Advances in human genetics in recent years have largely been driven by next-generation sequencing (NGS); however, the discovery of disease-related gene mutations has been biased toward the exome because the large and very repetitive regions that characterize the noncoding genome remain difficult to reach by that technology. For autosomal-dominant spinocerebellar ataxias (SCAs), 28 genes have been identified, but only five SCAs originate from non-coding mutations. Over half of SCA-affected families, however, remain without a genetic diagnosis. We used genome-wide linkage analysis, NGS, and repeat analysis to identify an (ATTTC) n insertion in a polymorphic ATTTT repeat in DAB1 in chromosomal region 1p32.2 as the cause of autosomal-dominant SCA; this region has been previously linked to SCA37. The non-pathogenic and pathogenic alleles have the configurations [(ATTTT) 7-400 ] and [(ATTTT) 60-79 (ATTTC) 31-75 (ATTTT) ], respectively. (ATTTC) n insertions are present on a distinct haplotype and show an inverse correlation between size and age of onset. In the DAB1-oriented strand, (ATTTC) n is located in 5 0 UTR introns of cerebellar-specific transcripts arising mostly during human fetal brain development from the usage of alternative promoters, but it is maintained in the adult cerebellum. Overexpression of the transfected (ATTTC) 58 insertion, but not (ATTTT) n , leads to abnormal nuclear RNA accumulation. Zebrafish embryos injected with RNA of the (AUUUC) 58 insertion, but not (AUUUU) n , showed lethal developmental malformations. Together, these results establish an unstable repeat insertion in DAB1 as a cause of cerebellar degeneration; on the basis of the genetic and phenotypic evidence, we propose this mutation as the molecular basis for SCA37.

Machado-Joseph disease (MJD) is a late-onset neurodegenerative disorder caused by a polyglutamine (polyQ) expansion in the ataxin-3 protein. We generated two transgenic mouse lineages expressing the expanded human ataxin-3 under the control of the CMV promoter: CMVMJD83 and CMVMJD94, carrying Q83 and Q94 stretches, respectively. Behavioral analysis revealed that the CMVMJD94 transgenic mice developed motor uncoordination, intergenerational instability of the CAG repeat and a tissue-specific increase in the somatic mosaicism of the repeat with aging. Histopathological analysis of MJD mice at early and late stages of the disease revealed neuronal atrophy and astrogliosis in several brain regions; however, we found no signs of microglial activation or neuroinflammatory response prior to the appearance of an overt phenotype. In our model, the appearance of MJD-like symptoms was also not associated with the presence of ataxin-3 cleavage products or intranuclear aggregates. We propose the transgenic CMVMJD94 mice as a useful model to study the early stages in the pathogenesis of MJD and to explore the molecular mechanisms involved in CAG repeat instability.

The gene encoding cytochrome c nitrite reductase (NrfA) from Desulfovibrio desulfuricans ATCC 27774 was sequenced and the crystal structure of the enzyme was determined to 2.3-A resolution. In comparison with homologous structures, it presents structural differences mainly located at the regions surrounding the putative substrate inlet and product outlet, and includes a well defined second calcium site with octahedral geometry, coordinated to propionates of hemes 3 and 4, and caged by a loop non-existent in the previous structures. The highly negative electrostatic potential in the environment around hemes 3 and 4 suggests that the main role of this calcium ion may not be electrostatic but structural, namely in the stabilization of the conformation of the additional loop that cages it and influences the solvent accessibility of heme 4. The NrfA active site is similar to that of peroxidases with a nearby calcium site at the heme distal side nearly in the same location as occurs in the class II and class III peroxidases. This fact suggests that the calcium ion at the distal side of the active site in the NrfA enzymes may have a similar physiological role to that reported for the peroxidases.

The cytochrome c nitrite reductase is isolated from the membranes of the sulfate‐reducing bacterium Desulfovibrio desulfuricans ATCC 27774 as a heterooligomeric complex composed by two subunits (61 kDa and 19 kDa) containing c‐type hemes, encoded by the genes nrfA and nrfH, respectively. The extracted complex has in average a 2NrfA:1NrfH composition. The separation of ccNiR subunits from one another is accomplished by gel filtration chromatography in the presence of SDS. The amino‐acid sequence and biochemical subunits characterization show that NrfA contains five hemes and NrfH four hemes. These considerations enabled the revision of a vast amount of existing spectroscopic data on the NrfHA complex that was not originally well interpreted due to the lack of knowledge on the heme content and the oligomeric enzyme status. Based on EPR and Mössbauer parameters and their correlation to structural information recently obtained from X‐ray crystallography on the NrfA structure [Cunha, C.A., Macieira, S., Dias, J.M., Almeida, M.G., Gonçalves, L.M.L., Costa, C., Lampreia, J., Huber, R., Moura, J.J.G., Moura, I. & Romão, M. (2003) J. Biol. Chem. 278, 17455–17465], we propose the full assignment of midpoint reduction potentials values to the individual hemes. NrfA contains the high‐spin catalytic site (−80 mV) as well as a quite unusual high reduction potential (+150 mV)/low‐spin bis‐His coordinated heme, considered to be the site where electrons enter. In addition, the reassessment of the spectroscopic data allowed the first partial spectroscopic characterization of the NrfH subunit. The four NrfH hemes are all in a low‐spin state (S = 1/2). One of them has a gmax at 3.55, characteristic of bis‐histidinyl iron ligands in a noncoplanar arrangement, and has a positive reduction potential.

Dissimilatory nitrite reduction, carried out by hexaheme proteins, gives ammonia as the final product. Representatives of this enzyme group from 3 bacterial species can also reduce NO to either ammonia or NzO. The redox regulation of the nitrite/nitric oxide activities is discussed in the context of the denitrifying pathway, [25]. Two pairs of magneti~~ly interacting hemes (low-spin/low-spin; low-spinlhighspin) operate in this complex, The high-spin heme binds NO and appears to be the enzyme-active site. Reaction of nitrite with fuIly reduced enzyme reoxidizes the lowspin hemes, but the EPR spectrum reveals persistence of the high-spin heme in the NO-bound form. Since the six-electron reduction of nitrite yields no NO as a free intermediate [25,26], NO appears to exist as an enzymebound and not as a gaseous product during the nitriteto-ammonia transit.Ammonia-generating nitrite reductases have typically been purified from spinach and display a complex active site comprised of a siroheme coupled to a single {4Fe,4S] center fl9]. Nitrite reduction by hexaheme enzymes from strictly and facultatively anaerobic bacteria, such as Desuvovibrio desulfuricans (ATTC 27774) [20], Woiinellu succinogenes [21j, Escherichia co& [223, and V. jkcfzeri [24], also give rise directly to ammonia. In concert, EPR andIn view of such involvement of NO with the hexaheme nitrite reductases, we have studied the capacity of representatives of this group to reduce free NO. Their influence in the pathway, NOz-to Nz is discussed as is the redox regulation of the nitrite/nitric oxide reduction activities in these multi-heme systems. MATERIALS AND METHODS
