The cyclocondensation of suitable CH‐acidic carbonyl compounds, aldehydes, and urea‐type blocks under acidic conditions provides multifunctionalized derivatives. The discovery of this three‐component condensation process was made by Biginelli in 1893, therefore this reaction is called the “Biginelli reaction,” “Biginelli condensation,” or the “Biginelli dihydropyrimidine synthesis.” While the early examples of this cyclocondensation process typically involved a beta‐ketoester, aromatic aldehyde, and urea, the scope has now been extended considerably by variation of three building blocks, allowing access to a large number of multifunctionalized pyridmidine derivatives. The importance of multicomponent reactions in combinatorial chemistry has generated a renewed interest in the Biginelli reaction and the number of patents and publications on this subject is growing.
In this chapter, all three‐component condensations involving suitable CH‐acidic carbonyl compounds, aldehydes, and urea‐type building blocks following the Biginelli concept are covered. Therefore, reactions involving 1,3‐diketones or nitroacetone as building blocks leading to dihydropyrimides that follow the discussed substitution pattern are included in contrast to earlier articles. Diese Heiztechnik hat sich von einer Laborkuriosität zu einer etablierten Methode entwickelt, die an Universitäten und in der Industrie stark genutzt wird. Die effiziente Verringerung der Reaktionszeiten (von Tagen und Stunden auf Minuten und Sekunden) ist nur einer der vielen Vorteile des schnellen Erhitzens mit Mikrowellen (“microwave flash heating”). Dieser Aufsatz gibt einen Überblick über neuere Anwendungen des kontrollierten Mikrowellenerhitzens in der modernen organischen Synthese und diskutiert einige grundlegende Vorgänge.
Flow chemistry studies can sometimes be difficult to reproduce. In this article we provide guidance to scientists for experimental details that should be considered as part of any organic chemistry-based continuous flow study. A focus is placed on information that should be provided within reported studies to enable experiments to be more easily and reliably reproduced. Topics covered include flow reactor components and assembly, important parameter effects and useful performance criteria. The article covers aspects of homogeneous systems, multiphase transformations, and catalytic reactions (homogeneous and heterogeneous). A more detailed discussion of photochemistry, biocatalysis and electrochemical flow systems is outside the scope of this review.
Über Schwefel zum Erfolg: Die Kreuzkupplung von organischen Schwefelverbindungen mit Boronsäuren unter neutralen Bedingungen in Gegenwart eines Palladium(0)‐Katalysators und einer stöchiometrischen Menge eines Kupfer(I)‐Cofaktors hat sich als wertvolle C‐C‐Verknüpfungsmethode etabliert (siehe Schema). Diese basenfreie Kupplung verläuft nach einem beispiellosen Mechanismus und hat deutliche Vorteile, wenn andere Pd0‐katalysierte Verfahren versagen.
Um die Fülle an bereits bekannten hoch selektiven und zuverlässigen Methoden zur C‐C‐Bindungsknüpfung zu ergänzen, besteht ein wachsendes Interesse an der Entwicklung neuer Verfahren mit einer veränderten oder orthogonalen Reaktivität. Liebeskind und Srogl beschrieben 2000 eine übergangsmetallkatalysierte Kreuzkupplung von Thioestern mit Boronsäuren zur Synthese von Ketonen unter neutralen Bedingungen. Diese desulfurierende Kreuzkupplung verläuft unter Palladium(0)‐Katalyse mit stöchiometrischem Kupfer(I)‐Zusatz, folgt einem beispiellosen Mechanismus und ist auf eine Reihe von Organoschwefelderivaten und nucleophilen Organometallreagentien anwendbar. Hier betrachten wir neuere Anwendungen dieser faszinierenden Reaktion in der organischen Synthese. Besondere Berücksichtigung finden Fälle, bei denen herkömmliche Methoden der C‐C‐Bindungsbildung versagt haben.
Ölbadchemie unter Mikrowellenbestrahlung: In Reaktionsgefäßen aus Siliciumcarbid (SiC) können Experimente, wie sie in einem Autoklaven mit Wärmeübertragung ausgeführt würden, in einem Mikrowellenreaktor simuliert werden, weil das elektromagnetische Feld der Mikrowellen durch die SiC‐Gefäße effizient abgeschirmt wird. Dieses Verfahren ermöglicht es, den Einfluss von Mikrowelleneffekten zu studieren.
Mit Stickstoffwasserstoffsäure in den Mikroreaktor! Ein Hochtemperatur‐Hochdruckprozess‐Prozessintensivierungsansatz lieferte Tetrazole auf sehr effiziente Art. Trotz des toxischen und explosiven Charakters von Stickstoffwasserstoffsäure gelang die sichere Synthese im kontinuierlichen Fluss mit Verweildauern von weniger als 2.5 Minuten bei 260 °C (siehe Bild).
