Tropical gar, Atractosteus tropicus, is a carnivorous fish species from Southern México with high value and acceptance in local markets. Therefore, the present study aims to spare proteins in diets for larviculture of this species. An experiment was performed using three different experimental diets with increasing carbohydrate levels based on potato starch (S) and decreasing protein content (P) and total energy: 16% S (16% S-44% P), 22% S (22% S-40% P) and 28% S (28% S-36% P). Tropical gar larvae (five days post-hatching, 2.1 mm average notochordal length) were assessed for growth, survival, degree of cannibalism, and digestive enzyme activities for 30 days. Highest growth and survival (24%), as well as the lowest cannibalism (33%), was seen in larvae fed the 28% S diet, and these larvae also had the highest lipase, amylase and glucosidase activities (0.28, 0.56 and 0.11 units, respectively). Protease activity (alkaline protease, trypsin, chymotrypsin, and leucine aminopeptidase) was higher in the larvae fed the 22% S diet than in those given the 16% S and 28% S diets. In conclusion, dietary inclusion of potato starch at 28% S enhanced growth, survival and some digestive enzyme activities, and decreased cannibalism in the larval gar. Potato starch could replace dietary protein as a major source of energy for A. tropicus larvae, thereby reducing the cost of diets.
This study undertakes the characterization of digestive proteases in the juvenile green cichlid, Cichlasoma beani. The results obtained showed a higher activity of alkaline proteases (0.14 ± 0.01 U mg protein −1 ) compared to acid proteases (0.07 ± 0.01 U mg protein −1 ) in this species. The optimum temperature of the alkaline proteases was 65 • C and these enzymes were more thermostable to temperature changes than the acid proteases, characterized by an optimal temperature of 55 • C. The pH optimum was 2 for acid proteases, and 11 for alkaline proteases, which were also more stable to changes in pH between 8 and 10. The use of specific inhibitors showed an acid protease inhibition of 88% with pepstatin A as inhibitor. In the zymogram SDS-PAGE analysis of alkaline proteases, five active fractions were revealed, indicating the presence of serine proteases. These results confirm that both alkaline and acid proteases are involved in the digestion of C. beani, and suggest that this species is omnivorous with carnivorous tendencies. The present study contributes to our knowledge about the digestive physiology of C. beani, and can be applied towards improved understanding of the kinds of protein sources that could be used in the development of inerts diets.

Short-tailed pipe fish (Microphis brachyurus) is a freshwater organism with high economic potential for the aquarium hobby, so it is necessary to implement methods to promote its culture through studies of digestive physiology. General activities of acid and alkaline proteases were evaluated, as well as the effect of pH, temperature and inhibitors. The optimal pH of stomach proteases was 2, while the optimal pH of intestinal proteases was 10. Optimal temperature for the acidic proteases was 35 ºC, while for alkaline proteases it was 45 ºC. Thermal stability showed high resistance at 35 ºC for both acid and alkaline proteases (above 100% residual activity). Acid proteases are resistant at pH 2 (50% of residual activity), meanwhile alkaline proteases were highly resistant at pH 10 (90% of residual activity). Acid proteases were inhibited by 80% with pepstatin A and alkaline proteases were inhibited with TLCK and TPCK for trypsin (75%) and chymotrypsin (80%), respectively. Finally, metallo-proteases were 75% partially inhibited some serine proteases by 75% with EDTA. In conclusion, M. brachyurus has a good digestive capacity, since they can degrade a wide variety of proteins due to their greater proteolytic activity.
Wolf cichlid, Parachromis dovii, is a species with a high potential for aquaculture in Central America; however, the knowledge of the digestive physiology in larvae period is limited. For these reason, this study evaluated the changes on digestive enzymes (alkaline and acid proteases, trypsin, chymotrypsin, aminopeptidase, carboxypeptidase, lipases, amylases, and phosphatases) during early ontogeny by biochemical analysis. All digestive enzymes were detected at first feeding (6 days after hatching, DAH, 9.49 mm,. Afterwards all enzymes reached two main peaks in activity at 14 or 22 DAH (15.10 mm, 364 DD and 20.83 mm, 550 DD, respectively). Later, there was a gradual decrease in activity for trypsin and acid and alkaline phosphatases until reach the lowest values at 41 DAH. In the case of acid proteases, chymotrypsin, aminopeptidase, carboxypeptidase, lipase and amylase, all activities reached their maximum values at the end of the larval period, except for alkaline proteases, which showed the maximum value at 14 DAH (15.10 mm, 364 DD). Parachromis dovii larvae have an early capability to hydrolyze exogenous food, agreeing with other carnivorous neotropical cichlid species, for this reason we proposed that the weaning process could begin at 14 DAH.El guapote lagunero (Parachromis dovii) es una especie con un alto potencial para la acuicultura en la región de América Central; sin embargo, existe un conocimiento limitado sobre la capacidad digestiva en el periodo larval. Por este motivo, este estudio evaluó los cambios de las enzimas digestivas (proteasas alcalinas y ácidas, tripsina, quimotripsina, aminopeptidasa, carboxipeptidasa, lipasas, amilasas y fosfatasas) durante la ontogenia temprana mediante análisis bioquímico. Todas las enzimas digestivas analizadas se detectaron en la primera alimentación (6 días después de la eclosión, DAH, 9.49 mm, 168 día-grados DD). Después, todas las enzimas alcanzaron dos picos máximos a los 14 o 22 DAH (15.10 mm, 364 DD and 20.83 mm, 550 DD, respectivamente). Después las actividades tripsina, fosfatasas ácidas y alcalina disminuyeron a sus valores más bajos a los 41 DAH. En el caso de las proteasas ácidas y alcalinas, quimotripsina, aminopeptidasa, carboxipeptidasa, lipasa y amilasa, los niveles de actividad aumentaron y alcanzaron su máximo valor al final del período larvario, excepto las proteasas alcalinas, que mostraron su máximo valor a los 14 DAH (15.10 mm, 364 DD). Las larvas de P. dovii tienen una capacidad temprana para hidrolizar alimentos exógenos, lo que concuerda con otras especies de cíclidos neotropicales carnívoros, por lo que proponemos que el proceso de destete inicie a los 14 DAH. In the Southeast of Mexico, there are many native cichlids with commercial interest such as redhead cichlid (Vieja melanurus) and twoband cichlid (V. bifasciata), which have a great local demand and excellent meat quality. However, it is necessary to implement their culture based on nutrition studies and digestive biochemistry. This study’s objective was to characterize these two cichlids’ digestive proteases (pH, temperature, and inhibitors) through biochemistry techniques. Results showed that V. melanurus and V. bifasciata have a digestive capacity analogous to other omnivore fishes, where the optimal pH values of stomach proteases (4 and 2, respectively) and intestinal proteases (6 and 12, respectively), the optimal temperature of acid (35°C and 55°C, respectively) and alkaline proteases (45°C and 55°C, respectively) are quite similar. Both species presented high thermal and pH stabilities. Inhibition showed that V. melanurus is more sensitive to specific inhibitors for alkaline proteases than V. bifasciata. In conclusion, V. bisfasciata and V. melanurus have different digestive protease patterns. Both species can hydrolyze different protein ingredients to formulate a specific diet. Nevertheless, V. bifasciata is more resistant to the presence of inhibitors, which allow it to include vegetable proteins in its diet.
A 70-day experimental trial was performed to evaluate the contribution of biofloc to the nutrition of Macrobrachium rosenbergii 15 days old postlarvae (PL) (average 82.00 ± 19.71 mg) fed different crude protein (P) and lipids (L) levels in zero-water exchange culture tanks. Six biofloc treatments (BFT) as experimental diets with 15, 20, or 25% P levels and 5 or 8% L levels were managed: BFT-15/5; BFT-15/8; BFT-20/5; BFT-20/8; BFT-25/5; BFT-25/8, and a clear-water control without biofloc fed with 25% P and 8% L (CW-25/8). The experiment was done in triplicate in 21-60 L plastic tanks containing 10 prawns tank-1. At the end of the experiment, survival of prawns was above 66%, with no significant differences among experimental groups (P > 0.05). The best growth performance was observed in the experimental prawns maintained in BFT-20/5, BFT-20/8, BF-25/5, and BFT-25/8 compared to the control (P < 0.001). The feed conversion rate of the prawns in BFT-20/5 was significantly lower than that in control (P < 0.001); the best P efficiency ratio and productive P value were observed in all BFT compared to control prawns (P < 0.001). Results showed that it is feasible to use low concentrations of P (20%) and L (5%) in diets to grow giant freshwater prawn PL in a biofloc system. Results also demonstrated that biofloc contributes to the P and L requirements of cultured freshwater prawn PL, as indicated by improved feed utilization, P retention, and growth performance.
