Die aus Tetracyclin, Formaldehyd und einer Reihe von Aminosäuren entstehenden Kondensationsprodukte sind unterschiedlich stabil und löslich. β‐Alanin, Cystein, Histidin, Tryptophan, Tyrosin und Glutaminsäure geben keine analysenreine Verbindungen.
Aliphatische, aromatische und aliphatisch‐aromatische Säureamide werden mit Formaldehyd und α‐Alanin nach Einhorn umgesetzt. Acetamid gibt ein einheitliches Kondensationsprodukt, dagegen wird durch eine Substitution in der Alkylgruppe die Reaktion gestört. Aromatische Säureamide reagieren leicht, auch wenn sie substituiert sind. Die beste Ausbeute erhält man mit Salicylsäureamid.
α‐Alanin, Valin, Norvalin, Phenylalanin und α‐Aminobuttersäure geben leicht die Einhorn‐Reaktion mit Acetamid. Dagegen entstehen mit Glycin, β‐Alanin und γ‐Aminobuttersäure nur schwer trennbare Substanzgemische. Dieses Verhalten hängt wahrscheinlich mit dem unterschiedlichen Bindungsvermögen der Aminosäuren gegenüber Formaldehyd zusammen. — Die Einhorn‐Reaktion mit Lysin und Arginin verläuft sowohl in alkalischem wie in saurem Milieu nicht einheitlich, wobei das Arginin stabilere Verbindungen bildet.
