The polyphosphinoboranes [PhPH−BH2] n (1), [iBuPH−BH2] n (2), [(p-nBuC6H4)PH−BH2] n (3), and [(p-dodecylC6H4)PH−BH2] n (4) were prepared from the corresponding phosphine−borane adducts RPH2·BH3 (R = Ph, iBu, p-nBuC6H4, p-dodecylC6H4) via a rhodium-catalyzed dehydrocoupling procedure at elevated temperatures (ca. 90−130 °C). Samples of polymers 1 and 2 and the new materials 3 and 4 were characterized by multinuclear NMR spectroscopy, and the molecular weights were determined by light scattering methods in THF or CH2Cl2 solutions. The absolute weight-average molecular weight of 2 was determined by static light scattering and found to be M w = 13 100, and values of M w of ca. 20 000 were estimated for the sample of polymers 1 and the new material 3 using dynamic light scattering (DLS). The molecular weights of polymers 3 and 4 were also analyzed by gel permeation chromatography using polystyrene standards, and values up to M w = ca. 80 000, M n = ca. 10 000 were determined for 3 and M w = ca. 168 000, M n = ca. 12 000 for 4. The chemical stability of 1 in THF toward HNEt2 or nBu3P was demonstrated using NMR spectroscopy and DLS analysis, which indicated that no significant polymer degradation occurred. WAXS analysis of 1 and 3 showed that the polymers are amorphous. The glass transition temperatures (T g) of polymers 2, 3, and 4 were analyzed by DSC and were detected at ca. 5, 8, and −1 °C, respectively. TGA analysis on 1−3 revealed T 5% values (temperature for which 5% or the weight is lost) of 240 °C for 1 and ca. 150−160 °C for 2 and 3. After heating to 1000 °C, ceramic yields in the range of 35−80% were obtained. The high ceramic yield for 1 (75−80%) indicates that this material is of interest as a pyrolytic precursor to boron phosphide-based solid-state materials.
An animalic note: The first total synthesis of the all-cis nupharamine 2, an alkaloid from beaver castoreum, is based on the stereoselective domino Mannich-Michael reaction of N-galactosylfurylaldimine to give 1 (Piv = pivaloyl), subsequent conjugate cuprate addition, and stereoselective protonation of the enolate. These reactions are all controlled by the carbohydrate. Protonation of the enolate after cleavage of the auxiliary leads to epimer 3.
Castoreum ("Bibergeil"), die getrockneten Duftdrüsen des kanadischen Bibers (Castor fiber L.), gehörte einst zu den wertvollsten tierischen Duftstoffen in der Parfümindustrie, [1,2] ist heute aber nur noch in homöopathischen Zubereitungen zugelassen. Es enthält eine Reihe von Stickstoffbasen, [3] die eigentlich als Furansesquiterpene einzuordnen sind, traditionell aber zu den Alkaloiden gerechnet werden.[4] Sie haben ein Chinolizidin-oder Indolizidin-Grundgerüst und werden unter dem Begriff Nuphar-Alkaloide zusammengefasst. Eine Minderkomponente, deren Gehalt < 0.0002 % beträgt, ist ein 5-(3'-Furyl)-8-methylindolizidin, dessen Konstitution mithilfe massenspektrometrischer Analysen aufgeklärt wurde.[1] Seine relative und absolute Konfiguration sind bis heute unbekannt.Alle bekannten Chinolizidine aus dem Castoreum sind an den Kohlenstoffatomen C-4 und C-10 S-konfiguriert. (À)-Desoxynupharidin (1) und sein Epimer (À)-1-epi-Desoxynupharidin (2) sind beide in den Duftdrüsen des Bibers enthalten. Auf Grundlage dieser Tatsache kommen für das natürliche 5-(3'-Furyl)-8-methylindolizidin die Diastereomere 3 und 4 in Frage. Für 4 sind bereits zwei enantioselektive Totalsynthesen beschrieben worden, [5,6] das Nupharamin 3 hingegen konnte bisher noch nicht enantioselektiv hergestellt werden.Wir beschreiben hier enantioselektive Totalsynthesen von 3 und 4. Zur Kontrolle der relativen und absoluten Konfiguration wird das Galactosylamin 5[7] als chirales Auxiliar verwendet. Um den Schlüsselbaustein N-Galactosyldidehydropiperidinon zu erhalten, wird Furan-3-carbaldehyd (6) mit 5 in siedendem 2-Propanol zum N-Galactosylimin 7 umgesetzt.Dieses reagiert in einer von Zinkchlorid katalysierten, hochstereoselektiven Domino-Mannich-Michael-Reaktion [8] mit dem methylsubstituierten Danishefsky-Dien 8[9] und ergibt nach saurer Aufarbeitung das Piperidinon 9 (Schema 1). Die Abschirmung der Re-Seite des Imins durch die anspruchsvolle 2-Pivaloyloxygruppe des Auxiliars bewirkt eine selektive Bildung des stereogenen Zentrums im Heterocyclus. [8,10,11] Die O-geschützte Alkylseitenkette [12] wird durch konjugierte Addition einer Organokupferverbindung eingeführt. Die besten Ergebnisse wurden mit dem "Komplexreagens" RCu·BF 3 (R = (CH 2 ) 3 OTIPS) erzielt.[13] Dieses wird aus der entsprechenden Grignard-Verbindung durch Transmetallierung mit CuBr·SMe 2 hergestellt. Die Addition der Organokupferverbindung erfolgt, unterstützt durch die abschirmende 2-Pivaloyloxygruppe, in hoher Ausbeute diastereoselektiv cis zum Furylsubstituenten (Schema 2). Das gebildete Enolat wird bei tiefen Temperaturen erstaunlich stereoselektiv protoniert, sodass als Hauptprodukt das all-cis-Isomer 11 a entsteht (Schema 3). Auch die Protonierung wird durch das Kohlenhydrat kontrolliert, das wegen des exo-anomeren Effekts und der sterischen Abstoßung zwischen der eingeführ-ten Seitenkette und dem 2-Pivaloyloxysubstituenten die Sesselkonformation 11 begünstigt.
