With the wide availability of massively parallel sequencing technologies, genetic mapping has become the rate limiting step in mammalian forward genetics. Here we introduce a method for real-time identification of N-ethyl-N-nitrosourea-induced mutations that cause phenotypes in mice. All mutations are identified by whole exome G1 progenitor sequencing and their zygosity is established in G2/G3 mice before phenotypic assessment. Quantitative and qualitative traits, including lethal effects, in single or multiple combined pedigrees are then analyzed with Linkage Analyzer, a software program that detects significant linkage between individual mutations and aberrant phenotypic scores and presents processed data as Manhattan plots. As multiple alleles of genes are acquired through mutagenesis, pooled "superpedigrees" are created to analyze the effects. Our method is distinguished from conventional forward genetic methods because it permits (1) unbiased declaration of mappable phenotypes, including those that are incompletely penetrant (2), automated identification of causative mutations concurrent with phenotypic screening, without the need to outcross mutant mice to another strain and backcross them, and (3) exclusion of genes not involved in phenotypes of interest. We validated our approach and Linkage Analyzer for the identification of 47 mutations in 45 previously known genes causative for adaptive immune phenotypes; our analysis also implicated 474 genes not previously associated with immune function. Este artigo procura reinterpretar o rito andino de marcação do gado denominado Santiago, que a comunidade camponesa de Auray (Andes Centrais do Peru) celebra todo dia 25 de julho de cada ano. O reconhecimento da complexidade dessas crenças e práticas religiosas é muito importante e decisivo para saber a percepção e o entendimento do pensamento do povo dos Andes Centrais com relação ao aumento, à diminuição e à proteção do gado e, sobretudo, com relação às condições sociais e naturais em que essas práticas se realizam hoje.
This article discusses the dialog between filmic anthropology's procedures and methods and anthropological practice, with a focus on ritual, which is captured by the moving image in a more direct and fluid way. For this, the ritual of reproduction and preservation of cattle in Santiago, a peasant village in the Peruvian Andes is used as an empirical base. As an anthropologist-filmmaker I will try to make explicit the relationship between the observed process filmed and the informant, combining two important epistemological grids: Claudine de France's deferred observation, and Clifford Geertz's interpretation from the native's point of view. Finally, based on this experience, I will make some observations about the use of methodological approximations from filmic anthropology in anthropological practice, between film and anthropology.
Já passou mais de setenta e um anos desde que Ruth Benedict nos apresentou sua obra “O Crisântemo e a Espada” (1946), trabalho que, de meu ponto de vista, contempla o nascimento da antropologia do cinema. São sessenta e quatro anos desde que Margaret Mead nos apresentou seu “Estudo de Culturas à Distância” (1953) pela primeira vez; a partir de então tem se realizado muitas pesquisas tomando o cinema como potenciais fontes interpretação nas ciências sociais. Sua integridade e constituição que um dia foram colocadas em questão, hoje ressurgem, sobretudo no ambiente acadêmico, com uma série de propostas que permitem esclarecer e sedimentar teórica e metodologicamente seu horizonte. É por isso, que baseados em nossas experiências e etnografias fílmicas, gostaríamos fazer algumas considerações epistemológicas e metodológicas da utilização das narrativas cinematográficas na pesquisa. Este artigo não entrará em debates e discussões dos novos estudos contemporâneos sobre cultura, não tenho a pretensão de um balanço exaustivo, quanto muito apontarei algumas considerações.
O documentário A Batalha de Adwa, realizado pelo cineasta Haile Gerima, permite o acesso a um fato da história etíope até então pouco conhecido pelo público brasileiro. Através de seu documentário, o cineasta etíope faz uma releitura desse fato histórico, propondo-se a retraçar seus aspectos marcantes e a entendê-los por meio de análise de fontes inovadoras, como entrevistas, canções e material iconográfico. Como qualquer obra fílmica, essa é repleta de representações e subjetividades que constituem os elementos necessários para a reconstituição de uma voz etíope sobre a história da batalha.
Este artigo tem como objetivo discutir os aspectos estilísticos e subjetivos dos conceitos de autoetnografia e documentário dispositivo em primeira pessoa, construídos pelo encontro entre Patrícia Ferreiro Pará Yxapy e Sophia Ferreira Pinheiro, cujo resultado é o documentário experimental Teko Haxy-Ser Imperfeita (2016). O filme é particularmente instigante para saber, como essas vozes estão construídas, do que falam essas vozes e, certamente, quais são as questões estéticas e subjetivas advindas desse documentário que deriva do ato de filmar a outra e a si mesmas. Para elaborar essa análise fílmica, utilizamos os princípios metodológicos da antropologia do cinema, notadamente, o conceito de etnografia fílmica e, os princípios metodológicos de mise en scène e auto-mise en scène oriundas da antropologia fílmica.
