Resumo: Para padronização de uma técnica manual para a obtenção de plasma rico em plaquetas (PRP) autólogo em bovinos com custo reduzido (método manual) e de boa qualidade (capacidade de concentrar plaquetas, alta concentração de fatores de crescimento e contaminação reduzida com leucócitos e eritrócitos), que poderá ser utilizado como um agente modulador da resposta imune de vacas com diferentes enfermidades, 450 ml de sangue total de nove vacas clinicamente saudáveis e com perfil hematológico normal foi coletado em bolsas de sangue CPDA-1 e processado dentro de quatro horas após a coleta. O sangue foi separado em alíquotas para avaliar 8 protocolos (P) de centrifugação dupla que variaram quanto a velocidade e o tempo de centrifugação. A contagem de plaquetas, eritrócitos e leucócitos na suspensão obtida (PRP) foi realizada pelo método manual em câmara de Neubauer: P5 (400g e 800g ambos durante 10 min) foi o protocolo com maior número de plaquetas, seguido por P3 (120g e 473g ambos durante 10 min), P4 (300g e 640g durante 10 min cada), P6 (640g durante 10 min e 640g durante 5 min), P8 (640g durante 5 min e 120g durante 10 min) e P7 (720g e 720g durante 5 min) e diferentes (p<0,05) dos menores valores encontrados em P1 (120g e 240g, ambos por 5 minutos) e P2 (120g e 473g ambos por 5 min). Em relação aos eritrócitos, P8, P7, P6, P5 e P4 apresentaram menores concentrações e maiores valores (p<0,05) foram observados em P3 e P2. Menores quantidades de leucócitos foram observadas em P5, P6, P8 e P7 com o maior valor obtido em P2 (p<0,05). Todos os protocolos (P1 a P8) foram eficientes em concentrar plaquetas sendo o valor mais baixo (3,65±0,79) observado em P1. Em relação aos fatores de crescimento ao se mensurar TGF- 1, os protocolos P1 e P8 evidenciaram valores mais elevados. De acordo com os resultados obtidos os protocolos P5 e P8 apresentaram os melhores resultados para confecção de PRP bovino.

Background: Canine transmissible venereal tumor (TVT) is one of the most common canine neoplasms in Brazil. Lesions develop mainly in the genital tissue and less frequently in other areas, in the extragenital form. Metastasis is rare, and the disease progression depends on the patient's immune status. The treatment of choice is chemotherapy with vincristine sulfate, considered an effective and safe modality. The present work describes the first case of extragenital TVT in the larynx with pulmonary metastasis at the time of diagnosis. Case: An approximately 4-month-old mixed-breed bitch was attended manifesting acute clinical respiratory signs. The patient had been rescued from the street 2 months beforehand. Physical examination revealed pale mucous membranes and crackling pulmonary auscultation. Thoracic radiography was performed in 3 projections, showing multiple nodules in the lung parenchyma, followed by cytopathological examination using fine needle puncture guided by thoracic ultrasound, confirming the diagnosis of TVT. Considering the cytological finding and the absence of primary tumor lesions in the genital, oral, cutaneous or nasal regions, endoscopy was performed in order to locate a possible primary focus. The exam revealed a small erythematous tumor with smooth and regular surface, measuring approximately 2.5 cm, located in the larynx, between the arytenoid cartilages. Chemotherapy was performed with vincristine sulfate with radiographic follow-up throughout the treatment. Full remission of the pulmonary and laryngeal nodules was achieved at the end of the protocol. The animal remained in complete remission for a period of 2 years. Discussion: TVT is more frequently observed in animals of active reproductive age, and rarely in animals under 1 year of age, as in the case described here. When considering extragenital lesions, the manifestation can be single or multiple, occurring more frequently in the cutaneous, oral and nasal forms. This case describes a laryngeal lesion with pulmonary involvement, without lesion in the genitalia or any other site. Cases of TVT in young and prepubescent animals, associated with extragenital involvement and aggressive behavior, similar to this case, have been described in literature, but never in this location. Literature suggests there is a relationship with the immune status of such patients, which do not have a well-developed immune system, facilitating disease progression. In these animals, transmission can be maternal, since in some cases the patient develops the disease without having contact with other animals except the mother. This form of transmission would facilitate the extragenital form in the oral cavity, airways and ocular region, related to licking, lactation and direct contact. Vincristine sulfate chemotherapy, considered the treatment of choice, was effectively used in this case, despite the atypical form. The TVT prognosis tends to be favorable, with the exception of the aggressive presentations with the presence of distant metastases. However, in the case described here, even though metastatic pulmonary nodules were present, complete remission was achieved, with survival exceeding 2 years. The development of TVT in the larynx is a rare form of the disease, and this report is possibly the first description of this tumor location. Thus, we highlight the importance of a complete investigation of the patient and of TVT as a differential diagnosis in this location. RESUMO A dislipidemia é um achado comum, porém não determinante, na síndrome metabólica equina (SME). O objetivo do presente trabalho foi caracterizar a dislipidemia em animais obesos com risco de SME. Para isso, 18 éguas foram alocadas em grupos, de acordo com escore corporal (EC) de 1 a 9: no grupo ideal, animais com EC de 4,5 a 5,5 (n= 6), no grupo sobrepeso, com EC de 6 a 7 (n= 6) e no grupo obeso, animais com EC de 7,5 a 9 (n= 6). Coletaram-se amostras de sangue em jejum de concentrado para determinação de triglicerídeos, colesterol total, glicemia e concentração de insulina. Valores preditivos de sensibilidade à insulina (RISQI) e de secreção β-pancreática (MIRG) foram calculados. O grupo obeso apresentou níveis maiores em relação aos outros grupos de triglicerídeos (P=0,001) e acima do ideal em concentrações de colesterol (P=0,012). Não foi observada diferença nas concentrações plasmáticas de glicose (P=0,53), de insulina (P=0,10) ou de RISQI (P=0,46). Houve diferença entre os grupos nos valores de MIRG (P=0,048), tendo o grupo obeso obtido resultados maiores quando comparado com o grupo ideal. O aumento do EC foi associado ao aumento das concentrações plasmáticas de colesterol e triglicerídeos, o que caracteriza um estado de dislipidemia e de elevação da secreção das células β-pancreáticas.

Cattle (Bos taurus and Bos indicus) are organised on the basis of leadership and dominance in such a manner that a disturbance by an external stressor causes negative effects on their health, productivity, well-being, and behaviour. One of these effects is the excessive release of glucocorticoids, which results in increased alertness. We evaluated the action of exogenous oxytocin (OT) on serum cortisol levels in crossbred Red Angus heifers. Twelve Red Angus crossbred heifers were moved daily from the pasture to the corral in weeks 1 and 2 for adaptation to human contact and handling in the cattle crush. In weeks 3 and 4, they were divided into two groups of six (T1 and T2). The T1 group was administered 20 IU (2 mL) of OT via intramuscular injection and the T2 group was administered 2 mL of saline solution 0.85% (SS). In weeks 5 and 6, they were only contained in the cattle crush for evaluation. On days 01, 07, 14, 21, 28, 35, and 42, blood samples were collected by jugular venepuncture in vacuum tubes without anticoagulants. Then, serum cortisol levels were measured using a radioimmunoassay. In the period of adaptation, during weeks 1 and 2, serum cortisol levels decreased in both the groups, with higher levels in the SS group; the same result was obtained in weeks 5 and 6. During treatment, however, there was a significant difference between the two groups in week 4, with a reduction in cortisol levels in the OT group. This result suggests a modulator effect of OT on neuroendocrine response to stress.
