Intraspecific resource partitioning and social affiliations both have the potential to structure populations, though it is rarely possible to directly assess the impact of these mechanisms on genetic diversity and population divergence. Here, we address this for killer whales (Orcinus orca), which specialize on prey species and hunting strategy and have long-term social affiliations involving both males and females. We used genetic markers to assess the structure and demographic history of regional populations and test the hypothesis that known foraging specializations and matrifocal sociality contributed significantly to the evolution of population structure. We find genetic structure in sympatry between populations of foraging specialists (ecotypes) and evidence for isolation by distance within an ecotype. Fitting of an isolation with migration model suggested ongoing, low-level migration between regional populations (within and between ecotypes) and small effective sizes for extant local populations. The founding of local populations by matrifocal social groups was indicated by the pattern of fixed mtDNA haplotypes in regional populations. Simulations indicate that this occurred within the last 20,000 years (after the last glacial maximum). Our data indicate a key role for social and foraging behavior in the evolution of genetic structure among conspecific populations of the killer whale.

The mechanisms that determine population structure in highly mobile marine species are poorly understood, but useful towards understanding the evolution of diversity, and essential for effective conservation and management. In this study, we compare putative sperm whale populations located in the Gulf of Mexico, western North Atlantic, Mediterranean Sea and North Sea using mtDNA control region sequence data and 16 polymorphic microsatellite loci. The Gulf of Mexico, western North Atlantic and North Sea populations each possessed similar low levels of haplotype and nucleotide diversity at the mtDNA locus, while the Mediterranean Sea population showed no detectable mtDNA diversity. Mitochondrial DNA results showed significant differentiation between all populations, while microsatellites showed significant differentiation only for comparisons with the Mediterranean Sea, and at a much lower level than seen for mtDNA. Samples from either side of the North Atlantic in coastal waters showed no differentiation for mtDNA, while North Atlantic samples from just outside the Gulf of Mexico (the western North Atlantic sample) were highly differentiated from samples within the Gulf at this locus. Our analyses indicate a previously unknown fidelity of females to coastal basins either side of the North Atlantic, and suggest the movement of males among these populations for breeding.

Mounting evidence that sharks are being over-fished to supply shark fin markets is causing widespread concern about the sustainability of these practices. The basking shark Cetorhinus maximus, whose fins command high market prices, has proven especially sensitive to exploitation. To prevent further population declines, this species is now protected in the territorial waters of several countries, and is listed on Appendix II of the Convention on International Trade in Endangered Species (CITES) requiring monitoring of trade in its products by all parties to CITES. Tracking trade in basking shark products, however, is often hampered by difficulties in identifying shark products to species of origin. Here, we present the development and application of a streamlined genetic forensics assay that does not require DNA sequencing to identify basking shark products. The dual-primer, species-specific polymerase chain reaction strategy provides diagnostic redundancy for robustness in legal venues. It is also effective for identifying basking shark products regardless of geographic origin, an important consideration, given the global distribution of the species and international sourcing of fins to the trade. Application of the assay confirmed the presence of basking shark fins in the Hong Kong and Japan markets, and indicated an apparent relationship between the Chinese fin trader category 'Nuo Wei Tian Jiu' and fins from basking sharks. The assay was also used in a law enforcement investigation to document illegal trade in basking shark fins in the United States where this species is prohibited from harvest and trade. These trade detections suggest that the high market value of basking shark fins is continuing to drive the exploitation, surreptitious and otherwise, of this highly threatened species, underscoring the need for improved trade monitoring. The streamlined assay developed here can assist in monitoring and conservation on a worldwide scale.

The preservation of organic remains and environmental indicators in those soft, damp and colourful deposits that visually indicate ancient excreta is familiar. This note reports a horse parasite, and draws attention to preservation of valuable material in deposits much less promising in nature.
