The deterioration of environmental quality in the Sinos River basin is directly associated with the impacts of intense industrialization and urbanization. An integrated environmental assessment (IEA) was conducted in July and September of 2012, in areas along the sources of the EstânciaVelha/Portão, Pampa and Schmidt streams using physical, chemical and biological methods. The water in the three sampling sites was not proper for human consumption, presented a low toxic contamination index (TCI) and mesotrophic characteristics. One site was included in Class 4, and two, in Class 3, according to current legislation. The rapid assessment protocol (RAP) indicated a natural environmental condition for habitat diversity and environmental impact in the three sites. The Tradescantia pallida (Rose) D.R. Hunt var. purpurea Boom biomarker showed water genotoxicity in two of the sites. The integrated diagnosis of water quality in these streams is fundamentally important to ensure the sustainable management of water resources and their multiple uses, as well to estimate their contribution to pollution in this river basin.Keywords: genotoxicity, water quality indices, source, rapid assessment protocol. Avaliação Ambiental Integrada de arroios da bacia do Rio dos Sinos no Estado do Rio Grande do Sul, Brasil ResumoA deterioração da qualidade ambiental da bacia do Rio dos Sinos está diretamente associada a impactos oriundos da intensa industrialização e urbanização. A avaliação ambiental integrada (AAI) foi realizada nos meses de julho e setembro de 2012, em áreas junto às nascentes dos arroios Estância Velha/Portão, Pampa e Schmidt por meio de metodologias que utilizam parâmetros físicos, químicos e biológicos. Os três sítios amostrais apresentaram águas impróprias para o consumo humano, baixo índice de contaminação por tóxicos (ICT) e características mesotróficas. Um sítio foi enquadrado na Classe 4 e dois na Classe 3, segundo a legislação vigente. Os resultados da aplicação do protocolo de avaliação rápida (PAR) indicaram uma situação ambiental natural, quanto à diversidade de habitats e aos impactos ambientais, nos três sítios amostrados. O biondicador Tradescantia pallida (Rose) D.R. Hunt var. purpurea Boom evidenciou genotoxicidade da água em dois sítios amostrados. O diagnóstico integrado da qualidade das águas destes arroios é de fundamental importância para assegurar o gerenciamento sustentado dos recursos hídricos e seus múltiplos usos, além de estimar sua contribuição à poluição desta bacia hidrográfica.Palavras-chave: genotoxicidade, índices de qualidade da água, nascente, protocolo de avaliação rápida.
The present study evaluated the genotoxic effects of the atmospheric air on Tradescantia pallida var. purpurea in urban areas with different intensities of vehicular traffic and in riparian forest fragments in the Sinos River Basin (Rio Grande do Sul, Brazil), considering the influence of climatic conditions prevailing in these environments. Bimonthly, from May 2012 to March 2013, cuttings with flower buds were exposed for 8 h in urban and riparian forest environments in the municipalities of Caraá, Taquara and Campo Bom in the upper, middle and lower sections, respectively, of the Sinos River Basin. Simultaneously, negative controls were made and climatic data were recorded. Micronuclei (MCN) frequencies were determined in young tetrads of pollen mother cells and expressed as MCN/100 tetrads. Significantly higher MCN frequencies were observed in buds exposed in urban and riparian forest environments in Taquara (up to 7.23 and 4.80, respectively) and Campo Bom (up to 4.90 and 4.23, respectively) than in buds exposed in Caraá (up to 2.90 and 2.50, respectively), in the majority of samplings, and in relation to the negative control (up to 1.93) in all months. Over the course of the period monitored, there were significant variations in MCN frequencies at all sampling points, with the exception of the urban environment in Caraá. For the urban environments, relation between the MCN frequency, vehicular traffic and mean temperature was observed. For the riparian forest fragments, there was no association between MCN frequency and climatic factors. Tradescantia pallida var. purpurea can be considered a useful tool to point out areas with increased atmospheric pollution, since the exposure of plants under severe climatic conditions is avoided to minimize their negative influence on the formation of micronuclei.Keywords: abiotic factors, vehicular emissions, bioindicator, environmental quality, micronucleus. Tradescantia pallida var. purpurea cuttings with flower buds are utilized in bioassays to diagnose genotoxic effects of water. The literature describes different substances used to adapt and recover the cuttings before and after exposure to water samples and also describes the effects of different exposure times. This study evaluated and compared the micronuclei (MCN) frequencies in T. pallida when cuttings with flower buds were submitted to different methodological conditions. The bioassay was then applied bimonthly during seven months to assess the genotoxic potential of a site located on the Sinos River in Campo Bom, Rio Grande do Sul, Brazil. Micronuclei frequencies in buds of cuttings adapted and recovered in distilled water and in Hoagland solution were 3.0 and 2.9, respectively, for cuttings exposed to river water, and 1.19 and 1.23 in controls. No significant differences among MCN frequencies were observed when cuttings were exposed for 8, 24 or 32 hours to river water (from 3.07 to 4.73) and in controls (from 1.13 to 2.00) in all samplings during a year. Adaptation and recovery of cuttings in distilled water or Hoagland solution and exposure for different times did not influence the response of T. pallida, indicating that all the conditions tested are viable for biomonitoring of water genotoxicity. Water samples from the Sinos River presented genotoxicity during the period monitored, evidenced by the MCN frequencies recorded which were significantly higher than the frequencies of the controls.
RESUMOA qualidade química e a genotoxicidade da água do arroio Luiz Rau em Novo Hamburgo (Rio Grande do Sul, Brasil) e a relação entre os danos genéticos observados em Tradescantia pallida var. purpurea e parâmetros químicos foram analisados. Amostras de água foram coletadas bimestralmente, entre setembro de 2012 e março de 2013 em dois sítios: próximo à nascente (S1) e próximo à foz (S2) do arroio. Ramos com botões florais foram expostos à água dos sítios e à água destilada (controle negativo). Frequências de micronúcleos (MCN) foram determinadas em células-mãe de pólen. Os seguintes parâmetros químicos: pH, sólidos dissolvidos totais, demanda bioquímica de oxigênio (DBO 5 ), oxigênio dissolvido, fósforo total (PT) e os elementos-traço cádmio, chumbo, cobre, cromo total e zinco foram analisados. Em todas as amostragens, as frequências de MCN observadas nos botões florais expostos à água dos dois sítios foram significantemente superiores (S1: 2,48 a 3,38, S2: 3,24 a 5,19) às observadas nos respectivos controles negativos (1,33 a 1,62). O PT esteve acima do limite legal ao longo de todo o período monitorado e a DBO 5 apresentou concentrações maiores àquelas estabelecidas pela legislação em dois meses no S1 e em três meses no S2. A análise de componentes principais mostrou relação entre a frequência de MCN, a DBO 5 e o PT, indicando a influência negativa de poluentes presentes na água sobre a espécie bioindicadora e reforçando a importância de se considerar os fatores ambientais de forma integrada em programas de monitoramento de corpos hídricos. between the genetic damage observed in Tradescantia pallida var. purpurea and the chemical parameters analyzed. Water samplings were collected bimonthly from September 2012 to March 2013 from two sites, near the headspring (S1) and near the mouth (S2). Cuttings with flower buds were exposed to water from the sites and distilled water (negative control). Micronuclei (MCN) frequencies were determined in pollen mother cells. The chemical parameters analyzed were pH, total dissolved solids, biochemical oxygen demand (DBO 5 ), dissolved oxygen, total phosphorus (TP) and the trace elements cadmium, lead, copper, total chromium and zinc. In all samplings, the MCN frequencies observed in buds exposed to water from both sites were significantly higher (S1: 2.48 to 3.38, S2: 3.24 to 5.19) than those observed in the respective negative controls (1.33 to 1.62). The TP was above the legal limit throughout the monitored period and DBO 5 presented concentrations higher than those established by legislation in two months at S1 and three months at S2. The principal component analysis showed a relationship between MCN frequency, DBO 5 and TP, pointing to the negative influence of pollutants present in water on the bioindicator species and reinforcing the importance of considering the environmental factors in an integrated way in water-body monitoring programs.
