The aim of this work was to investigate the role of the antioxidant enzymes in salt tolerance comparing the salt-sensitive (Pérola) and a salt-tolerant (Pitiúba) cultivar of cowpea [Vigna unguiculata (L.) Walp.]. Salt stress (100 mM NaCl for 8 d) reduced the leaf growth rate more in the sensitive cultivar. The salt-induced decrease in the relative water content, Na + accumulation and increase in leaf electrolyte leakage was similar in both cultivars. Salt stress induced a higher increase in the activities of superoxide dismutase (SOD), ascorbate peroxidase (APX) and phenol peroxidase (POX) in the tolerant cultivar than in sensitive one.
ABSTRACT(Activities of antioxidant enzymes and root growth inhibition in cowpea seedlings exposed to diff erent salt levels). Phenol peroxidase (POX) is a dual enzyme that is involved with hydrogen peroxide scavenging and lignin biosynthesis, contributing to growth inhibition by secondary wall thickening. In order to relate growth inhibition to salt-induced oxidative modulation, the activities of superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and POX were evaluated in cowpea roots under salinity. Four-day-old seedlings of the Pitiúba and Pérola cultivars were exposed to 0, 25, 50, 75 and 100 mM NaCl in germination paper under controlled conditions. Aft er two days of treatment, root length was reduced under 100 mM NaCl by 56 and 26% in Pitiúba and Pérola, respectively, which was associated with enhanced electrolyte leakage and cell death in the root apex. NaCl salinity did not trigger lipid peroxidation, indicating that cell death was probably due to membrane damage instead of oxidative stress. Salt stress reduced the activity of SOD, CAT and APX and increased the POX activity, demonstrating that this enzyme plays a role in oxidative protection in cowpea roots exposed to NaCl salinity. In conclusion, salt-induced growth inhibition in cowpea roots could be attributed, at least in part, to a coordinate action involving an increase in POX activity and a drop in CAT and APX activities. A salinidade e o déficit hídrico são os fatores abióticos que têm afetado acentuadamente a produção de espécies vegetais de importância socioeconômica nas regiões áridas e semi-áridas. Portanto, o objetivo do presente trabalho foi de avaliar os efeitos do cloreto de sódio (NaCl) e do polietilenoglicol (PEG 6000), agentes estressores comumente utilizados para simular os estresses salino e hídrico, respectivamente, na germinação de sementes de cártamo (Carthamus tinctorius L.), uma oleaginosa da família Asteraceae. A germinação foi conduzida em sistema de rolo, com delineamento experimental de sete tratamentos osmóticos (controle; "0,450; "0,852; "1,271 MPa), sendo três com concentrações isosmóticas de NaCl (100; 200 e 300mM) e PEG (188,35; 267,75 e 357,24 g/L), e cada tratamento com seis repetições. Durante a germinação, foram avaliados parâmetros como a porcentagem de germinação, o índice de velocidade de germinação (IVG), e três dias após o semeio, foram registrados o índice de crescimento relativo da radícula (ICRR) e a massa fresca dos eixos das plântulas. A porcentagem de germinação e o IVG não sofreram alterações significativas no tratamento osmótico de "0,450 MPa induzido por NaCl quando comparado com o controle, enquanto no mesmo tratamento, induzido pelo PEG, estes dois parâmetros apresentaram uma intensa diminuição e um leve aumento, respectivamente. O ICRR e a massa das plântulas apresentaram diminuições proporcionais à redução do potencial osmótico ao utilizar ambos os compostos. Com base nos resultados alcançados, pôde-se concluir que a germinação de sementes de cártamo é influenciada pelo tipo de estresse aplicado e sua respectiva dose. Adicionalmente, o estresse osmótico induzido por PEG 6000 é mais agressivo que o salino, e potenciais osmóticos mais baixos ("0,851 e "1,271 MPa) são limitantes para a germinação desta espécie.
RESUMO -Estudos de meios de cultura que facilitem a germinação de sementes recalcitrantes são de grande importância para a fruticultura. A mangabeira (Hancornia speciosa Gomez) é uma fruteira nativa da região Nordeste do Brasil. Sua propagação por métodos tradicionais é dificultada pelo fato de suas sementes serem recalcitrantes e a polpa do fruto ter uma ação inibitória sobre a germinação das sementes. Na tentativa de maximizar a percentagem de germinação in vitro desta espécie, foi testada a influência da sacarose (0; 10; 30; 60 e 90 g/L), do ácido giberélico (0; 0.1; 0.3 e 0.5 mg/L) e de diferentes meios de germinação (água destilada, água de coco; MS sólido e MS líquido). Também foi testado o efeito da escarificação (sementes com ou sem tegumento). As sementes obtidas de frutos maduros foram escarificadas ou não, e inoculadas em meios contendo os diferentes tratamentos. A taxa de germinação foi calculada trinta dias após a inoculação das sementes. Sementes sem tegumento obtiveram maior percentagem de germinação em todos os meios de cultura estudados, sendo que a maior percentagem foi obtida no tratamento MS líquido. A adição de sacarose tanto em meio MS sólido quanto em MS líquido não favorece a germinação e pode prejudicar em concentrações iguais ou maiores que 20g/ L. A maior percentagem de sementes germinadas em MS suplementado com GA 3 , tanto em meio líquido como em meio sólido, ocorreu na concentração de 0,1 mg/L. Maiores taxas de germinação in vitro de sementes de mangabeira podem ser obtidas através da retirada do seu tegumento e posterior inoculação em meio MS líquido suplementado com 0,1 mg/L GA 3 . The salt stress effect in root growth and antioxidative response were investigated in two cowpea cultivars which differ in salt tolerance in terms of plant growth and leaf oxidative response. Four-day-old seedlings (establishment stage) were exposed to 100 mM NaCl for two days. The roots of the two cultivars presented distinct response in terms of salt-induced changes in elongation and dry weight. Root dry weight was only decreased in Pérola (sensitive) cultivar while root elongation was mainly hampered in Pitiúba (tolerant). Root relative water content remained unchanged under salinity, but root Na + content achieved toxic levels as revealed by the K + /Na + ratio in both cultivars. Then, root growth inhibition might be due to ionic toxicity rather than by salt-induced water deficit. Although electrolyte leakage markedly increased mainly in the Pérola genotype, lipid peroxidation decreased similarly in both saltstressed cultivars. APX and SOD activities were reduced by salinity in both cultivars reaching similar values despite the decrease in Pitiúba had been higher compared to respective controls. CAT decreased significantly in Pitiúba but did not change in Pérola, while POX increased in both cultivars. The salt-induced decrease in the CAT activity of Pitiúba root is, at principle, incompatible to allow a more effective oxidative protection. Our results support the idea that the activities of SOD, APX, CAT and POX and lipid peroxidation in cowpea seedling roots were not associated with differential salt tolerance as previously characterized in terms of growth rate and oxidative response in plant leaves.Key words: antioxidative enzymes, oxidative stress, root growth inhibition, salt stress, Vigna unguiculata resuMo O objetivo deste trabalho foi avaliar o efeito do estresse salino sob o crescimento radicular e a atividade de enzimas antioxidantes em duas cultivares de feijão caupi contrastantes em termos de crescimento e resposta oxidativa de folhas. Plântulas com quatro dias de idade foram expostas a NaCl 100 mM durante dois dias sob condições controladas. A massa seca das raízes foi afetada somente na cultivar Pérola (sensível) enquanto que o comprimento radicular foi mais reduzido na Pitiúba (resistente). O estresse salino não afetou o conteúdo relativo de água, contudo o conteúdo de Na + nas raízes atingiu níveis tóxicos de acordo com a razão /Na + em ambas as cultivares, indicando que a inibição do crescimento radicular pode ser estar mais relacionada à toxicidade iônica mais do ao estresse osmótico induzido pelo sal. Embora o vazamento de eletrólitos tenha aumentado principalmente na cultivar sensível, a peroxidação de lipídeos decresceu de forma similar em ambas as cultivares. As atividades de APX e SOD foram diminuídas pela salinidade nas duas cultivares, atingindo valores similares, apesar do decréscimo em Pitiúba ter sido maior, em relação aos controles. The effects of Al 2 (SO 4 ) 3 Á18H 2 O on growth of root and apical root cells were studied in seedlings of rice cultivars differing in Al resistance including I Kong Pao and Aiwu (Al-sensitive) and IRAT 112 and IR6023-10-1-1 (Al-resistant). Inhibition of root growth was a typical effect of Al, and the extent of the inhibition depended on both cultivar and Al concentration. Al impaired the activity of the root meristem as indicated by reductions in its size, mitotic activity and the diameter of the meristematic cell nucleoli. Cell size in the elongation zone of the root was also reduced by Al. The reliability of the haematoxylin staining method to classify rice cultivars according to their Al-sensitivity failed to discriminate the Al-resistant IR6023-10-1-1 cultivar from the two sensitive cultivars. The results are discussed in relation to the Al resistance mechanisms operating in rice.
O efeito de diferentes concentrações de ANA e BAP na micropropagação do abacaxizeiro, bem como no cultivo em hidroponia das plântulas obtidas in vitro, foram estudados em brotos de abacaxizeiro da variedade Pérola, inoculados em meio de cultura básico MS, suplementado com os fitorreguladores BAP e ANA em diferentes concentrações. Caracteres morfológicos quantitativos relacionados ao crescimento dos brotos e das plântulas de abacaxizeiro foram avaliados respectivamente durante os cultivos in vitro e em hidroponia e mostraram que, o tratamento T1 (BAP = 1,0 mg L-1e ANA = 0,5 mg L-1) proporcionou a maior taxa média de regeneração de brotos e conseqüentemente uma maior produção de matéria fresca. Entretanto, a altura dos brotos e a formação de suas raízes foram maiores nos tratamentos T2 (BAP = 0,5 mg L-1 e ANA = 0,25 mg L-1 ) e T3 (BAP = 0,25 mg L-1 e ANA = 0,12 mg L-1 ). Após sessenta dias de cultivo em hidroponia, todas as plântulas oriundas do tratamento T1 apresentaram um bom desenvolvimento, expresso pela maioria dos caracteres morfológicos avaliados. O sistema de micropropagação utilizado neste trabalho possibilitou a obtenção de brotos de abacaxizeiro Pérola, em quantidade suficiente e ao mesmo tempo de fácil individualização, seguida da regeneração de plântulas que foram cultivadas em hidroponia.
