The biosynthesis of terpene hydrocarbons has been investigated in maritime pine (Pinus pinaster Ait.) seedling primary leaves under light and darkness and with different precursors. Impossible in darkness, the synthesis of monoterpenes (mainly α‐ and β‐pinene) is strongly activated by light. Only 14C‐carbonate and 14C‐acetate can be incorporated into monoterpenes. Activation by light is comparatively much more effective for seedling leaves previously cultivated under short days than in leaves from seedlings given long days. The spectral bands which are efficient for the synthesis of monoterpenes are located around 480 and 685 nm with 14C‐carbonate and 480 and 630 nm with l‐14C‐acetate. Furthermore, this light activation does not occur if leaf pieces instead of whole leaves are used for the incorporation experiments. When 2‐14C‐mevalonic acid and 1‐14C‐isopentenyl pyrosphosphate are applied as precursors, no radioactivity is recorded in monoterpene hydrocarbons even after light exposures. In contrast, sesquiterpene hydrocarbons (caryophyllene and humulene) are easily synthesized under light or darkness in intact or fragmented leaves from the different precursors of photosynthetic or exogenous origin. From these results the compartmentalization in the synthesis of C10 and C15 hydrocarbons appears clear. There is a metabolic cooperation between the photosynthetic tissues and the specific site of elaboration of C10 hydrocarbons, which site is located in the parts where the epithelial cells of resin ducts are functional. The synthesis of sesquiterpene hydrocarbons takes place in the whole leaf without activation by light.
A plastid vesicle preparation isolated from exocarpium of young Citrofortunella mitis (calamondin) fruits was able to synthesise monoterpene hydrocarbons when incubated with isopentenyl pyrophosphate. The electron-microscope comparison between this organelle fraction and the various plastid classes present in the peel tissues has shown the structural identity between these plastid vesicles and the leucoplasts of the epithelial cells lining the secretory pockets. The monoterpene biosynthesis required the presence of dimethylallyl pyrophosphate, Mn(2+) or Mg(2+) and was increased by addition of 2-mercaptoethanol. Evidence is provided that the leucoplast vesicles act as a complete system in which occur all the successive steps involved in monoterpene hydrocarbon elaboration from isopentenyl pyrophosphate.
En ce qui concerne le 3-carène, , sur Pinus monticola, a montré que la concentration en 3-carène des tissus corticaux est contrôlée par un couple de gènes, l'allèle dominant étant responsable de la richesse en ce monoterpène. TOBOLSKY (1968) travaillant sur des familles de demi-frères (descendances maternelles) de Pin sylvestre constate simplement que les peuplements dont provenaient les descendances doivent être très hétérozygotes pour le ou les gènes contrôlant le 3-carène. Il étend la même conclusion au myrcène, au limonène, au P-phellandrène et au terpinolène; par ailleurs, SQUILLACE (1971) a montré que chez Pinus elliotii, les teneurs en (3-pinène et en myrcène sont contrôlées par un seul couple d'allèles.Pour d'autres genres, les conclusions concernant le déterminisme génétique des teneurs en monoterpènes sont moins précises. Sur Abies lasiocarpa, ZAVARIN et al. (1970) ont montré que le limonène semble être hérité indépendamment du groupe |3-pinène-phellandrène. Ils suggèrent au vu d'un histogramme multimodal, un contrôle génétique par une série d'allèles multiples pouvant occuper un même locus. Mais cette étude ne fait pas intervenir de tests de descendances et ne peut évidemment pas aboutir à des conclusions précises sur le contrôle génétique du limonène. Enfin, WILKINSON et al. (1971) suggèrent dans le cas de Picea glauca un contrôle du limonène, de l'oc-pinène et du (3-pinène par un système polygénique.Ces différents travaux n'ont donc pas conduit à des conclusions nettes sur le détermi-nisme génétique du limonène; mais, par contre, les conclusions de HANOVER sur le contrôle monogénique du 3-carène chez Pinus monticola sont irréfutables. Peuvent-elles être étendues LES TERPÈNES DU PIN MARITIME (il) 309 aux autres espèces du genre Pinus, en particulier à Pinuspinasterl C'est l'objet de la seconde partie de notre travail. L'examen de la figure 1 montre, pour le bois, une augmentation de la concentration de l'a-pinène et une chute de celle du ^-pinène lorsque l'âge augmente. Les moyennes de familles, très regroupées pour les plants de 3 ans, se différencient nettement à partir de 5 ans.Dans les tissus extérieurs au bois ( fig. 2), on observe que les teneurs en a-pinène, bien différenciées pour les plants de 3 ans (1968), convergent à partir de 5 ans (1966) pour pratiquement se confondre chez les plants de 9 ans (1962). Pour le (3-pinène, les différences entre moyennes semblent être à peu près stables à partir de 3 ans. Pour le 3-carène, les diffé-rences entre familles se manifestent très tôt et se maintiennent. Cependant, une inversion du classement est à remarquer pour les plants de 3 ans (1968) et ceux de 7 ans (1964) entre les familles dérivant des clones 00.07 et 02.05. Sans doute cela est-il en partie dû au fait que les pères utilisés en pollinisation contrôlée ne sont pas les mêmes que ceux qui ont pollinisé l'arbre en forêt. De plus, les mères très pauvres en 3-carène, comme 00.08 et 13.08 donnent des descendances beaucoup plus riches qu'elles en ce monoterpène. Dans le cas du myrcène, une différen...
