal was admitted tested positive for Mycobacterium gordonae, after an 8-week incubation period; however, the animal had already died by the time the results were obtained. Postmortem examination confirmed systemic mycobacteriosis, identified as M. gordonae in blood, liver and lung cultures and by PCR amplification of DNA extracts from frozen tissue samples. This is the first report of a disseminated infection caused by M. gordonae in a free-ranging sea turtle. Although the source of this Mycobacterium remains unknown, it could potentially be related to anthropogenic activities (e.g., waste dumping or leakage from landfill sites). Increasing surveillance of Mycobacterium in wild sea turtles is of paramount importance to better understand the role of mycobacterial species as causative agents of infections in marine species.
