Abstract. Mediterranean fruit fly Ceratitis capitata (Diptera: Tephritidae) larvae are capable of developing in one of many hosts that may vary greatly in quality. We hypothesized that they will respond to the larval environment in a manner beneficial to their subsequent reproductive performance. Accordingly, we investigated the effects of various larval diets (varying in the amount of protein and sugar they contain) on the size, development time, nutritional status and reproductive maturation (ovarian development and onset of sexual behaviour) of females and males. We found that flies which undergo larval development in artificial host fruit that contain sugar and protein (`protein-fed') were larger, developed faster and emerged with more nutritional reserves than flies that were protein-deprived as larvae. Protein-fed males, regardless of their size, became sexually active before males that developed in hosts with no protein. Protein-fed females produced more mature eggs than protein-deprived ones. Moreover, protein-fed females tended to copulate sooner than females that developed in hosts with no protein. In addition, regardless of female larval diet, females with more mature eggs tended to copulate sooner than females with less mature eggs. In light of these results, the importance of the larval environment for adult reproductive success is discussed.
Post-teneral diets containing protein have been shown to enhance the copulatory success of sterile male Mediterranean fruit flies, Ceratitis capitata (Wied.) (Diptera: Tephritidae). However, ingesting protein was also found to negatively affect male survival, in particular when males faced starvation following release in the field. Accordingly, the objective of the present study was to determine the effects of various post-teneral diets, presented to sterile males prior to release, on their subsequent ability to forage for carbohydrates and protein in the field. Using mark-release recapture and analytic biochemical methods, we found that both protein-fed and protein-deprived males foraged successfully for protein and sugar in a field enclosure when these resources were available. We conclude that protein-fed sterile males are able to exploit sources of nutrition in the release environment, and their inability to overcome starvation is not a concern for control operations using the sterile insect technique.
A [3H]-PBAN (pheromone biosynthesis-activating neuropeptide) analog was synthesized, and binding of the radioligand to a specific PBAN-antiserum was achieved. The inhibition of binding of the radioligand by unlabeled PBAN, several PBAN analogs, and other competitors was studied and a specific radio-immunoassay was developed. Using this radioimmunoassay we found PBAN-like immunoreactivity in methanol extracts of hemolymph and neural tissues from females. Higher levels of PBAN-like immunoreactivity in extracts of brain-suboesophageal ganglion complexes, corpora cardiaca, thoracic ganglia, and abdominal ganglia were observed during the 4-5th h scotophase when compared to the PBAN-like immunoactivity levels during the 6-11th h photophase. On the other hand, the concentrations of PBAN-like immunoreactivity, in the terminal abdominal ganglion were higher during the photophase relative to minimal levels observed during the scotophase, indicating an accumulation before the onset of pheromone production. These differences in concentrations of PBAN were also reflected in the stimulation of in vitro pheromone glands, whereby significant stimulations were obtained by scotophase and photophase brain extracts, scotophase thoracic ganglia extracts, and photophase terminal abdominal ganglia extracts. No detectable levels of PBAN were found in hemolymph extracts during the sampling periods.
The Spodoptera littoralis multicapsid nuclear polyhedrosis virus (SlMNPV) is a member of the Baculoviridae that shows a distant genetic relationship to the prototype Autographa californica MNPV (AcMNPV). Using an AcMNPV gene-specific probe, we identified and mapped an ecdysteroid UDP-glucosyltransferase (egt) gene in the genome of SlMNPV. Sequence determination of a part from the hybridizing DNA fragment revealed an open reading frame of 1548 nucleotides that exhibits 38% and 44% identity to the egt amino acid sequences of AcMNPV and Lymantria dispar MNPV (LdMNPV), respectively. Sequences flanking the SlMNPV egt gene, including the promoter region, were found to be unique to the virus. The presence of this nonstructural gene in SlMNPV and several other baculoviruses points to the importance of egt for the viral infection process.
