Potency analysis of mesenchymal stromal cells (MSCs) is required for their use in advanced clinical trials. Assay matrix strategy evaluating more than a single property of MSCs is an emerging strategy in potency analysis. Here we developed an assay matrix approach focusing on the secretory chemokine responses of MSCs using multiplex analytical method. MSCs’ innate fitness in secreting matrix of chemokines is correlated with their metabolic fitness in differential degrees. In addition, innately secreting chemokines are correlated among themselves in a unique pattern. MSC’s matrix chemokine responses to exogenous stimulation of IFNγ and/or TNFα are distinct. However, the combination of IFNγ and TNFα is superior than individual stimulations in eliciting robust and broad matrix chemokine responses of MSCs. Correlation matrix analysis has identified that chemokine responses to IFNγ and/or TNFα display unique correlative secretion patterns. MSC and peripheral blood mononuclear cells coculture analysis has identified the correlation matrix responses of chemokines that predicted immune suppression. In addition, MSC-mediated blocking of T-cell proliferation predominantly correlates with chemokines in an inverse manner. Knockdown of chemokines has demonstrated that MSC-sourced inherent chemokines do not actively play a role in T-cell suppression and thus are the bystander predictors of T-cell suppression. The present analysis of MSC’s matrix chemokine responses can be deployed in the advanced potency analysis of MSCs.
Galaxy is a popular framework for execution of complex analytical pipelines typically for large data sets, and is a commonly used for (but not limited to) genomic, genetic and related biological analysis. It provides a web front-end and integrates with high performance computing resources. Here we report the development of the blend4php library that wraps Galaxy’s RESTful API into a PHP-based library. PHP-based web applications can use blend4php to automate execution, monitoring and management of a remote Galaxy server, including its users, workflows, jobs and more. The blend4php library was specifically developed for the integration of Galaxy with Tripal, the open-source toolkit for the creation of online genomic and genetic web sites. El desarrollo de software es una actividad demandada por el mundo entero, en el que se requiere de habilidades técnicas y blandas para desempeñarse con destreza en una carrera tan globalizada y competitiva. Las investigaciones revisadas muestran la existencia de una brecha entre la industria y la academia. Los centros de formación proveen a los profesionales de habilidades técnicas, sin embargo en estos ultimos tiempos manifiesta la necesidad de contar con habilidades blandas, por lo que es clave estudiarlas y encontrar maneras de poder insertarlas en los profesionales que se integran a la industria. Existe una metodología llamada PSP creada por Watts Humphrey, que ayuda a cada individuo a fortalecer sus destrezas en el proceso de desarrollo de software, a pesar de eso, el método no es muy difundido en la académia y la industria. Esta investigación muestra la importancia de las habilidades blandas según empresas de la región Latino América y el aporte de PSP puede hacer para reducir la brecha de estas habilidades
DevOps is a philosophy and framework that allows software development and operations teams to work in a coordinated manner, with the purpose of developing and releasing software quickly and cheaply. However, the effectiveness and benefits of DevOps depend on several factors, as reported in the literature. In particular, several studies have been published on software test automation, which is a cornerstone for the continuous integration phase in DevOps, which needs to be identified and classified. This study consolidates and classifies the existing literature on automated tests in the DevOps context. For the study, a systematic mapping study was performed to identify and classify papers on automated testing in DevOps based on 8 research questions. In the query of 6 relevant databases, 3,312 were obtained; and then, after the selection process, 299 papers were selected as primary studies. Researchers maintain a continuing and growing interest in software testing in the DevOps context. Most of the research (71.2%) is carried out in the industry and is done on web applications and SOA. The most reported types of tests are unit and integration tests.
