RESUMO-O conhecimento da diversidade genética de espécies nativas é de grande valia quando se objetiva o melhoramento e a conservação de populações naturais. Neste sentido, o objetivo deste trabalho foi selecionar iniciadores ISSR (inter repetições de sequências simples) para Hancornia speciosa (Apocynaceae), assim como quantificar a variabilidade genética em uma população natural. Foram amostrados 15 indivíduos de uma população localizada em Natal-RN. Amostras de caule foram coletadas para a posterior extração do DNA. Para a seleção, 19 primers ISSR foram testados, dos quais seis foram eficientes, apresentando locos nítidos e em maior número (UBC 808; UBC 810; UBC 826; UBC 827; UBC 841 e UBC 842), totalizando 63 locos. Desses, apenas 30 (47,62%) apresentaram polimorfismo. O valor de PIC (conteúdo de informações polimórficas) para os primers selecionados atingiu a média de 0,37, variando de 0,26 a 0,44. A diversidade genética foi considerada baixa dentro da população, com o número de alelos observados (na =1,48), número de alelos efetivos (ne = 1,32), índice de diversidade de Nei (He = 0,18) e índice de Shannon (I = 0,26). Os padrões de diversidade alélica encontrados indicam a ocorrência de um gargalo populacional recente. A utilização de marcadores ISSR para Hancornia speciosa mostrou-se eficaz para a quantificação da diversidade genética dos indivíduos, servindo como aporte para estratégias e planos que visem à conservação e à manutenção da espécie. GENETIC DIVERSITY AND ISSR INITIATORS SELECTION IN A NATURAL POPULATION OF MANGABA (Hancornia speciosa Gomes) (APOCYNACEAE).ABSTRACT -The knowledge of genetic diversity of native species has great value when it aims at the improvement and conservation of natural populations. In that direction, the objective of this study is to select ISSR primers (inter simple sequence repeat) to Hancornia speciosa (Apocynaceae), as to quantify the genetic variability of a natural population. It was sampled 15 individuals of a population located in Natal/RN. Stem samples were collected for subsequent DNA extraction. For the selection, 19 IRSS primers were tested, six of them were effective, presenting clearer and numerous loci (UBC 808, UBC 810, UBC 826, UBC 827, UBC 841, UBC 842) totaling 63 loci. Of which, only 30 (47.62%) presented polymorphism. The value of PIC (polymorphic information content) for the selected primers averaged 0.37, ranging from 0.26 to 0.44. Genetic diversity was considered low in the population, with the number of observed alleles (na = 1.48), number of effective alleles (ne = 1.32), diversity index of Nei (He = 0.18) and Shannon index (I = 0.26). The allelic diversity patterns found indicates the occurrence of a recent population bottleneck. The use of ISSR markers for Hancornia speciosa is effective as measuring the genetic diversity of individuals, working as a support for strategies and plans that aim the conservation and maintenance of the species.
This work aimed to develop nanoemulsions (NE) containing cholesterol and Amphotericin B (AmB) evaluating the influence of a lipophilic amine (stearylamine; STE) on drug encapsulation efficiency (EE), cytotoxicity on macrophages and in vitro antileishmanial activity. The EE of AmB in NE was nearly 100% regardless of STE concentration. Stability studies showed that AmB-loaded NE with or without STE were stable revealing that AmB content and EE remained constant after 180days. In significant contrast, the EE for AmB in NE without cholesterol drastically decreased showing that this co-surfactant significantly improved the retention of drug in NE. The electronic absorption and circular dichroism (CD) data revealed that the signal characteristic of self-associated free AmB, the most toxic form to the host cells, was virtually absent in the spectra of AmB-loaded NE. In agreement, NE-induced toxicity toward macrophages was significantly lower than that observed for the conventional AmB. STE enhanced both cytotoxicity and the activity against intracellular amastigotes of AmB-loaded NE. However, selectivity index values for AmB-loaded NE were considerably higher than that observed for conventional AmB. AmB-loaded and cholesterol-stabilized NE constitutes an attractive alternative for the treatment of leishmaniasis.
Objectives All-trans retinoic acid (ATRA) is one of the most successful examples of differentiation agents and histone deacetylase inhibitors, such as tributyrin (TB), are known for their antitumor activity and potentiating action of drugs such as ATRA. Nanostructured lipid carriers (NLC) represent a promising alternative to the encapsulation of lipophilic drugs such as ATRA. This study aimed to develop, characterize, and evaluate the cytotoxicity of ATRA-TB-loaded nanostructured lipid carriers (NLC) for cancer treatment. Methods The influence of in situ formation of an ion pairing between ATRA and a lipophilic amine (benethamine; BNT) on the characteristics of NLC (size, zeta potential, encapsulation efficiency) was evaluated. Tributyrin (TB), a butyric acid donor, was used as a component of the lipid matrix. In vitro activity on cell viability and distribution of cell cycle phases were evaluated for MCF-7, MDA-MB-231, HL-60, and Jurkat cell lines. Results The presence of the amine significantly increased the encapsulation efficiency of ATRA in NLC. Inhibition of cell viability by TB-ATRA-loaded NLC was more pronounced than the free drug. Analysis of the distribution of cell cycle phases also showed increased activity for TB-ATRA-loaded NLC, with the clear effect of cell cycle arrest in G0/G1 phase transition. The presence of TB played an important role in the activity of the formulation. Conclusion Taken together, these findings suggest that TB-ATRA-loaded NLC represent a promising alternative to intravenous administration of ATRA in cancer treatment.
The continuing fragmentation of forests has been a threat to the maintenance of genetic resources. Genetic diversity is fundamental to the survival of species in natural environments in the long term, as well as being the basis for genetic improvement. The objective of this study was to evaluate the genetic diversity in natural populations of Hancornia speciosa and to contribute to the development of conservation strategies. We sampled 105 individuals of H. speciosa, distributed in seven populations. The ISSR (Inter-Simple Sequence Repeat) markers provided 70 loci, of which 81% were polymorphic. The mean genetic diversity of Nei (h) was 0.19, and the Shannon index (I) was 0.27. The h and I diversity indices ranged respectively from 0.16 to 0.24 in the PAD (Parque das Dunas) population and from 0.21 to 0.29 in MAC (Macaíba) population. Resulting from a Bayesian analysis, the genotypes were divided into four groups (K = 4). The allelic diversity patterns observed indicated the occurrence of the genetic bottleneck in all populations, according to the stepwise mutation model (SMM). The infinite allele model (IAM) revealed an imbalance between mutation and genetic drift only in the PAD population. Genetic conservation strategies for H. speciosa should cover each genetic group that was differentially structured. We recommend in situ conservation and the creation of germplasm banks, especially with the PAD population which demonstrated the lower genetic diversity and decreased effective population size according to the two mutational models.
Resumo: A qualidade do DNA é de fundamental importância quando se objetiva avaliar a diversidade genética em espécies arbóreas por meio de reação em cadeia da polimerase (PCR). Impurezas presentes no DNA podem influenciar negativamente as sensíveis reações da PCR. O objetivo do presente estudo foi avaliar a qualidade do DNA extraído a partir de folha e caule de Hancornia speciosa. O DNA foi extraído do tecido caulinar e foliar de cinco indivíduos. A quantificação foi realizada por meio de espectrofotometria e eletroforese em gel de agarose. O material genético obtido a partir do caule apresentou pureza semelhante ao extraído da folha, obtendo valores para a razão entre as absorvâncias (A260/A280) de 1,46 para o caule e de 1,42 para a folha, ficando, ambos, um pouco abaixo do valor considerado ótimo que é entre 1,5 e 2,5. O tecido caulinar apresentou um DNA de qualidade semelhante ao da folha, sendo sua utilização viável para estudos moleculares em H. speciosa. Introdução A análise molecular é bastante empregada em estudos de diversidade genética que visam o melhoramento e a conservação de espécies florestais (REDDY et al., 2002) Com isso, a qualidade do DNA extraído é de fundamental importância para o bom funcionamento da PCR. Pesquisas recentes, com enfoque genético, realizadas para a H. speciosa, mostraram que o DNA extraído a partir tecido foliar apresentou boa qualidade, resultando em amplificações via PCR e posterior visualização de padrões de bandas após a eletroforese (COSTA; et al., 2011;MARTINS et al., 2012).
Os consumidores da era da Internet estão cada vez mais informados, ativos, com mais opções de escolhas, e, por isso, mais exigentes e difíceis de satisfazer. Atualmente, as empresas procuram oferecer experiências junto com seus produtos e serviços buscando gerar valor para o cliente. A co-criação de valor acontece quando o consumidor cria produtos e experiências em trabalho conjunto com a empresa ou marca. Essa prática vem sendo tratada na literatura como estratégia da empresa, mas há poucos trabalhos publicados sobre co-criação do ponto de vista dos clientes. Esta dissertação tem o objetivo de entender como se dá o envolvimento com o processo de co-criação a partir da ótica dos consumidores, buscando contribuir para a lacuna de conhecimento sobre o tema. Um quadro teórico conceitual foi montado a partir da literatura sobre co-criação, sobre experiência de marca e sobre atitudes, identificando os conceitos fundamentais para se entender o fenômeno sob análise. Tal quadro foi base para realização de nove entrevistas em profundidade com consumidores que participaram de um caso de co-criação e analisadas por meio da técnica de análise de conteúdo. Os resultados apresentaram 31 categorias relacionadas à co-criação, destacando entre elas 20 sentimentos distintos gerados a partir das experiências vivenciadas pelos consumidores. Foram identificados elementos antecedentes do comportamento co-criador, destacando atitudes positivas para a realização de ações em ambientes presenciais, interação entre empresaconsumidor, consumidor-consumidor e inspirações da empresa para incentiva novas ideias. O grupo influenciador representado por professores e o tempo como fator limitante no controle percebido dos consumidores completam os antecendentes encontrados sobre o comportamento co-criador. A contribuição do trabalho se deve ao fato de capturar experiências e impressões do tema co-crição sob a perspectiva dos próprios consumidores cocriadores.Palavras-chave: Co-criação, comportamento do consumidor, experiência de marca.
