Protein freeze-thawing is frequently used to stabilize and store recombinantly produced proteins after different unit operations in upstream and downstream processing. However, freeze-thawing is often accompanied by product damage and, hence, loss of product. Different effects are responsible, including cold denaturation, aggregation effects, which are caused by inhomogeneities in protein concentration, as well as pH and buffer ingredients, especially during the freeze cycle. In this study, we tested a commercially available small-scale protein freezing unit using immunoglobin G (IgG) as monoclonal antibody in a typical formulation buffer containing sodium phosphate, sodium chloride, and Tween 80. Different freezing rates were used respectively, and the product quality was tested in the frozen sample. Spatially resolved tests for protein concentration, pH, conductivity, and aggregation revealed high spatial differences in the frozen sample. Usage of slow freezing rates revealed high inhomogeneities in terms of buffer salt and protein distribution, while fast rates led to far lower spatial differences. These protein and buffer salt inhomogeneities can be reliably monitored using straight forward analytics, like conductivity and photometric total protein concentration measurements, reducing the need for HPLC analytics in screening experiments. Summarizing, fast freezing using steep rates shows promising results concerning homogeneity of the final frozen product and inhibits increased product aggregation.

ProblemstellungEin zentrales Problem in der Biotechnologie ist die Aufarbeitung der produzierten Wertstoffe. Etwa 5 % (bei Massenprodukten) bis zu 90 46 (bei Pharmazeutika) der Kosten des Herstellungsprozesses entstehen im down-stream-processing. Eine interessante alternative Aufarbeitungsoperation zur selektiven Separation von kolloidalen Partikeln und Makromolekiilen ist dabei die Adsorption in Wirbelschichten. In den letzten Jahren wurde diese Methode intensiv bearbeitet und zeigt ein groBes Potential, da bei einer Adsorption in der Wirbelschicht gegenuber einem Festbett z. B. keine vollstandig partikelfreie Losung vorliegen mug [l]. Die Aufarbeitung kann somit in weniger Schritten und damit sowohl schneller als auch kostengunstiger durchgefuhrt werden. 2.1.2 Krafte in der magnetisch stabilisierten Wirbelschicht Bilanziert man uber ein einzelnes Partikel in der magnetisch stabilisierten Wirbelschicht, so setzt sich die Resultierende aus Gewichtskraft F , Auftriebskraft Fa, Widerstandskraft F , mit dem Widerstandsbeiwert c , und die durch das Magnetfeld erzeugte Kraft FM zusammen [Z]'). Durch den Einsatz magnetisierbarer Adsorptionspartikel kann man rnit Hilfe eines Magnetfeldes die Partikelbewegung einschranken. Eine solche magnetisch stabilisierte Wirbelschicht, auch MSFB (magnetically stabilized fluidized bed) abgekurzt, ist der Gegenstand dieser Untersuchungen. Die Stabilisierung der Wirbelschicht erfolgt durch ein homogenes Magnetfeld, welches durch stromdurchflossenene Ringspulen in einer Spulenanordnung nach H E L M H 0 LTZ erzeugt wird. Dabei wird der der Abstand zweier benachbarter Spulen so gewahlt, daB dieser dem Spulenradius entspricht. Im homogenen Magnetfeld wirkt keine Kraft von aul3en auf die Partikel, die Stabilisierung basiert auf den Wechselwirkungen zwischen den magnetisierten Partikeln im System und der durch die Partikel lokal veranderten Magnetfelddichte. Bei einem homogenen Magnetfeld, das einem Gradienten der magnetischen Feldstarke von Null entspricht, wird keine resultierende Kraft vom Magnetfeld auf das Partikel ausgeubt [3]. 2.1...
