O Citrato de Sildenafila (CS) é um fármaco amplamente utilizado por homens, para o tratamento, principalmente, contra disfunção erétil. Muitas vezes o tratamento não tem acompanhamento médico tornando-se assim um fator preocupante para a comunidade médico-científica. Este estudo teve como objetivo investigar as possíveis alterações do uso crônico de Citrato de Sildenafila no hipotálamo posterior de ratos. Foram utilizados 10 cérebros de ratos (Rattus norvegicus) machos da linhagem Wistar. Os animais foram alojados em caixas contendo três animais, tratados com ração comercial e água à vontade e mantidos em ciclo de 12 horas claro-escuro e, divididos em 2 grupos: G1 – grupo controle (sem medicação) e G2 – grupo Sildenafil (10 mg/kg). Os resultados obtidos mostram que houve uma diferença significativa do grupo controle (25,43 ± 11,82) em relação ao grupo tratado com CS (20,65 ± 9,22), ou seja, no grupo CS, a densidade neuronal foi 18,79% menor que no grupo controle. Estes dados permitem concluir que, o uso indiscriminado de Citrato de Sildenafila diminui o número de corpos celulares de neurônios na área hipotalâmica posterior do cérebro de ratos machos. Esta perda neuronal pode ocasionar alterações clínicas graves nos sistemas em que o hipotálamo está envolvido.
