Меланома кожи является злокачественной опухолью с ранним началом метастазирования и низкой эффективностью лечения диссеминированных форм. Одним из механизмов метастазирования злокачественных опухолей является диссеминации злокачественных клеток, которая является результатом взаимодействия опухолевых клеток и клеток с различными клетками дистантных органов, формирующих впоследствии микроокружение метастазов. Известно, что одними из регуляторных молекул, способных выделяться в экстраклеточные везикулы и принимающих участие в регуляции перепрограммирования микроокружения опухоли, являются микроРНК -короткие некодирующие РНК. При меланоме кожи одной из наиболее низкоэкспрессируемых микроРНК является молекула микроРНК miR-204-5p. В данном исследовании оценено влияние специфического ингибитора молекулы микроРНК miR-204-5p на интенсивность пролиферации in vivo и подвижность клеток меланомы in vitro, а также на профиль транскриптома легких -одного из дистантных органов метастазирования меланомы. Определено, что ингибирование микроРНК miR-204-5p вызывает увеличение интенсивности пролиферации клеток меланомы В16, но не их способности к миграции. Помимо этого, перепрограммирование транскриптомного профиля в легких связано с изменением экспрессии генов, являющихся компонентами механизмов внутриклеточной сигнализации, опосредующих функционирование фибробластов и иммунокомпетентных клеток в данном органе.
