Sự dimer protein đóng vai trong quan trọng trong hầu hết các hoạt động sinh học tế bào như: hoạt động enzyme, thụ thể protein màn, nhân tố truyền tín hiệu nhân tố phiên mã. Do đó, kiểm soát sự dimer của protein giúp kiểm soát được các quá trình sinh học này. Trong nghiên cứu này, phân tử Curcubit[8]uril được sử dụng để kiểm soát sự dimer của protein có chứ đuôi FCG (phenylalanine-cystein-glycine) ở đầu N (N-terminus) của protein. Cặp protein chỉ thị CFP/YFP (cyan fluorescent protein/yellow fluorescent protein) chứa đuôi dung hợp FCG được tạo ra bằng hệ thống intein tự cắt (auto cleavage). Phân tử Curcubit[8]uril cảm ứng sự dimer của protein CFP/YFP có chứa đuôi FCG được đánh giá bằng kỹ thuật trao đổi năng lượng FRET (fluoresecnt resonance energy transfer). Tỷ lệ FRET của bước sóng 525 nm/475 nm tăng mạnh từ 0.5 lên 1.2 khi có sự hiện diện của phân tử curcubit[8]uril, đồng nghĩa với việc phân tử này có khả năng cảm ứng sự dimer của protein CFP và YFP có chứa đuôi FCG. Kiểm soát sự dimer của protein bởi curcubit[8]uril có tiềm năng trong việc kiểm soát các protein chức năng sinh học chứa đuôi dung hợp FCG.
Caspase-9 đóng vai trò quan trọng trong quá trình chết của tế bào. Trong tế bào, caspase-9 tồn tại ở trạng thái đơn phân không hoạt tính, và trở nên có hoạt tính khi ở trạng thái dimer. Do đó, kiểm soát sự dimer của caspase-9 giúp cho kiểm soát được hoạt tính của caspase-9. Trong nghiên cứu này, hoạt tính của caspase-9 được hoạt hóa bởi trình tự DNA chứa hai cấu trúc G-quadruplex (DNA2G4). Caspase-9 được dung hợp với peptide RHAU bằng kỹ tthuật sinh học phân tử tạo nên protein tái tổ hợp RHAU23-caspase-9. RHAU23-caspase-9 được biểu hiện trong vi khuẩn E.coli dưới sự cảm ứng của IPTG và được tinh chế bằng sắc ký ái lực với cột His. Kết quả cho thấy DNA2G4 khả năng cảm ứng sự dimer của RHAU23-caspase-9 thông qua tương tác đặc hiệu giữa RHAU và G4, dẫn đến kích thích hoạt tính của enzyme. Sự hoạt hóa RHAU23-caspase-9 bởi DNA2G4 giúp hoạt tính enzyme tăng hoạt tính lên khoảng 2,0 lần so với hoạt tính của RHAU23-caspase-9. Kiểm soát hoạt tính của caspase-9 có tiềm năng trong việc ức chế sự phát triển của các tế bào ung thư.
G-quadruplex là cấu trúc bậc 2 của DNA hay RNA mà đóng vai trò quan trọng trong các quá trình sinh học như sao chép, phiên mã, dịch mã và kéo dài các telomeres. Do đó, cấu trúc G-quadruplex được xem như một phân tử mục tiêu trong việc thiết kế các thuốc trúng đích ứng dụng trong y sinh. Trong nghiên cứu này, nuclease mới được phát triển có khả năng nhắm mục tiêu G-quadruplex song song và thực hiện chức năng cắt tại vị trí đặc hiệu. Enzyme mới được tạo ra bằng phương pháp kết hợp phân tử peptit RHAU53 có khả năng nhận diện G-quadruplex song song với vùng protein xúc tác cắt của Fok1, tạo nên protein dung hợp RHAU53-Fok1. Protein dung hợp này có thể được biểu hiện trong hệ thống E. coli dưới sự cảm ứng của IPTG và được tinh chế bằng sắc ký ái lực với cột His. Đặc biệt, RHAU53-Fok1 có thể nhận diện và bám đặc hiệu vào cấu trúc G-quadruplex song song và cắt DNA sợi kép bên cạnh cấu trúc đó. Enzyme cắt DNA ở nhiều vị trí cho ra 1 sản phẩm chính và các sản phẩm phụ. Enzyme mới này được xem là công cụ tiềm năng cho việc lập bản đồ vị trí cấu trúc G-quadruplex trong bộ gen.
Alpha-momorcharin (α-MMC) thuộc tuýp I protein bất hoạt ribosome (ribosomeinactivating protein) được tìm thấy trong trái khổ qua. Hiện nay, α-MMC đang được nghiên cứu như một protein chức năng có tiềm năng ứng dụng trong y dược và nông nghiệp. Ngoài khả năng chống lại khối u trong giai đoạn đầu của ung thư của α-MMC trong y dược, những nghiên cứu gần đây cho thấy protein này có khả năng kháng virus, kháng vi khuẩn, kháng nấm trong nông nghiệp cây trồng. Hiểu được cơ chế hoạt động của α-MMC sẽ góp phần quan trọng trong việc tìm ra phương pháp cũng như là sản xuất chế phẩm vi sinh ứng dụng vào lĩnh vực nông nghiệp. Trong nghiên cứu này, chúng tôi thảo luận cơ chế hoạt động của α-MMC và khả năng kháng nấm và virus trong cây trồng.
