RESUMO - Durante o processo de produção de sementes, estresses abióticos como salinidade do solo e déficit hídrico são fatores determinantes e que influenciam no estabelecimento e desenvolvimento uniforme do estande em campo. Nesse sentido, cultivares tolerantes a altas concentrações salinas nestas fases são fundamentais nos programas de melhoramento. Diante disto, objetivou-se com a presente pesquisa verificar a influência da produção de sementes sob condições de estresse hídrico e salino na germinação e no vigor, assim como na atividade da enzima α-amilase em sementes de milho. Para isso, foram conduzidos dois experimentos para a produção de sementes de milho, um em condições de estresse salino e o outro sob condições de déficit hídrico. Os experimentos foram conduzidos em delineamento de blocos completos casualisados, em esquema de parcelas subdivididas. As sementes colhidas em diferentes estádios de desenvolvimento, determinados pela linha de leite (LL), foram submetidas a análises fisiológicas e de vigor, bem como determinou-se a expressão da enzima α-amilase por eletroforese. Diante dos resultados foi possível concluir que a germinação e o vigor das sementes produzidas em solos com condutividade de 3 dS m-1 não são afetados pela salinidade, e o desenvolvimento das plantas em condições de estresse hídrico e salino não afetou a atividade da enzima α-amilase em sementes colhidas na LL5. PHYSIOLOGICAL QUALITY AND ALPHA-AMYLASE EXPRESSION OF MAIZE SEEDS IN CONDITIONS OF SALT AND WATER STRESS ABSTRACT - During seed production process, abiotic stresses such as soil salinity and drought are determining factors that influence the establishment and development of uniform stand on the field. Thus, tolerant cultivars to high salt concentrations in these phases are critical in breeding programs. This way, we aimed to investigate seeds production in conditions of water and salt stress on maize seeds germination and vigor as well as the α-amylase enzyme activity. Two experiments were performed for maize seeds production in salt stress conditions and drought. The experiments were conducted in a randomized complete block design in a split-plot arrangement. Seeds harveste at different development stages, determined by milk line (ML), were submitted to vigor and physiological analysis as well as the expression determination of α-amylase enzyme by electrophoreses. With the results it was concluded that both, the seeds germination and vigor produced in soil with conductivity 3 dS m-1, are not affected by salinity, and the plants development in water and salt stress conditions did not affect the activity of α-amylase enzyme in seedsharvested in ML5 maturity stage. The objective of this study was to evaluate gene expression related to water deficit tolerance in maize lines. For this, lines previously classified as tolerant (91-T and 32-T) and non-tolerant (24-NT and 57-NT) to water deficit were used. The seeds of the four lines were evaluated for emergence and emergence speed index, and the seedlings were evaluated for root and shoot length under two conditions of water availability (70 and 10% substrate water retention capacity). In transcript analysis, the expression of several genes associated with water deficit tolerance, ZmDBP3, ZmALDH9, ZmAN13, and ZmDREB2A, was evaluated by qRT-PCR for the 91-T and 57-NT lines. It can be concluded that soil water deficiency did not reduce root development. However, the shoot length was significantly lower under dry conditions. Through transcript analysis, the genes ZmDBP3 and ZmAN13 were identified as potential markers for the early selection of maize lines tolerant to water deficit.
<p>O fluxo gênico, ou escape gênico, é definido como a troca de alelos entre indivíduos, isto é, a transferência de alelos de uma variedade/espécie para outra. Os fatores climáticos, como velocidade e direção do vento, temperatura e umidade relativa podem interferir no fluxo gênico por afetar a distância, a duração e a quantidade de pólen liberado. A utilização da engenharia genética nos programas de melhoramento genético de forrageiras tropicais e a falta de informação sobre o fluxo gênico nessas gramíneas gerou a necessidade de se conhecer o fluxo de genes em espécies de valor econômico, principalmente do gênero <em>Brachiaria </em>e<em> Panicum</em> por serem as mais utilizadas na dieta de ruminantes no Brasil. Os objetivos desta revisão foi esclarecer o que é o fluxo gênico e como pode ocorrer na natureza; além disso, mostrar os modelos experimentais de investigação usados no melhoramento genético de plantas forrageiras e gerar conhecimento para futuras aplicações em sistemas agrícolas transgênicos com espécies dos gêneros <em>Brachiaria</em> e <em>Panicum</em> em regiões tropicais. O fluxo gênico é um evento natural entre diferentes espécies de plantas e pode ser controlado por barreiras físicas, temporais ou modo de reprodução. O delineamento mais utilizado para a investigação de fluxo gênico em gramíneas é o concêntrico, e análises moleculares são excelentes ferramentas para auxiliar na investigação de fluxo gênico em plantas. Conhecer o fluxo gênico em plantas é indispensável para garantir a liberação e regularização de eventos geneticamente modificados.</p><p>Palavras-chave: Escape gênico.
