Введение. Инфекция клеток насекомых вирусом ядерного полиэдроза (ВЯП) приводит к появлению в инфицированных клетках вирусспецифичной ДНК-полимеразы [1, 2]. Этот фермент отличается от клеточных ДНК-полимераз α, β, γ по физико-химическим и энзиматическим свойствам и предположительно кодируется вирусным геномом. Используя специфическое мечение белков вируса после ВЯП-инфекции клеток Spodoptera frugiperda, Ванг и Келли [2] показали, что вирусная ДНКполимераза является полипептидом с молекулярной массой 126 000. Получить в очищенном виде этот фермент и изучить его энзиматические свойства авторам не удалось. Ранее нами было показано, что после ВЯП-инфекции в куколках тутового шелкопряда возрастает активность хозяйской α-полимеразы и появляется вирусспецифичная ВЯП-полимераза. Необычным свойством ВЯП-полимеразы была высокая активность на однонитевой ДНК фага М13 в отсутствие затравки. В настоящей работе мы провели очистку ВЯП-полимеразы из инфицированных куколок тутового шелкопряда, описали матричные свойства фермента и механизм утилизации ВЯП-полимеразой ДНК фага М13. Материалы и методы. Однонитевую ДНК фага М13 и ДНК плазмиды pBR322 получали по описанным методам [3, 4]. Вирус ядерного полиэдроза получали и куколки тутового шелкопряда инфицировали, как описано ранее [5]. Суспензию ВЯП инъецировали в куколки на 3-й день метаморфоза. Инфицированные куколки инкубировали при 25 °С в течение 72 ч и собирали на жидком азоте. Процедура очистки ВЯП-полимеразы включала получение гомогената, супернатанта, фракционирование сульфатом аммония, хроматографию на фосфоцеллюлозе, оксиапатите и ультрацентрифугирование в градиенте глицерина (табл. 1). Экспериментальные детали выделения фермента будут описаны в отдельном сообщении. Препарат ВЯП-полимеразы хранили при-20 °С в буферном растворе А: 10 мМ фосфат калия, рН 7,5, 50 %-ный глицерин, 0,1 мМ ЭДТА, 5 мМ 2-меркаптоэтанол.
