La infección por SARS-CoV-2 promueve un descontrol del sistema inmune con predominio de una respuesta inflamatoria de rápida instauración y el desencadenamiento de una tormenta de citocinas. El objetivo de esta revisión es comprender mejor los mecanismos inmunológicos y su descontrol activados por SARS-CoV-2. La activación de la inmunidad innata, contra la infección, es el factor clave para el control viral o su progresión hacia COVID-19. La respuesta inmune adaptativa, igualmente es perturbada por SARS-CoV-2 con lo cual se posibilita una mayor tasa de infección, así como también el descontrol inmunológico sistémico que puede desencadenar shock y daño tisular en diferentes órganos, muerte por COVID-19 o autoinmunidad post COVID-19.
El artículo se centra en la utilización de la nueva herramienta, CRISPR (repeticiones palindrómicas cortas agrupadas a intervalos regulares), la cual permite editar los genomas de los seres vivos de manera más precisa que otras técnicas; a lo largo del artículo se mencionan trabajos relacionados con la detención de la angiogénesis, cáncer, Sarcoma de Kaposi en inmunodeficiencias, Parkinson, regeneración y modificación genética en humanos, todas estas investigaciones tiene en común la utilización de la herramienta CRISPR. También se comenta las complicaciones éticas que conlleva utilizar esta tecnología en el ADN de células embrionarias humanas, que según diferentes criterios, podrían llevar a generar seres humanos “mejorados”, es decir no solo sin susceptibilidad a enfermedades degenerativas o incurables, sino también modificados en aspectos físicos que no necesariamente estarían ligados a alguna patología.
Para enfrentar la pandemia por COVID-19 Bolivia implementó medidas de contención reflejadas en el Plan Nacional de vacunación que contempla realizar pruebas diagnósticas rápidas, vacunar masivamente mediante mecanismo COVAX y compra directa de vacunas y coordinar la implementación de vacunación universal gratuita. El personal de salud fue el primer grupo vacunado y hasta diciembre de 2021 se habilitó la vacunación para menores de edad de 5 a 11 años. Con relación a los puntos de vacunación, Beni es el departamento con menos puestos y Santa Cruz concentra la mayor cantidad; a pesar de los esfuerzos solo el 52,8 % de la población boliviana cuenta con un esquema completo donde Santa Cruz y Pando registran el mayor y menor porcentaje de vacunados respectivamente, según reporte hasta octubre de 2022, si se compara este porcentaje con los demás países latinoamericanos se observa que Bolivia es el 8vo país con menos vacunados. La distribución de vacunas anti COVID-19 reveló problemas importantes en Bolivia, en relación con requerimientos mínimos de insumos para conservación de vacunas, mecanismos y políticas de aplicación de dosis a los grupos vulnerables. Introducción: La Proteína Quinasa Activada por AMP (AMPK), es una enzima monitora y reguladora central del estado energético celular, por tanto, es responsable de la respuesta celular al suministro y demanda de energía. El AMP actúa como activador en condiciones de déficit energético, mientras que el ATP la inactiva cuando las condiciones energéticas son más favorables. Debido a su función central en el metabolismo, la AMPK surge como un blanco proteico prometedor para el tratamiento de diferentes enfermedades como la Diabetes Mellitus tipo 2 (DM2), Síndrome Metabólico (SM), Cáncer, entre otros. Existen múltiples isoformas de AMPK que se regulan y expresan diferencialmente en todo el organismo. La isoforma AMPK–β2 se expresa casi exclusivamente en músculo esquelético y dado que este es el órgano primario para el almacenamiento y eliminación de Glucosa, AMPK–β2 puede dirigir su homeostasis por una ruta independiente a la Insulina. La molécula activadora SC4 tiene una gran selectividad por AMPK–β2 y debido a su función biológica, podría servir como modelo farmacológico para coadyuvar el tratamiento de enfermedades metabólicas. Objetivo: Análisis de la dinámica molecular de activación de la AMPK–β2. Metodología: En el presente estudio, se emplean herramientas bioinformáticas como Chimera 1.15 y Phyton Molecular Viewer. Resultados: El análisis in silico permitió comprender varios aspectos estructurales relacionados con la acción de SC4 sobre la estructura trimérica de la AMPK, los aminoácidos con los que interacciona y cómo su estructura química le otorga gran selectividad. También fue útil para en un futuro, ampliar los criterios de extracción, identificación y/o diseño de compuestos activos a partir de fuentes naturales, con propiedades funcionales similares o aún mejores a SC4, para así poder emplearlos con un enfoque terapéutico que beneficie a nuestra población.
