Bezerros foram imunizados com duas vacinas contra o herpesvírus bovino tipo 1 (BHV-1; vacina A, n=28; B, n=28) ou com uma vacina contendo antígenos do BHV-1 e BHV-5 (n=32) e a atividade neutralizante sérica antiBHV-1 e BHV-5 foi testada após três doses vacinais. Todos os animais (n=88) produziram anticorpos com atividade neutralizante antiBHV-1 (títulos médios geométricos [GMT] de 13,1; 14,8 e 34,3 para as vacinas A, B e C, respectivamente) e 82 animais (93,2%) desenvolveram atividade neutralizante antiBHV-5 (GMTs: 10,6; 11,5 e 29,8, respectivamente). Nos três grupos vacinais, os títulos médios antiBHV-1 não diferiram dos títulos antiBHV-5. No entanto, comparando-se os títulos médios entre as vacinas, os títulos neutralizantes para o BHV-1 e BHV-5 foram superiores nos animais do grupo C (p<0,01 ). Esse grupo também apresentou a maior proporção de reagentes ao BHV-5 (96,9%; contra 85,7% da vacina A e 92,9% da vacina B). Esses resultados demonstram que a vacina C induziu títulos neutralizantes superiores contra os dois vírus e que anticorpos induzidos por antígenos do BHV-1 (vacinas A e B) possuem atividade neutralizante relevante também contra o BHV-5.
A resposta sorológica e proteção fetal conferida por três vacinas inativadas contra o vírus da diarréia viral bovina (BVDV) (vacinas A, B e C) foram avaliadas através de vacinação e posterior desafio de ovelhas prenhes com amostras brasileiras de BVDV-1 e BVDV-2. Níveis baixos a moderados de anticorpos neutralizantes anti-BVDV-1 foram detectados na maioria dos animais (45/47) aos 30 dias após a segunda dose vacinal (títulos médios geométricos [GMT] de 124,7; 74,6 e 26,7 para as vacinas A, B e C, respectivamente). Em contraste, atividade neutralizante anti-BVDV-2 não foi detectada em vários animais (12/47) e foi de magnitude inferior nos três grupos vacinais (GMTs 19,1; 14,1 e 15,1). Os títulos médios de anticorpos reduziram-se significativamente no dia 180, sendo que vários animais já não apresentavam atividade neutralizante detectável frente ao BVDV-1 (grupo B=1/19; C=8/14) e principalmente frente ao BVDV-2 (A=7/14; B=13/19; C=13/14). Nessa data, os títulos médios de anticorpos contra as amostras utilizadas no desafio eram de 91,9; 15,1 e 60,6 (SV-126.8, BVDV-1) e de 10; <10 e 28,3 (SV-260, BVDV-2) nos grupos A, B e C, respectivamente. Nos três grupos vacinais, os níveis de anticorpos neutralizantes contra essas amostras não foram suficientes para prevenir a replicação, disseminação virêmica e transmissão transplacentária dos vírus aos fetos. O vírus foi detectado no sangue e nos fetos de todas as ovelhas vacinadas com as vacinas A (10/10), B (9/9) e C (8/8). Esses resultados demonstram que as vacinas testadas induziram níveis moderados a baixos de anticorpos neutralizantes contra o BVDV-1 e, principalmente contra o BVDV-2 na maioria dos animais; e que esses níveis não foram suficientes para prevenir a infecção fetal frente ao desafio com amostras de BVDV-1 e BVDV-2. Adicionalmente, os resultados confirmam a adequação de ovelhas prenhes para estudos de proteção fetal por vacinas contra o BVDV.
Anticorpos monoclonais (AcMs) produzidos com uma amostra brasileira do herpesvírus bovino tipo 5 (BHV-5) foram utilizados na adaptação e padronização de técnicas rápidas para o diagnóstico de infecções pelo BHV-1 e BHV-5. A detecção de antígenos virais por imunofluorescência em células descamativas (IFCD) foi comparada com o isolamento viral, em secreções nasais de 16 bezerros, sendo 6 inoculados com o BHV-1 e 10 com o BHV-5. De 203 amostras testadas, 182 (89,6%) apresentaram resultados concordantes nos dois testes (143 positivas; 39 negativas). Comparando-se com o isolamento, a IFCD apresentou sensibilidade de 96,6%, especificidade de 70,9%, e precisão de 89,6%. Os AcMs também foram utilizados para detectar antígenos do BHV-5 em impressões frescas do cérebro de bezerros acometidos de enfermidade neurológica e em células de cultivo utilizadas na técnica de soroneutralização (SN), permitindo a obtenção dos resultados em 24 horas. A técnica, denominada de soroneutralização rápida (SNR), apresentou sensibilidade de 97,3%; especificidade de 95,5% e precisão de 96,2% em comparação com a SN tradicional; e sensibilidade de 94,7%, especificidade de 97,3% e precisão de 96,1% em comparação com um ELISA comercial. Esses resultados demonstram que esses AcMs podem ser muito úteis para uso em técnicas rápidas de diagnóstico de infecções suspeitas de BHV-1 ou de BHV-5, sobretudo em situações de surto, quando a tomada de decisões adequadas depende de um diagnóstico rápido e confiável.
