This work presents variation of oxidative catalytic activities of methanol and formic acid on Pt nanoparticles of various sizes and a comparison to the results observed on Pt(111), Pt(100), and polycrystalline Pt. The Pt nanoparticles dispersed on platelet carbon nanofiber are cuboctahedral particles, whose sizes span from 5.6 to 1.1 nm. The electrochemically active surface areas, measured using charges of hydrogen adsorption/desorption and stripping of adsorbed CO, are reasonably consistent with those calculated theoretically with a simple cuboctahedron model. However, Pt nanoparticles with extremely small size (<1.8 nm) aggregate to reduce their surface areas. The size effect of Pt nanoparticles in oxidation of methanol and formic acid is discussed in terms of specific activity (current per unit surface area) and mass activity (current per unit mass).
Thiamine diphosphate (ThDP)-dependent enzymes are ubiquitously present in all organisms and catalyze essential reactions in various metabolic pathways. ThDP-dependent phosphoketolase plays key roles in the central metabolism of heterofermentative bacteria and in the pentose catabolism of various microbes. In particular, bifidobacteria, representatives of beneficial commensal bacteria, have an effective glycolytic pathway called bifid shunt in which 2.5 mol of ATP are produced per glucose. Phosphoketolase catalyzes two steps in the bifid shunt because of its dual-substrate specificity; they are phosphorolytic cleavage of fructose 6-phosphate or xylulose 5-phosphate to produce aldose phosphate, acetyl phosphate, and H 2 O. The phosphoketolase reaction is different from other well studied ThDP-dependent enzymes because it involves a dehydration step. Although phosphoketolase was discovered more than 50 years ago, its three-dimensional structure remains unclear. In this study we report the crystal structures of xylulose 5-phosphate/fructose 6-phosphate phosphoketolase from Bifidobacterium breve. The structures of the two intermediates before and after dehydration (␣,-dihydroxyethyl ThDP and 2-acetylThDP) and complex with inorganic phosphate give an insight into the mechanism of each step of the enzymatic reaction.
Binary expression systems like the LexA-LexAop system provide a powerful experimental tool kit to study gene and tissue function in developmental biology, neurobiology, and physiology. However, the number of well-defined LexA enhancer trap insertions remains limited. A rapid increase in the number of patients with coronavirus disease 19 (COVID-19) may overwhelm the available medical resources. We aimed to evaluate risk factors for disease severity in the early stages of COVID-19. The cohort comprised 293 patients with COVID-19 from 5 March 2020, to 18 March 2020. The Korea Centers for Disease Control and Prevention (KCDC) classification system was used to triage patients. The clinical course was summarized, including the impact of drugs (angiotensin II receptor blockers [ARB], ibuprofen, and dipeptidyl peptidase-4 inhibitors [DPP4i]) and the therapeutic effect of lopinavir/ritonavir. After adjusting for confounding variables, prior history of drug use, including ARB, ibuprofen, and DPP4i was not a risk factor associated with disease progression. Patients treated with lopinavir/ritonavir had significantly shorter progression-free survival than those not receiving lopinavir/ritonavir. KCDC classification I clearly distinguished the improvement/stabilization group from the progression group of COVID-19 patients (AUC 0.817; 95% CI, 0.740–0.895).
Bifidobacterium breve was cloned and overexpressed in Escherichia coli. The enzyme was purified to homogeneity and crystallized by the sitting-drop vapourdiffusion method. Crystals were obtained at 293 K using 0.05 mM thiamine diphosphate, 0.25 mM MgCl 2 , 24%(w/v) PEG 6000 and 0.1 M Bicine pH 9.0. The crystals belonged to the tetragonal space group I422, with unit-cell parameters a = b = 174.8, c = 163.8 Å , and diffracted to beyond 1.7 Å resolution.
This study investigated the effects of nano-silica (NS) and silica fume (SF) on the hydration reaction of high-volume fly ash cement (HVFC) composites. In order to solve the dispersibility problem caused by the agglomeration of NS powder, NS and NSF solutions were prepared. NS content and SF content were used as main variables, and an HVFC paste was prepared in which 50% of the cement volume was replaced by fly ash (FA). The initial heat of hydration was measured using isothermal calorimetry to analyze the effects of NS and SF on the initial hydration properties of the HVFC. In addition, the compressive strength was analyzed by age. The refinement of the pore structure by the nanomaterial was analyzed using mercury intrusion porosimetry (MIP). The results show that the addition of NS and SF shortened the setting time and induction period by accelerating the initial hydration reaction of HVFC composites and improved the compressive strength during the initial stage of hydration. In addition, the micropore structure was improved by the pozzolanic reaction of NS and SF, thereby increasing the compressive strength during the middle stage of hydration.
