A fungal strain of Aspergillus niger was recovered from sediments collected in the Northeast coast of Brazil (Pecém's offshore port terminal). Cultivation in different growth media yielded a new ester furan derivative, 1, along with malformin A1, malformin C, cyclo (trans-4-hydroxy-L-Pro-L-Leu), cyclo (trans-4-hydroxy-L-Pro-L-Phe), cyclo (L-Pro-L-Leu), cyclo (L-Pro-L-Phe), pseurotin D, pseurotin A, chlovalicin, cyclo (L-Pro-L-Tyr) and cyclo (L-Pro-L-Val). Compound 1 was cytotoxic against HCT-116 cell line, showing IC = 2.9 μg/mL (CI 95% from 1.8 to 4.7 μg/mL).
The cytotoxic activities of extracts (50 μg/ml) from 48 fungal strains, recovered from sediments of Pecém's offshore port terminal (Northeast coast of Brazil), against HCT-116 colon cancer cell lines were investigated. The most promising extract was obtained from strain BRF082, identified as Dichotomomyces cejpii by phylogenetic analyses of partial RPB2 gene sequence. Thus, it was selected for bioassay-guided isolation of the cytotoxic compounds. Large-scale fermentation of BRF082 in potato dextrose broth, followed by chromatographic purification of the bioactive fractions from the liquid medium, yielded gliotoxin (4) and its derivatives acetylgliotoxin G (3), bis(dethio)bis(methylsulfanyl)gliotoxin (1), acetylgliotoxin (5), 6-acetylbis(dethio)bis(methylsulfanyl)gliotoxin (2), besides the quinazolinone alkaloid fiscalin B. All isolated compounds were tested for their cytotoxicities against the tumor cell lines HCT-116, revealing 4 and 3 as the most cytotoxic ones (IC50 0.41 and 1.06 μg/ml, resp.).
A fungal strain of Aspergillus sp. (BRF 030) was isolated from the sediments collected in the northeast coast of Brazil, and the cytotoxic activity of its secondary metabolites was investigated against HCT-116 tumour cell line. The cytotoxicity-guided fractionation of the extracts from this fungus cultured in potato-dextrose-sea water for 14 days at room temperature yielded the hetero-spirocyclic γ-lactams pseurotin A (1), pseurotin D (2) and pseurotin FD-838 (7), the alkaloids fumitremorgin C (5), 12,13-dihydroxy fumitremorgin C (6), methylsulochrin (4) and bis(dethio)bis(methylthio)gliotoxin (3). Among them, fumitremorgin C (5) and 12,13-dihydroxy fumitremorgin C (6) were the most active. The cytotoxic activities of the extracts from Aspergillus sp. grown from 7 to 28 days were investigated, and they were associated with the kinetic production of the compounds. The most active extracts (14 and 21 days) were those with the highest relative concentrations of the compounds fumitremorgin C (5) and 12,13-dihydroxy fumitremorgin C (6).
O artigo discute a formação continuada para docentes da Educação Profissional e Tecnológica (EPT). Para tanto, inicialmente, apresenta um breve histórico sobre a formação de professores desta modalidade de ensino. Em seguida, mostra uma discussão sobre a formação continuada, a importância das experiências e dos saberes docentes. Em um terceiro momento, discorre sobre ideias, propostas de formação no contexto da EPT realizadas por autores, como Moura (2014), Machado (2008), Oliveira e Mororó (2018), Tardif (2014), que embasam o estudo. Por fim, conclui apresentando alguns desafios postos na atualidade para a formação docente na EPT e coloca como indispensável que a formação docente possa ter como base a realidade vivida pelos professores, além de ver como relevante os conhecimentos construídos na prática, ao longo da profissão, sem desconsiderar os saberes teóricos da educação. O trabalho é de cunho bibliográfico e documental.
The present article consists of a reflection on the teaching of history in the perspective of inclusion. In this sense, the main purpose of this work is to present a suggestion of a lesson plan based on the teaching of inclusive history, in which all, disabled or not, are contemplated. In order to achieve this goal, a bibliographic review was elaborated based on Circe Bittencourt, Paulo Santos, Maria Ciavatta and other authors. It was concluded that the History teacher, in order to base his teaching practice on an inclusive approach, should value the students' protagonism, address human diversity as a fundamental element of the historical process, seek interdisciplinary dialogue, seek basic knowledge about disabilities and other aspects.
Em busca de saudabilidade, o consumo de frutas é crescente, já que são ricas em compostos bioativos, tais como compostos fenólicos, flavonoides, carotenoides, vitaminas, etc (SHINWARI; RAO, 2018).Porém, é fato que a umidade desses alimentos é alta, no geral acima de 80%, sendo, então, altamente perecíveis.A industrialização de frutas e hortaliças é realizada em diversas etapas, dentre elas: colheita, transporte, seleção, lavagem, sanitização, processamento, embalagem e armazenamento. Durante o armazenamento e o transporte estimam-se perdas de 25% a 30% (OLIVEIRA, 2015), sendo pertinentes operações que reduzam esse valor.Reduzir as perdas durante a colheita e pós-colheita de frutas frescas, a partir do uso de partes sãs de frutas consideradas impróprias para comercialização, aumentar sua vida de prateleira e disponibilidade ao longo do ano é de interesse de quaisquer indústrias de alimentos. Muitas são as operações disponíveis para tanto, tais como produzir geleias.Na produção de geleias, o uso de calor, açúcar em altas concentrações e acidificação contribuem para aumentar a pressão osmótica do meio e reduzir sua atividade de água, resultando em ambiente inapropriado ao desenvolvimento de microorganismos (GAVA; SILVA; FRIAS, 2008), podendo, portanto, ser considerada uma técnica de conservação de frutas.Segundo a Resolução nº12 de 1978 do Ministério da Saúde, que dispõe sobre padrões de identidade e qualidade para alimentos: "Geleia de fruta é o produto obtido pela cocção, de frutas, inteiras ou em pedaços, polpa ou suco de frutas, com açúcar e água e concentrado até consistência gelatinosa" (BRASIL, 1978).A pectina é o polissacarídeo usualmente responsável pela formação de géis nas geleias, sendo usualmente adicionada durante sua cocção. O termo pectina diz respeito a uma família de compostos de ácido galacturônico ou galacturonoglicano, solúveis em água, que contém éster metílico em graus de neutralização variados, o qual forma géis (DAMODARAN; PARK;FENNEMA, 2010). Shinwari e Rao (2018) afirmam que, apesar de boa parte das frutas conter ácidos e pectina, esses componentes devem ser adicionados às geleias para atingir um valor mínimo de 1% de pectina e pH 3,0 para formar uma rede de gel consistente e característica. Oetterer, Regitano-D'Arce e Spoto (2006) apontam que nas geleias de frutas ocorre o equilíbrio entre o ácido, a pectina e o açúcar, originando um produto gelatinizado.
