The main goal of our research was to search for SSRs in the Eucalyptus EST FORESTs database (using a software for mining SSR-motifs). With this objective, we created a database for cataloging Eucalyptus EST-derived SSRs, and developed a bioinformatics tool, named Satellyptus, for finding and analyzing microsatellites in the Eucalyptus EST database. The search for microsatellites in the FORESTs database containing 71,115 Eucalyptus EST sequences (52.09 Mb) revealed 20,530 SSRs in 15,621 ESTs. The SSR abundance detected on the Eucalyptus ESTs database (29% or one microsatellite every four sequences) is considered very high for plants. Amongst the categories of SSR motifs, the dimeric (37%) and trimeric ones (33%) predominated. The AG/CT motif was the most frequent (35.15%) followed by the trimeric CCG/CGG (12.81%). From a random sample of 1,217 sequences, 343 microsatellites in 265 SSR-containing sequences were identified. Approximately 48% of these ESTs containing microsatellites were homologous to proteins with known biological function. Most of the microsatellites detected in Eucalyptus ESTs were positioned at either the 5’ or 3’ end. Our next priority involves the design of flanking primers for codominant SSR loci, which could lead to the development of a set of microsatellite-based markers suitable for marker-assisted Eucalyptus breeding programs
RESUMO -O objetivo deste trabalho foi investigar a diversidade, a estrutura genética e o tamanho efetivo, retido em um banco de germoplasma, de duas populações de Myracrodruon urundeuva Fr. All. (aroeira), procedentes de Aramina-SP e Selvíria-MS. As populações foram avaliadas a partir da amostragem de 25 progênies de polinização aberta de cada população. De cada progênie, 17 a 20 indivíduos foram amostrados e genotipados para oito locos microssatélites. Os maiores valores para o número total de alelos (A t ), número médio de alelos por loco (A), número efetivo de alelos (A e ), heterozigosidade observada (H 0 ) e esperada (H e ) foram detectados na população Selvíria (A t = 105, A = 13,13, A e = 3,98, H 0 = 0,669 e H e = 0,749), enquanto Aramina teve A t = 94, A = 13,75, A e = 3,10, H 0 = 0,535 e H e = 0,678. A diferenciação nas frequências alélicas das duas populações, com relação ao pólen cruzado (0,159) e do óvulo (0,235), indica que cerca de 84% e 77%, respectivamente, da diversidade genética está dentro das populações. O coeficiente médio de coancestria foi maior (Selvíria Θ = 0,165, Aramina Θ = 0,169) e o tamanho efetivo médio das progênies (Selvíria N e(v) = 3,04, Aramina N e(v) = 2,69) foi menor do que o esperado em progênies de populações panmíticas (Θ = 0,125, N e(v) = 4). O tamanho efetivo total retido no banco ex situ foi estimado em 67,5 na população Selvíria e 71,1 na população Aramina, valores menores do que o requerido (N e = 150) para a conservação de populações em curto prazo. Entretanto, as duas populações apresentaram alta diversidade genética, o que as qualifica para serem utilizadas em programas de conservação e melhoramento genético da espécie, desde que seja aumentado o tamanho efetivo populacional conservado ex situ.Palavras-chave: Microssatélites, Espécie arbórea tropical e Endogamia. -In experimental areas of the Education and Researches Ilha Solteira and Jaboticabal UNESP/Campus farms were selected and tagged 20 hermaphrodite plants and 20 feminine of cultivar Sunrise Solo, Improved Sunrise Solo cv.72/12 and Baixinho of Santa Amália.The seeds origined of the selected fruits were cropped to be analysed the self-pollination efficiency and frequency of the sex in the progenies. After that, samples of the young leaf of the matrix plants were colected for the extration of the DNA. It was built five library enriched of microsatellite sequencies, using probes (TCA) 10 , (TC) 13 , (GATA) 4 , (CAC) 10 e (TGAG) 8 .It was possible the development of the primers only in the library that has utilized the probe (TCA) 10. This probe allowed the design of 32 primer pairs. From these, 31 presented pattern of unique band in agarose Metaphor and in acrilamide. For primer S36 were observed 2 bands, but with no polymorphism to differentiation in the sexual form at papaya tree culture. However, these primers can be tested, in the futures, in the investigation of the others features in segregated populations of this specie and the related species, germoplasm analysis, cultivars identification, parent evolution and molecular markers for the assisted plant breeding programs.
Pertencente à família Lauraceae, o abacateiro compreende três raças hortícolas: antilhana, guatemalense e mexicana. Os marcadores moleculares são uma ferramenta rápida e eficaz para estudos genômicos, uma vez que detectam o polimorfismo diretamente ao nível do DNA e não sofrem qualquer tipo de influência ambiental. Com base nesse polimorfismo, é possível fazer inferências sobre as relações entre o genótipo e o fenótipo dos indivíduos, o que, em última análise, permite aumentar a eficiência dos programas de melhoramento. Diante o exposto, o objetivo foi investigar a diversidade genética entre sete variedades de abacate a partir de 5 lócus de marcadores moleculares microssatélites (SSR). Nas amostras de abacateiros avaliadas, encontrou-se um total de 18 alelos, com uma média de 3,6 alelos por lócus. O dendrograma gerado a partir de análise de agrupamento UPGMA agrupou, separadamente do resto dos genótipos, a cultivar Geada da raça Antilhana, possivelmente por esta variedade ser uma raça pura, e o restante foi agrupado em dois grandes grupos das raças, a Guatemalense e a Mexicana. Os genótipos das sete variedades de abacate apresentam diversidade genética nos cinco lócus de marcadores moleculares microssatélites (SSR) avaliados, o que indica que são materiais promissores para utilização em futuros programas de melhoramento.
