Inflammatory bowel diseases (IBD) are a major problem for public health, with an increased incidence and impact on life quality. The effect of pre- and probiotic combination has been less studied in IBD. Using genomic and proteomic array technologies, this study examined the efficacy of a new combination of natural alternatives: prebiotics (grape pomace extract, GP) and probiotics (lactobacilli mixture, Lb mix) on inflammation and intracellular signalling routes in a cellular model of inflammation. Caco-2 cells challenged with lipopolysaccharide (LPS) for 4 h were treated with GP extract (50 μg/ml gallic acid equivalent) and Lb combination (3 × 108 colony-forming units/ml total Lb) for 24 h. The profile expressions of forty key inflammatory markers and twenty-six signalling kinases were analysed. Other markers involved in inflammation were also investigated (NF-κB/RELA, Nrf2, aryl hydrocarbon receptor, Cyp1A1, Cyp1B1); 57·5 and 60 % of investigated genes and proteins, respectively, were down-regulated by the synbiotic combination. Relevant cytokines and chemokines involved in response to microbial infection and inflammation were reduced under the level induced by LPS treatment and toward the unchallenged control. As expected, the reduction effect seems to imply mitogen-activated protein kinase and NF-κB pathway. Most of the signalling molecules activated by LPS were decreased by GP extract and Lb mix. Our study indicates that the synbiotic combination of GP extract and Lactobacillus sp. mixture exerted anti-inflammatory properties, which are able to decrease the majority of inflammatory genes, their proteins and associated signalling markers. Due to protective role of GP compounds on lactobacilli probiotic, this synbiotic combination might serve as a promising adjunctive therapy in intestinal inflammations.
The aim of this work was to evaluate the influence of a diet containing 5% dried GP on the antioxidant status (total antioxidant status (TAS), antioxidant enzyme activity (catalase-CAT, superoxide dismutase-SOD, and gluthatione peroxidase-GPx), and lipid peroxidation) on the key organs of the liver, kidneys, and spleen in relation to health status as indicated by blood biochemical parameters and total polyphenol content in the blood, organs (liver, spleen, kidney, mesenteric lymph nodes, heart, and brain) and Longissimus dorsi muscle in piglets. The GP diet results in a significant increase of TAS in the liver, spleen, and kidneys, with increased CAT activity in the spleen and kidneys, increased SOD activity in the liver, kidneys, and spleen, and increased GPx activity in the kidneys, as well as a decrease in lipid peroxidation in the liver and kidneys. The GP included in the piglets’ feed contained polyphenols that showed antioxidant activity and were absorbed in the plasma, contributing to maintaining the good health of the animals. The inclusion of 5% GP inclusion in the diets of piglets is beneficial for overall normal blood constituent metabolism and helps to maintain piglet health by increasing the polyphenol content in blood plasma and antioxidant activity in the liver, spleen, and kidneys.
Poultry meat is a valuable source of nutrients and the enrichment with health-promoting substances such as polyunsaturated fatty acids (n-3 PUFA) is an important factor for consumers’ choice. Camelina meal (Camelina sativa) is an animal feedstuff used to achieve this goal, but the administration of n-3 PUFA-enriched diets in broiler nutrition can accelerate the oxidative processes in meat leading to a decreased quality of final product. The aim of this study was to investigate the effect of the organic Cr as chromium picolinate (CrPic) on meat quality, fatty acid profile of fat and oxidative stability of meat from broilers fed supplemented dietary Camelina meal. An experiment was conducted on 240 Ross 308 broiler chicken aged 14 days which were assigned to 6 dietary treatments in a randomized complete block design with a 2 × 3 factorial arrangement. Within the treatment arrangement two concentrations of Camelina meal (0% and 3%) and three concentrations of Cr3+ (0, 200 and 400 μg/kg) were used. Dietary treatments were: (1) Control diet (C) containing a corn–soybean diet with no added Camelina meal or Cr3+; (2) a C diet containing an additional 200 μg/kg of Cr3+ as CrPic; (3) a C diet containing an additional 400 μg/kg of Cr3+ as CrPic; (4) a C diet containing an additional 3% Camelina meal; (5) diet 2 containing an additional 3% Camelina meal; (6) diet 3 containing an additional 3% Camelina meal. Chromium supplementation significantly (P<0.05) increased the CP concentrations and significantly (P<0.05) decreased the crude fat concentrations in breast samples. The Camelina meal groups presented higher values of unsaturated fatty acids, particularly n-3 fatty acids (P<0.05). In CrPic groups, increased retention of Zn and Fe (P < 0.05) was observed in breast samples, compared to control group, and thiobarbituric acid reactive substances values were significantly (P<0.05) smaller. Myoglobin fraction (metmyoglobin and oximyoglobin) concentrations differ significantly (P<0.05) from the control group, under the influence of Cr3+ supplements. This study found that broilers fed with CrPic supplements showed improved mineral composition and oxidative stability of breast meat, proving an effective protection of lipid molecules from oxidation in PUFA-enriched meat.
Weaning is associated with several stress factors and their effects on the piglet's body are fairly well known. Thus, changes were estimated in certain serum and faecal parameters after weaning owing to dietary protein level, though essential amino acid (AA) levels were maintained or reduced. Eighteen Topigs piglets were assigned randomly to three homogenous groups. The dietary protein level was reduced by 10% in diets 1 and 2 compared with diet C. Diet 1 had similar levels of essential AA to diet C, while the levels of essential AA in diet 2 were reduced by 10%. Blood samples were collected by jugular venipuncture in the farrowing unit on four occasions: before and after separation from the sow; the day after transfer to the nursery; and seven days after weaning. Faecal excreta were collected daily. No major health problems arose, and total pathogen germs were not different among treatments. Cortisol concentration reached similar values to those from the farrowing unit seven days after weaning. Whatever the diet, vitamin E in plasma decreased significantly in the first seven days post weaning. The reduction of dietary protein, as well as essential AAs, adversely affected the concentration of Cu (by 17.3%) in plasma. Selenium concentration in plasma increased slightly, irrespective of diet. The authors conclude that providing dietary protein at a particular level (diet 1), while maintaining AAs at normal level, limits faecal nitrogenous content without significantly modifying stress indicators (except vitamin E) or faecal composition.
Abstract— Six trials were conducted to study the conditions of preparation and treatment of moist forage samples for the in situ measurement of their ruminal dry matter and nitrogen degradations. Drying, even at 60 °C, lowered effective nitrogen degradability against moist forage; drying at 80 °C lowered it by 10 points (P < 0.01) (1 point = 1 % on a scale from 0 to 100). Freeze-drying had a weak negative effect (-3.1 points; P < 0.05). The nitrogen degradability of hays increased with decreasing particle size (+7.7 points, P < 0.01, from 12 to 0.8 mm mesh size). The objective of this study was to identify relevant descriptors of ruminal pH post-prandial evolution that can replace the mean pH (considered unsatisfactory). These descriptors are to be used in the attempts to predict ruminal pH from dietary characteristics, in order to quantify the potential of a diet to induce subacute ruminal acidosis from its intrinsic characteristics. A total of 219 pH curves, reported as graphics in 48 published articles describing the post-prandial evolution of ruminal pH (first 8 h), were digitized by image analysis then summarized in 15 pH variables. Relationships among pH variables and the principal components (PCs) of pH variability were analyzed in order to identify possible alternatives to mean pH, as the average value of all pH data the curve is composed of. Two groups of pH variables were identified according to their relationship with the most important principal components. A first group, including mean pH, was closely related to PC 1 , which accounted for 78% of data variability; hence, correlations between variables of this group were generally high. Of these, threshold-related variables were distinct as their withinstudy correlations with mean pH were rather moderate (0.69 on average). This suggests they might carry supplementary information that could explain the variation in ruminal pH induced by within-study factors, e.g. diet characteristics. However, caution should be taken in their use because of their truncation at 0 h and their non-normal distribution. Variables from the second group were independent of the PC 1 , and thus of the first group of variables, whereas they were mostly related to PC 2 and PC 3 . This implies they are complementary to mean pH. Of this second group, the rate of pH decreases or the time period when pH reaches its minimum might be useful to better describe the ruminal status, from the point of view of the risk of subacute ruminal acidosis.
